1.Effect of disodium quercetin-7,4′-disulfate on enzyme activity of recombinant human phosphoinositide 3-kinase p110 catalytic subunit

Objective To study the effect of disodium quercetin-7,4′-disulfate (SQDS) on activity ofrecombinant human phosphoinositide 3-kinase (PI3-K) p110?catalytic subunit. Methods Recombinanthuman PI3-K p110?catalytic subunit was expressed by gene engineering. PI3-K activity was assayed byincubating recombinant PI3-K p110?with phosphatidylinositol-4,5-bisphosphate and [?-32P] ATP;the32P-radiolabeled lipids were extracted with chloroform and methanol,then assessed by scintillation counter.Results SQDS showed inhibition on the recombinant p110?catalytic subunit with IC5014.88?mol/L.Conclusion SQDS is an inhibitor of PI3-K. The recombinant PI3-K p110?catalytic subunit might be usedas a molecular target for simpler screening and development of more effective inhibitors of PI3-K.

3.Effects of sodium quercetin-7-sulfate on recombinant human protein kinase CK2 holoenzyme

AIM To study the effects and kinetics of sodium quercetin 7 sulphate (SQMS) on recombinant human protein kinase CK2 holoenzyme. METHODS The recombinant human CK2 holoenzyme activity was assayed by detecting incorporation of 32 P of [? 32 P]ATP into the substrate at various conditions. RESULTS SQMS was shown to strongly inhibit the holoenzyme activity of recombinant human protein kinase CK2 with an IC 50 of 1 37 ?mol?L -1 , Kinetic studies of SQMS on recombinant human CK2 showed: the inhibition was competitive with ATP and noncompetitive with casein. CONCLUSIONSQMS is an inhibitor of protein kinase CK2, and the inhibitory action of SQMS on CK2 is competitive with ATP and noncompetitive with casein.

5.A comparative analysis on the effect of laparoscopic repair of gastric perforation with open surgery on pulmonary function

Objective To compare the effect of traditional open surgery and laparoscopic repair of gastric perforation on postoperative pulmonary function.Methods Fifty patients were divided into two groups according to therapeutic method.Group A underwent traditional open gastric perforation repair(n=25).Group B underwent laparoscopic repair of gastric perforation(n=25).The pulmonary functions were examined at 1 day,3 days and 7 days postoperatively.Results All patients had different degrees of postoperative pulmonary function decline.After operation,the forced vital capacity(FVC)and forced expiratory volume in 1 second(FEV1)and vital capacity(VC)in group A were significantly lower than those in preoperation(P<0.05).The patients in laparoscopic operation group decreased significantly only on the first day postoperative.The postoperative pulmonary function of patients undergoing open surgery was significantly lower than that of patients undergoing laparoscopic surgery(P<0.05).Conclusion Laparoscopic repair of gastric perforation has less damage to the lung function than traditional open gastric perforation repair.Laparoscopic surgery plays an important role in the recovery of postoperative pulmonary function.

Objective To observe the efficacy of percutaneous vertebroplasty (PVP) combined with zoledronic acid in treatment of spinal metastatic carcinoma.Methods Fifty cases of spinal metastatic carcinoma were divided into control group and treatment group by random digits table method with 25 cases each.The control group was given zoledronic acid 4 mg,intervals of 28 d intravenous drip 1 time.The treatment group was given PVP besides zoledronic acid.The visual analog scale score,activity ability score and analgesic usage score after treatment for 3 months were observed and compared.The quality of life was evaluated and the occurrence of relapse of fracture was also recorded.Results After treatment for 3 months,the visual analog scale score,activity ability score and analgesic usage score were (2.00 ± 0.17),(2.12 ± 1.38),(1.47 ± 0.10) scores in treatment group,and (7.84 ± 0.18),(3.21 ± 0.13),(3.30 ± 0.09) scores in control group,and there were significant differences between two groups (P < 0.01 or < 0.05).The occurrence of relapse of fracture was 2 cases (8%,2/25) in treatment group and 9 cases (36%,9/25) in control group,and there was significant difference between two groups (P < 0.05).Conclusions PVP combined with zoledronic acid in treatment of spinal metastatic carcinoma has many advantages such as small incision,short surgery time and rapid relief from pain.It can obviously improve the quality of life,and it is worthy of spreading in the clinic.
10.Dorsal foot flap transplantation for skin and soft tissue defects of the hand:survival rate and success analysis

BACKGROUND:Skin and soft tissue defects of hands are easy to appear due to different factors, such as trauma. Flap transplantation is a clinical treatment for soft tissue defects of hands. Dorsal foot flap, which is thicker with constant vessel traveling, is very suitable for repair of hand soft tissue defects. OBJECTIVE: To explore the success rate and survival rate of dorsal foot flap transplantation in the repair of hand skin and soft tissue defects. METHODS: A retrospective analysis was performed on the clinical data of 86 patients with skin and soft tissue defect of hands. These patients were respectively given conventional skin wound skin grafting (control group) and dorsal foot flap transplantation (observational group). Hand appearance, infection rate, perception recovery and complications were observed during the postoperative folow-up. RESULTS AND CONCLUSION:Skin flaps al survived after grafting, and the texture of flaps was good. Patients were satisfied with hand shape and the recovery of hand function. After flap transplantation, one patient had local skin necrosis at the donor region, and the scar healed after dressing; another patient had venous crisis, and was given vascular re-anastomosis. In the observation group, the postoperative appearance was better than that of the control group, the infection rate was lower than that of the control group, the perception recovery was better, and there were fewer complications (P < 0.05). These findings suggest that the dorsal foot flap transplantation is better to repair skin and soft tissue defects in the hand.
