1.Construction,expression and identification of yeast two-hybrid bait vector pGBKT7-PML-V

Objective:To construct the bait expression vector pGBKT7-PML-V of retinoic acid receptor variant protein,for screening the target proteins interacting with the bait protein through the yeast two-hybrid system.Methods:The fragments of PML-V binding domain was amplified by PCR,and then was cloned into the bait expression vector pGBKT7.After being verified by sequencing,the bait vector pGBKT7-PML-V was transformed into AH109 yeast cells.Then the expression of the bait protein was analyzed by Western Blotting.Toxicity,leakage and serf-activation of the bait protein were detected.Results: PML-V was amplified and cloned into pGBKT7 successfully.The bait vector was transformed into AH109 as well and no toxicity and self-activation were found.The expression of the bait protein was confirmed by Western Blotting.Conclusion:The bait expression vector of PML-V was constructed successfully,which layed the foundations for screening target proteins interacting with the bait protein using the yeast two-hybrid technique.
2.Construction and identification of the bait expression vector of GR?-LBD of human HL-60 cell

Objective:To construct the bait expression plasmid pGBKT7-GR?-LBD of glucocorticoid receptor 1igand binding domain in order to lay the foundation for constructing small molecule ligand yeast three-hybrid system.Methods:A fragment of GR?-LBD of human HL-60 cell was amplified by RT-PCR,and then was subcloned into the bait expression vector pGBKT7.After verified by sequencing,the bait vector pGBKT7-GR?-LBD was transfected into AH109 yeast cells.And the expression of the bait protein was analyzed by Western-blotting.Toxicity and self-activation of the bait protein were detected simultaneously.Results:GR?-LBD was amplified and cloned into pGBKT7 successfully.The bait vector was transformed into AH109 yeast cells successfully,and no toxicity and self-activation were found.The expression of the bait protein was confirmed by western blot.Conclusion:The bait expression vector pGBKT7-GR?-LBD was constructed successfully,which lays the foundation for constructing small molecule ligand yeast three-hybrid system.
3.Effect of losartan on expression of PTHrP and PTH1R induced by high glucose in NRK-52E cells

To study the effect of losartan on high glucose up-regulated expression of parathyroid hormone-related peptide (PTHrP) and its type 1 receptor (PTH1R) in NRK-52E cells. The expression of PTHrP and PTH1R were detected by RT-PCR and Western blot in control group (0 mmol/L glucose) ,normal glucose group (5 mmol/ L glucose) ,moderately high glucose group (16.7 mmol/L glucose) ,high glucose group (25 mmol/L glucose) ,and after intervention by 10 μmol/L losartan for 72 h (only Western blot). The expression of PTHrP and PTH1R were up-regulated by high glucose (PTHrP mRNA : 0. 66 ± 0.08, 0. 84 ± 0. 13,1.57 ± 0. 15, and 1.73 ± 0.21 ; PTHrP protein :0.63±0.12,0.68±0.06,1.02±0. 11, and 1.04±0.08 ; PTH1R mRNA :0.26±0. 08,0.28±0.07,2.35± 0. 10,and 2.47±0. 05 ; PTH1R protein:0. 88±0. 05,0. 87±0. 10, 1.05±0. 11, and 1.12±0. 09) ,and losartan inhibited the effects of high glucose (PTHrP 0.74±0. 15, PTH1R 0.98±0.06, both P<0.01). The results suggest that losartan could inhibit the expression of FTHrP and PTH1R induced by high glucose in NRK-52E.

5.The application of sacral block anesthesia in pediatric interventional therapy

Objective To discuss the management and feasibility of sacral block anesthesia in pediatric interventional therapy. Methods A total of 80 pediatric patients were randomly and equally divided into two groups. Patients in group A received sacral block anesthesia together with basic anesthesia with propofol, while patients in group B received intravenous anesthesia with propofol. Small amount of ketamine as maintaining dose was used in both groups when needed. Results The interventional management was successfully completed in all patients. A marked decrease in blood pressure occurred in three patients of group A receiving sacral block anesthesia. In group B receiving intravenous anesthesia, a decrease, of SpO_2 to below 90 percent was seen in 8 cases, and obvious bradycardia developed in 12 cases. All these patients were treated with intravenous medication or by reducing the dose of propnfol. Additional small dose of ketamine was needed in 4 patients during the procedure. Conclusion Sacral block anesthesia combined with intravenous anesthesia is one of the effective anesthesia management schemes for pediatric interventional therapy.
6.Discussion on "removing stasis and regulating qi and blood" in the scraping therapy.

