1.Clinical pathological analysis of singularity uterine leiomyoma

Objective To explore the clinical and pathological characteristics of singularity uterine leiomyoma.Methods The cause of disease and the clinical features in 15 patients with singularity uterine leiomyoma were analyzed.The patients were examined by pathology and immunohistochemistry.Results The main clinical manifestations were irregular vaginal bleeding,pelvic tumor and anemia.3 patients combined with pregnancy,4 patients had the treatment history of exogenous progesterone.The tumor appeared grayish red,and with complete envelope by macroscopic examination;The multi-and mono-nucleated giant cells were seen under surgical microscopy,the mitosis count was 0~3/10HPF.The tumors showed weak positive staining for Ki-67 and p53,negative staining for ER in 8 cases and positive staining for PR in all cases.Conclusions Singularity uterine leiomyoma belongs to benign tumor,its clinical features and treatments are similar to common leiomyoma;It has certain relation with exogenous and endogenous progesterone.The differentiation of leiomyoma with leiomyosarcoma is basically depended on HE staining observation;The immunohistochemistry of leiomyoma has certain feature.
2.The effect of danshensu (DS-182) of the injury of oxygen free radicals in myocardial mitochondria from rat myocardium

The role of DS-182 in protection of cellular damages induced by oxygen free radicals was studied in rat cardiac mitochondria. The changes of cardiac mitochondrial enzyme (cytochrome oxidase) and P/O, RCR were used as indices of mitohondrial injury. The experimental animals were divided into three groups: control group, injury group (Fe~(2+)+ascorbic acid), and protection group (DS-182+Fe~(2+)+ascobic acid). Results obtained showed that incubation of mitochondrial suspension with ascorbic acid and Fe~(2+) resulted in decrease of activity of cytochrome oxidase, P/O (with ?-ketoglutarate andsuccinate as substrate respectively) and RCR, which indicated that the structure and function of mitochondria were damaged. Danshensu (DS-182) can inhibit the decrease of cytochrome oxidase activity, P/O and RCR. Thus, DS-182 with acts as an effective scavenger for oxygen free radicals prevented the mitochondrial injury.

4.Risk Analysis on the Collinear Production of Solid Preparations Based on Failure Mode and Effects Analysis as the Quality Risk Management Tools

Objective: To analyze and estimate the risks of failure modes in a pharmaceutical Co.Ltd.in Nanjing producing solid preparations by collinear production, and come up with measurements to improve the quality level accordingly.Methods: The quality risk level evaluation table for the failure modes in the various steps of collinear production was obtained through the failure mode and effects analysis (FMEA).The risk control measurements were put forward in order to control and improve different failure modes with various unacceptable risk levels.Results: The risks of solid preparations by collinear production were greatly reduced through FMEA.Conclusion: The FMEA method is an effective way to improve the quality level of collinear products.Meanwhile, there are limitations of FMEA, and other risk management methods should be combined to control the overall risk of drug production.Moreover, the failure modes at the same risk level with different properties should be analyzed and controlled accordingly.The above methods can improve the risk management level of a company and reduce the risks of cross-contamination, mistakes and air-transferring in order to enhance the efficiency of quality management system and produce safe and effective drugs.
5.Diagnostic Value of Multichannel Intraluminal Impedance in Gastroesophageal Reflux Disease

Multichannel intraluminal impedance( MII)is a new technique that monitors comprehensively the function of esophagus and the nature and composition of gastroesophageal reflux, and can be applied for the diagnosis of gastroesophageal reflux disease( GERD),especially suitable for the diagnosis of weakly acidic reflux,non-acid reflux, refractory GERD,and GERD with atypical symptoms. This article reviewed the study on MII and its clinical application in diagnosis of GERD.
6.Influence on Proliferation and Invasion of Human Breast Carcinoma Cell Line MDA-MB-231 by Using MicroRNA Interference Technique to Suppress MTDH Expression

Objective To investigate the effects of RNA interference technique silencing MTDH on proliferation,metastasis and invasion of human breast cancer cell MDA-MB-231.Methods The MTDH miRNA was transfected into Human breast cancer cell MDA-MB-231 by means of lipofectamine 2000.The silencing efficiency of MTDH was examined by RT-PCR and western blot.The influence of MDA-MB-231 cell proliferation,metastasis and invasion ability from inhibition MTDH gene was conducted by using MTT assay,scratch experiments,and Transwell assay.Results MTDH miRNA effectively inhibited MTDH mRNA and protein levels.The best inhibition rate were 79.41% and 80.56% (P < 0.05).With miRNA interferring cells,the ability of cells proliferation was decreased (P < 0.05),and cells invasive and metastasis were depressed (P < 0.05).Conclusion MTDH expression is obviously suppressed after transfected by MTDH miRNA in MDA-MB-231 cells.The inhibition of MTDH expression from microRNA leads to significant suppression of proliferation,invasion and metastasis of breast cancer cell MDA-MB-231.

AIM: To evaluate the effects of radiotherapy combined IL 2 in the treatment of the transferred remnant of nasopharyngeal carcinoma in jugular lymph. 8.Clinical evaluation of early impairment of renal function in elderly patients with chronic heart failure by detecting serum cystatin C

Objective To investigate the clinical value of serum cystatin C in the early renal function impairment in elderly patients wtih chronic congestive heart failure (CHF). Methods Serum cystafin Cwas measured in 60 cases with chronic CHF (CHF group), 40 cases with cardiovascular diseases and compensated heart function (compensated heart function group), and other 20 elderly normal volunteers(controlgroup). The correlation between cystafin C, B-type natriuretic peptide (BNP) and LVEF was evaluated. Results The levels of eystatin C in CHF group [(1.86±0.37)mg/L]was higher than that in compensated heart function group [(1.32±0.56) mg/L]and control group [(1.05±0.43) mg/L], P < 0.01. Cystatin C was positively correlated with BNP (r = 0.528,P< 0.01)and negatively correlated with LVEF (r=-0.463, P<0.01). In CHF group, the level of cystatin C and the incidence rate of renal failure was higher in the patients with BNP≥500 ng/L than those with BNP < 500 ng/L. Conclusions Cystatin C is a marker dearly impairment of renal function in elderly patients with chronic CHF. The more severe the heart failure, the more obvious the glomorular filtration function impairment shows.

10.Quantitative study on loss of dopaminergic neurons in SNpc of C57/BL6 mice induced by MPTP

Objective To observe the distribution of dopaminergic neurons in SNpc and to establish standard curve in normal mice so as to measure the changes of dopaminergic neurons in number in SNpc of the mice toxicated with MPTP. Methods Ten male C57BL6 mice aged 8-12 weeks, weight 20-22 g, were randomized to receive 20 mg/kg MPTP or physical saline every 3 h for 4 times, then killed 7 d later. The mouse mesocerebrum was taken out and fixed, frozen, sectioned. All sections containing SNpc were observed under the guide of mouse brain atlas. Every other sections were chosen to stain tyrosine hydroxylase (TH) to show dopaminergic neurons immunohistochemically. The TH positive cells in SNpc were counted in each section and the standard curve was established. Results The standard curve of SNpc compact part position and TH positive cells was established. By comparing the standard curves for the MPTP intoxicated mice and the saline mice, TH positive cells in SNpc from MPTP toxicated mice decreased significantly, which confirmed the validity and feasibility of the standard curve. Conclusion The establishment of standard curve greatly facilitates the comparison of specimen from different groups and makes the assessment of dopaminergic neuron loss more accurate and efficient. The standard curve can serve as an excellent reference curve for the assessment of dopaminergic neurons in SNpc in normal C57BL6 mice.
