Objective To explore the effectiveness of Silk Fibroin Hydrolytes/Traditional Chinese Medicine Complex on prevention of alcohol-induced liver injury in mice.Methods Establish acute alcoholism liver injury models using alcohol(50 %).Experimental mice were randomly divided into five groups:control group,model group,small dose level group,middle dose level group and high dose level group.Control group and model group were given distilled water BW/d 20ml/kg,and the three sample groups were given different level of Silk Fibroin Hydrolytes/Traditional Chinese Medicine Complex for 30 days continuously.Results Content of MDA,GSH and TG in mice livers have significant difference in the middle and high dose level groups compared with model group(P
2.A study on correlation between verbal memory and working memory in patients with alcohol dependence

Objective To explore the correlation between verbal memory and working memory in patients with alcohol dependence (AD).Methods A total of 40 patients with AD and 40 health controls were included in this study.N-back task was used to evaluate the verbal working memory.20 sets of phrases were used to evaluate long-term verbal memory.Results Correct number of verbal memory in AD group ((5.15±0.92)times) was lower than that of HC group ((7.35±3.07) times) after one-week baseline test,and the difference was statistically significant (t=4.344,P< 0.01).Compared with HC group ((1 436.37±192.50) ms,(1.08±0.89) times,(4.00±0.85) times,respectively),AD group((1 535.40± 192.50) ms,(1.90± 1.57)times,(2.60± 1.39)times) showed significantly impaired reply reaction time of 2-back,error number of 1-back and 2-back (t=-2.112,P=0.038;t=-2.899,P=0.005;t=-5.433,P<0.01).The error number of 2-back task in AD group was negative correlated with verbal memory after one-week baseline test (r=-0.427,P=0.006).Conclusion The verbal memory is impaired in AD patients.Long-term verbal memory damage have a certain correlation with working memory.
3.Transfection of human endostatin gene in umbilical cord CD34~+ hematopoietic stem cell

Objective:To transfer human endostatin gene into umbilical cord CD34 + hematopoietic stem cells and detect its expression and excretion. Methods: Human endostatin gene was transferred into human umbilical cord CD34 + hematopoietic stem cells by retroviral pLNCX to build endostatin-transferred cell line.RT-PCR and Western blot analysis were applied to examine the transfection and expression of endostatin gene. Results:RT-PCR proved that genome of endostatin-transferred CD34 + hematopoietic stem cells contained a 550 bp fragment of human endostatin .The expression and excretion of human endostatin from endstatin-transferred hematopoietic stem cells were confirmed by Western blot analysis. Conclusion:Human endostatin gene can be transferred into CD34 + hematopoietic stem cells.

Objective To determine the association of glutathione-stransferase M1(GSTM1), T1(GSTT1) genetic polymorphisms, smoking and alcohol consumption with the risk of gastric cancer. Methods 1:1 matched case-control study and molecular epidemiology technique were used to analysis the genetic polymorphisms of GSTM1, GSTT1 genotype among 60 cases of gastric cancer patients and sex-and age-matched controls, who were selected from Nanjing Zhongda Hospital and Jiangsu Province Cancer Hospital. Objective:To explore TCM syndrome variation regularity on acute stage of ischemia stroke.Methods:To collect clinical dynamic state information of ischemia stroke from 72h to 14days by using prospective clinical research method and investigate TCM syndrome variation regularity in acute stage of ischemia stroke.Results:Wind,fire and phlegm syndromes were the most frequent syndromes in acute stage of ischemia stroke.They appeared varied regularity.There were forty-four combination patterns about six basic syndromes.The wind syndrome and fire-phlegm syndrome were increasing in acute stage of ischemia stroke.The wind-phlegm syndrome was decreasing in the first three days,the wind-fire syndrome peaked on the third day,the phlegm was decreasing on the fifth day.The wind-fire-phlegm was the most frequent and undulate syndrome on the fifth day.The wind-phlegm-stagnation syndrome was increasing for the first three days.Conclusion:There were fastly morbility,changeable and complicated syndrome change in acute stage of ischemia stroke.It was very necessity and urgency for complementing diagnostic criteria of stroke differentiation.
6.Effect of halometasone in combination with scutellaria baicalensis georgi on the vitiligo mice

Aim To study the effect of halometasone in combination with scutellaria baicalensis georgi on the vitiligo mice induced by monobenzone. Methods 40% monobenzone cream was applied to induce vitiligo in C57BL/6 mice. Through the halometasone, halo-metasone and scutellaria baicalensis georgi combined with 40% monobenzone cream, the influence of halo-metasone and scutellaria baicalensis georgi on mice de-colorizing was studied. Hair decolorizing was observed with the naked eye, the skin decolorizing was observed by reflectance confocal microscopy ( RCM ) , and CD8 +T cell infiltration was tested with immunofluores-cence detection. The serum levels of interleukin-6(IL-6 ) and tumor necrosis factor-α( TNF-α) were deter-mined by enzyme linked immunosorbent assay ( ELISA) . Results Mice in model group showed de-pigmentation at both the monobenzone application part and non-application part. The halometasone group did not show significant therapeutic efficacy. In halometa-sone and scutellaria baicalensis georgi treatment group, there was less decolorization, the occurrence ratio, the scores of occurring time and size were lower compared with model group. There were fewer infiltrated lympho-cytes and CD8 +T cells. Halometasone and scutellaria baicalensis georgi group also showed that the serum levels of IL-6,TNF-α decreased. Conclusion Halo-metasone and scutellaria baicalensis georgi have thera-peutic effect on vitiligo mice induced by monobenzone.
7.The effect of methadone maintenance treatment on decision-making biases in male heroin dependent patients

