2.Analysis of toxicity of traditional Chinese herbal medicine and its connotation.

Based on traditional Chinese medicine theory and clinical experience, traditional Chinese herbal drug toxicity has its own special connotation. From the perspective of history and logic, the different comprehension of toxicity between Western medicine and traditional Chinese medicine was discussed after retracing the meaning of "drug toxicity" in traditional Chinese medicine. The authors suggest that it's not feasible to study the Chinese medicine coping mechanically and applying indiscriminately the concept and the research idea about modern drug toxicity since there is different understanding of "drug toxicity" between traditional Chinese medicine and Western medicine. Many control elements are involved in the use of traditional Chinese herbal drugs, and Chinese drug components and actions are complex as compared with Western drugs. More and more drugs with toxicity will be found due to the relativity of drug toxicity. Currently, the study of Chinese drug toxicity should pay more attention to the relation between the toxicity and Chinese drug nature, compatibility and the corresponding disease or syndrome pattern after making definition of Chinese drug toxicity and its connotation.
3.Analysis of Essential Oil from Fructus Ponciri Trifoliatae Immaturi by GC-MS

Objective To establish a GC-MS method for the analysis of the chemical components of essential oil from Fructus Ponciri Trifoliatae Immaturi.Methods The essential oil was extracted by steam distilyation ,then separated by capillary gas chromatography. The components from essential oil were identified by comparing with related standard chromatogram and their amount were determined by normalization method.Results Twenty-one components have been identified from Folium Fructus Ponciri Trifoliatae Immaturi and their amount accounted 99.04 %of total essential oil. The main components are limonene (55.68 %),?-myrcene(21.96 %),?-Pinene(5.17 %),cis-caryophyllene(4.61 %)and ?-Ocimene (1.71 %).Conclusion This method is reliable and effective and can be applied to analyze the essential oil components extracted from Fructus Ponciri Trifoliatae Immaturi.

5.Clinical pathological analysis of singularity uterine leiomyoma

Objective To explore the clinical and pathological characteristics of singularity uterine leiomyoma.Methods The cause of disease and the clinical features in 15 patients with singularity uterine leiomyoma were analyzed.The patients were examined by pathology and immunohistochemistry.Results The main clinical manifestations were irregular vaginal bleeding,pelvic tumor and anemia.3 patients combined with pregnancy,4 patients had the treatment history of exogenous progesterone.The tumor appeared grayish red,and with complete envelope by macroscopic examination;The multi-and mono-nucleated giant cells were seen under surgical microscopy,the mitosis count was 0~3/10HPF.The tumors showed weak positive staining for Ki-67 and p53,negative staining for ER in 8 cases and positive staining for PR in all cases.Conclusions Singularity uterine leiomyoma belongs to benign tumor,its clinical features and treatments are similar to common leiomyoma;It has certain relation with exogenous and endogenous progesterone.The differentiation of leiomyoma with leiomyosarcoma is basically depended on HE staining observation;The immunohistochemistry of leiomyoma has certain feature.
6. Effects of resistin gene on TNF-α-induced expression of ICAM-1 in vascular endothelial cells

Objective: To study the effect of resistin gene on TNF-α-induced expression of ICAM-1 in ECV304 cells. Methods: Resistin gene was cloned into vector pEGFP-C1 to construct pEG/ Resi plasmid; the latter was then used to transfect ECV304 cells via liposome. TNF-α(20 μg/L) was added to the system 6 h after transfection. RT-PCR was used to detect the expression of resistin and ICAM-1 mRNA in the cells 18, 30, 42 and 54 h after transfection; meanwhile, the resistin protein levels were determined by Western blotting analysis. ICAM-1 protein levels in the supernatants of ECV304 cells were determined by ELISA. Results: Restriction enzyme digestion and sequencing demonstrated that the recombinant plasmid was successfully constructed and was expressed in ECV304. ICAM-1 levels in resistin-transfected group were consistently higher than that in the non-transfected group in the presence of TNF-α. ICAM-1 mRNA and protein expression in the transfected-group was significantly higher than that in the non-transfected group during the peak periods (30 and 42 h) of resistin expression (P<0.05); ICAM-1 mRNA and protein expression increased with the increase of resistin gene expression. Conclusion: Resistin gene can enhance TNF-α-induced expression of ICAM-1 in ECV304 cells.
7.Establishing of the HSV-2 infected cell culture system