Stasis is the critical causative factor and pathological production considered in clinic treatment. Promoting the circulation in blood vessel and removing stasis are equally important in treatment. Stasis is interactive with qi and blood. Removing stasis aims to regulating the circulation of qi and blood. Scraping therapy is one of the major approaches to stasis removal in TCM. In clinical practice, the stasis can be removed with scraping, bleeding and cupping, etc. Additionally, removing stasis with scraping should be combined with regulating qi and blood to enhance the therapeutic effect.

Objective To study the role of parathyroid hormone-related peptide ( PTHrP) in transdif-ferentiation of NRK-52E cells induced by high glucose. 9.Value of serum intestinal fatty acid binding protein and calcium levels for evaluating the effect of emergency surgical on the treatment of severe acute pancreatitis

Objective To study the value of serum intestinal fatty acid binding protein(I-FABP) and calcium levels for evaluating the effect of emergency surgical on the treatment of severe acute pancreatitis(SAP). Methods A total of 68 firstly diagnosed as SAP from June 2014 to October 2016 were collected into this study. The arrest time was less than 72 h and they were underwent emergency endoscopic or laparoscopic surgery. The serum I-FABP and calcium levels were detected 12 hours after surgery and analyze the values of serum I-FABP and calcium levels in evaluating the clinical effects by receiver operating characteristic curve(ROC). Results There were 33 cases with excellence, 21 cases with availability and other 14 cases with no-effect, the total efficacy rate was 79.4%. The gender, age, arrest time, APACHE Ⅱ score, derum amylase, blood glucose and total cholesterol levels were no statistical differences in effective group and no-effect group.The serum I-FABP levels 12 hours after surgery in effective group were lower and calcium levels were higher than no-effect group(P<0.05).The evaluating accuracy was 0.856, sensibility was 86.5%,specificity was 92.2%,cut-off value was 368 ng/mL of serum I-FABP by ROC analysis. And they were 0.844,83.5%,86.7% and 1.96 mmol/L of serum calcium level by ROC analysis. Conclusion Serum I-FABP combined with calcium can be used for evaluating the effects of emergency surgical on the treatment of the patients with SAP.
10.Sites of action of propofol and isoflurane on somatosensory pathway

Objective To determine the sites of action of propofol and isoflurane on somatosensory pathway using median nerve somatosensory evoked potential(MnSSEP) .Methods Twenty-six ASA I - II patients aged 20-50 yrs were randomly divided into two groups: I propofol group( n = 13) and H isoflurane group( n = 13) .In propofol group patients received propofol infusion at a rate of 10 mg?kg-1?h-1 .Oxygen was administered via mask and respiration was assisted or controlled to maintain SpO2 at 96%-100% and PETCO2 at 35-45 mm Hg. The propofol infusion was continued until the patient failed to respond to verbal command. Six minutes later if the patient was still breathing spontaneously, the rate of propofol infusion was increased to 20-40 mg?kg-1?h-1 . In isoflurane group anesthesia was induced with propofol 2 mg ? kg-1 and intubation was facilitated by succinylcholine and anesthesia was maintained with isoflurane inhalation and intermittent iv boluses of vecuronium. The end-tidal isoflurane concentration was maintained at 0.5,1.0 and 1.5 MAC. Each concentration was maintained for 15 minutes. MAP, HR, SpO2,PCTCO2, T0(naso-pharyngeal) and BIS were continuously monitored. MnSSEP( Viking IV D type) was measured and recorded before induction of anesthesia(baseline) and in propofol group when patients became unconscious and apneic; in isoflurane group when end-tidal isoflurane reached 0.5,1.0 and 1.5 MAC.Results In group I there was no change in both latencies and amplitudes of N9 and N13'.With increasing infusion rate, propofol gradually prolonged the latencies and decreased the amplitudes of N60, P45, N35, N20 and P25 waves. In isoflurane group there was no change in the latencies and amplitudes of N9. There was no change in the latencies of N13', but the amplitudes decreased at 1.0 and 1.5 MAC. With increasing concentration, isoflurane gradually prolonged the latencies and decreased the amplitudes of P45, N60, N35, N20 and P25 waves. At 1.5 MAC the inter-peak latencies between N13'-N20 and N13'-P25 were prolonged. Conclusions The sites of action of different infusion rates of propofol and different concentrations of isoflurane are different on somatosensory pathway. The higher the doses, more widespread are the sites of action.