Objective To investigate the characteristics of decision-making impairment in male patients with heroin dependence in methadone maintenance treatment(MMT). Methods A total of 45 male patients in MMT were compared with matched 32 health controls on the Iowa Gambling Task. Results Participants with anxiety or depression had to be excluded, MMT subjects were required to have negative urine toxicology screening tests.The MMT group selected more net disadvantageous cards (number of cards selected from the disadvantageous decks minus number of cards selected from advantageous decks) than health controls( (9.11 ± 19.60, -4.25 ±10.27 respectively), t=3. 884, P<0.01). A 2(group) ×5(block) ANOVA (analysis of variance) on the cards of net disadvantageous from the gambling task revealed a significant main effect of group( F= 12.414, P<0.01 ).The ANOVA also revealed a significant main effect of block( F=6.871, P<0.01 ). The study indicated the totalnumber of net disadvantageous cards for MMT subjects was negative correlation to the age of initial drug use ( r=- 0. 320, P < 0. 05 ). Meanwhile, methadone dose and the time of enrollment MMT did not reveal a significant main effect on decision-making biases. Conclusion The deficit of decision making for male heroin dependent patients might be attributed to the dysfunction of the ventromedial prefrontal cortex, and this effect does not resolve with abstinence, even very long term abstinence. Methadone may not get worse this impairment in male patients who have a history of heroin use during maintenance treatment intervention.
8.Clinical application of vaccination with four peptides-pulsed dendritic cells in postoperative patients with malignant melanoma

Objective To investigate the safety and clinical response of dendritic cells (DC)vaccine loaded with HLA-A2-restricted peptides MAGE-3、 Tyr、 MART-1 and GP-100 against malignant melanoma. Methods Twenty three HLA-A2 positive patients with malignant melanoma were enrolled.Peripheral blood mononuclear cells were separated into adherent cells for induction of DC loaded with four peptides. DC vaccine was administered subcutaneously once a week in inguinal region. The immunological responses and clinical responses were evaluated. Results Dendritic cell vaccination were well tolerated in all patients and there was no toxicity observed. Serum levels of IL-2, IL-12 and IFN-γincreased significantly after first vaccination and fourth vaccination. The positive rate of DTH test was 50% (5/10), and 2 fold increase of CD8+ IFN-γ+ cells were observed in 6 of 10 patients. Stable disease was observed in 11 of 23 patients, one patient had a complete metastasis regression, four patients had 50% regression of metastasis,four patients had a minor response, and disease progressed in three patients. Conclusion DC vaccines loaded with peptides MAGE-3、 Tyr、 MART-1 and GP-100 can elicit non-specific and specific immune responses, leading disease control. DC vaccine is considered one of safe and effective treatments for malignant melanoma.
9.A study of attention networks in the patients with obsessive-compulsive disorder and generalized anxiety disorder

Objective To investigate the attention network in patients with obsessive-compulsive disorder (OCD) and generalized anxiety disorder (GAD).Methods 31 GAD patients,31 OCD patients and 30 healthy controls (HC) were tested with attention network test.Results There were significant differences in the main effects with respect to the alerting network(15.87±5.24,26.77±4.33,34.87±3.47) (F=4.619,P=0.012)) and the executive network(114.84±9.64,122.45±5.57,96.57±5.45) (F=3.388,P=0.038) among the OCD,GAD and HC groups.The three groups did significantly differ in RT of the attention network (F=19.808,P=0.000).For the HC group,there was significant main effects with respect to cue conditions(F=29.699,P=0.000).There was a significant correlation between the executive network and Hamilton anxiety scale (r=0.351) and also a significantly negative correlation between the alerting network and Hamilton depression scale (r=-0.267).Conclusion There are attention network damages in patients with OCD and GAD,which may be involved in the impairment of emotion.
10.Investigation of attentional bias to word-related cues in male alcohol dependent patients

ObjectiveTo explore the attentional biases in male alcohol dependent (AD) patients and the correlations between the attentional bias and alcohol-related factors.MethodsA total of 30 recently detoxified male individuals with alcoholism were compared with 37 healthy controls ( HC ) on the Chinese Emotional Stroop Task using negative,neutral,and alcohol-related words.ResultsThe comparison between AD group( ( 1382.13 ±323.38) ms,( 1365.76 ±313.03)ms,( 1433.20 ±342.23) ms,respectively) and HC group( (797.27 ±216.97)ms,( 794.11 ± 209.41 ) ms,(799.40 ± 215.82 ) ms respectively) on the reaction time of neutral,negative and alcohol-related words were significant ( t =8.822,P < 0.001 ; t =8.922,P < 0.001 ; t =9.234,P < 0.001 ).The error number of of the neutral and negative- related words of the patients ( ( 3.70 ± 2.56) time,( 4.23 ± 2.53 ) time)was worse than that of HC group( ( 2.11 ± 1.87 ) time,( 1.92 ± 1.82 ) time) ( t =2.939,P =0.005 ; t =4.355,P <0.01 ).Error number of the alcohol- related words between two groups were not significant;Its alcohol-related words attentional bias negative correlation to the age of initial alcohol use(P< 0.05 ),and positive correlation to continue drinking and score of self-rating depression scale (P<0.05).Age of addiction and the score of self-rating anxiety scale enter the regression equation of alcohol-related words.ConclusionThese results demonstrate that alcoholics have attentional bias in alcohol-related words and reward-related brain regions may be associated with craving among male patients with attentional bias.
