AlM:To evaluate the effect of intralamellar cryolysis of cornea with amniotic membrane transplantation on painful bullous keratopathy ( PBK) .?METHODS: Randomly selected 156 cases ( 156 eyes ) with PBK who underwent surgery of intralamellar cryolysis of cornea with amniotic membrane transplantation. Followed up with 2 ~ 3mo, the symptoms of eye pain, corneal epithelial blisters, foreign body sensation and postoperative complications were observed.?RESULTS:Pain symptoms disappeared in all patients, and corneal epithelial blisters disappeared in 130 cases (83. 3%). All agonizing pain symptoms disappeared, but patients had occasional foreign - body sensation, occasional corneal epithelial blisters in 24 cases ( 15. 4%) . Two weeks after surgery, corneal stroma dissolved, 2 cases ( 1. 3%) of them were cured by conjunctival flap cover.?CONCLUSlON:The operation of intralamellar cryolysis of cornea with amniotic membrane transplantation can relieve the pain in 98. 7% of PBK patients and simple therapy for treating PBK. Hence, it's be worth to advocate for relieve the pain of patients.

Objective To explore the effects of extrinsic and intrinsic recognition loading on gait in patients with Parkinson disease. Methods Eight patients with Parkinson disease and six control subjects were instrutted to walk with extrinsic and intrinsic recognition loading respectively. The gait parameters in two conditions were measured with three-dimension motion analysis equipment.FAB scale and Stroop Test were used as the evaluation of executive function. Results The velocity(cm/s),the cadence(step/min),single support and swing phase(%)in the audition stimulates condition in the patients group((113.4±14.32)step/min,(78.90±16.35)cm/s,(40.50±2.58)%,(40.50±2.58)%)were significantly better than those in the calculation condition(respectively(91.27±15.54)step/min,(63.79±21.49)cm/s,(37.95±2.61)%,(37.95±2.61)%).In the calculation condition, the coefficients of variability in stride length, swing and sinfle support phase were significantly higher in the patient group(respectively(6.69±3.99),(8.56±5.69),(8.56±5.69))than in thecontrol group(respectively(3.23±1.34),(5.02±2.54),(5.02±2.54));in the audition condition, except that, the coefficients of variability in step length and velocity also were significantly higher in the patient group(respeetively(11.92±5.86),(6.89±4.98))than the control group(respectively(7.35±3.32),(2.5±1.53)).In the patients group, the score of FAB(15.63±1.51)was lower and error rates of Stroop test(0.087±0.056)was higher than those in the control group(respectively(17.67±0.52),(0.027±0.03))significantly.The error rate of stroop-3 was significantly negative correlated with the gait variables of patient group in the calculation condition. Conclusion The extrinsic audition stimulates has lower effect on the gait of patients of Parkinson's disease than the intrinsic recognition loading.

It is necessary to provide a SPARQL query tool for biomedical researchers due to the massive data of bi-omedical ontology. However, manual input of SPARQL query statements is quite complicated and easy to produce errors. A Cytospace-based parameterized SPARQL query system was thus developed for biomedical ontology, which can generate a module with SPARQL query conditions used as parameters. Biomedical researchers can thus rapidly, easily and directly retrieve the data in several biomedical ontology knowledge libraries instead of manual input of SPARQL query statements.

Objective To construct the recombinant plasmid that highly expressed human subcellular PTEN,in order to provide a basis for further study on its anti-tumor effect. Methods PTEN cDNA was amplified by RT-PCR based on mRNA of placenta. The PCR product was ligated into T-vector,and transfected into E.coli;the obtained T-PTEN plasmid was identified with restrictive digestion and sequencing.PCR was used to incorporte nuclear signal of localization(NSL) into PTEN when T-PTEN was used as template. Then the PCR product was ligated into T-vector,and transfected into E.coli, and T-NSL-PTEN plasmid was obtained.pcDNA3.1 and T-NSL-PTEN were ligated after digested with EcoRⅠand BamHⅠ,and transfected into E.coli, the recombinant vector pcDNA3.1-NSL-PTEN was obtained, and identified with digestion and sequcncing. Results The recombinant expression vector DUM-PTEN and PUM-NSL PTEN were identified by restrictive digestion and DNA sequencing. As expected,by EcoRⅠ and BamHⅠ digestion,it showed the band of 1 200 bp.The sequencing result showed the NSL was incorporated successfully. The recombinant pcDNA3.1-PTEN was obtained with 1 200 bp,the sequencing result showed that its sequence was same as target gene;the recombinant pcDNA3.1-NSL-PTEN was comfirmed by restrictive digestion and sequencing, and the NSL was incorporated successfully. Conclusion The recombinant expression plasmid pcDNA3.1-NSL-PTEN is constructed successfully which can highly express human subcellular PTEN.

flow cytometry.A strong linear relationship was observed in the standard curve of real-time PCR of each bacteria. Conclusion These three non-culture methods can be used in the quantitative analysis of oral microorganisms.Real-time PCR and laser scanning confocal microscopy are better than the traditional culture-based CFU count, and real-time PCR is the most sensitive method.

Objective To detect the differential gene expression between Streptococcus sobrinus(S.sobrinus) 6715 and its fluoride-resistant strains. Methods The fluoride-resistant strains of S.sobrinus 6715 was induced by increasing the concentration of fluoride step by step.Total RNA of both S.sobrinus 6715 and its fluoride-resistant strains was extracted,mRNA was separated and purificated, and then cDNA was obtained by reversed transcription.Suppression subtractive hybridization(SSH) technology was used to detect the differential gene expression between them. The differential gene expression fragments were cloned and compared with the GenBank by BLAST.Results After comparing with the GenBank by BLAST,it was identified that there were two differential gene expression fragments, fruA and SMU.438c. Conclusion The cDNA subtractive lib of differential gene expression between S.sobrinus 6715 and its fluoride-resistant strains was successfully constructed through SSH,which paves a way for the further study of fluoride-resistant mechanism.