Objective To establish a HSV-2 infected cell culture system,so as to confirm the suppressing effect of medicine on HSV-2 including effective concentration and safety.Methods A monkey kidney cell line,Vero ,and a human tracheal epithelial cell line,Hep-2 were used in HSV infected cell culture system.HSV-2 was propagated in Vero cells and Hep-2 cells was served as targets of HSV-2 infection.Then cytopathic effect(CPE) was performed.Results HSV-2 was propagated abundance in Vero cells after4 days:The CPE occurred in Hep-2 cells after 12 hours infected by HSV-2.Conclusion The HSV-2 infected cell culture system we established was stable for utilization.As well as to confirm the suppressing effect of medicine on HSV-2.
8.Retrobulbar injection with triamcinolone acetonide for macular edema caused by retinal vein occlusion

Objective To evaluate the therapeutic efficacy of retrobulbar injection with triamcinolone acetonide (TA) for macular edema caused by retinal vein occlusion (RVO). Design Retrospective case series. Participants 21 eyes of 21 patients with macular edema caused by RVO. Methods Retrobulbar injection with TA 40 mg was performed on 21 eyes. The visual acuity, intraocular pres-sure, fundus fluorescein angiography (FFA) and macular central thickness under the optical coherence tomography (OCT) were recorded and analyzed before and after treatment. The follow-up duration was 6-11 months. Main Outcome Measures The visual acuity and the macular central thickness of OCT. Result Visual acuity was improved at the end of follow-up in 16 eyes (76.2%) and unchanged in 5 eyes (23.8%). Macular central thickness was (287.55±121.70) μm at the end of follow-up and (617.23±185.58) μm before treatment. The difference was statistically significant(P=0.000). After treatment, the comparison of visual acuity and macular central thickness be-tween CRVO patients and BRVO patients, as well as between ischemic RVO patients and non-ischemic RVO patients had no significant difference (P>0.05). In patients of course less than 6 months, the visual prognosis after treatment is better than that of before treatment (P=0.011, 0.01), while in patients of course more than 6 months, no difference of vision could be detected before and after treatment (P= 0.583). The result of FFA showed decrease of fluorescein leakage. During follow-up, intraocular pressure of all patients was in the nor-real range. Recurrence of macular edema was found in 8 eyes 2 to 6 months after treatment. Conclusions Retrobulbar injection with TA is a safe, effective, and easy method to treat macular edema caused by RVO. Now the treatment of lymphoma mainly uses combined chemotherapy and radiotherapy.Es- pecially hemopoietic stem cell transplantation is effective in the treatment to the patients with refractory and later stage.During the recent years the domestic and foreign scholars have obtained the encouraging results with arsenic trioxide to treat lymphoma.This article has reviewed the experimental study and the clinical re- search progress about arsenic trioxide in application to lymphoma cells.
10. Effects of ovariectomy on autophagy and senile plaques in the brain of adult APP/PS1 double transgenic mice

Objective To investigate the effect of ovarian estrogen on autophagy and senile plaques (SP) in the brain of adult APP/PS1 double transgenic mice, and inquire into the mechanism of Alzheimer disease (AD) neuropathogenesis due to estrogen deficiency. Methods APP/PS1 AD mice (3-month old) were divided into ovariectomy group (OVX-AD) and sham group (sham-AD). Two months after operation, transmission electron microscopy (TEM) was employed to observe the morphological structures of the brains of AD model mice, and immunohistochemical staining was performed to examine the changes in SP and autophagy-related proteins LC3 and Beclin-1 in the brain of AD model mice. Results Immunohistochemical staining showed that the number of SP in the brain of OVX-AD mice incrcased significantly compared with that in sham-AD mice, and distribution of SP was observed more extensively, including the cortex and hippocampus. The autophagy related proteins LC3 and Beclin-1 positive neurons were decreased greatly in the OVX-AD mice brain compared with that of sham-AD mice. TEM revealed a larger number of swollen and dark neurons in hippocampus of OVX-AD mice, while more autophagosome was observed around the neuronal processes in OVX-AD mice brain compared with sham-AD mice. Conclusion Estrogen deficiency may lead to delayed maturation of autophagy, decrease the activity and weaken the function of autophagy, which in turn may result in aggravation of pathological features of AD.
