1.The role of O-GlcNAc modification in glutamine-induced myocardial heat shock protein 70 expression in a rat model of endotoxic shock

Objective To investigate the role of O-linked N-acetylglutamine (O-GlcNAc) modification in glutamine-induced myocardial heat shock protein (HSP70) expression in a rat model of endotoxic shock. Methods Thirty-two 8 weeks old male SD rats weighing 250-300 g were randomly divided into 4 groups (n = 8 each):group Ⅰ control (group C); group Ⅱ LPS; group Ⅲ G+ LPS and group glutamine + LPS (group A + G + LPS). Endotoxic shock was induced by iv administration of LPS 10 mg/kg in group Ⅱ , Ⅲ and Ⅳ.Glutamine 0.75 g/kg was administered iv at 1 h before iv LPS in group Ⅲ and Ⅳ . Alloxan (an inhibitor ofO-linked N-acetylglucosamine transferase) 50 mg/kg was administered iv together with glutamine at 1 h before LPS in group Ⅳ. The animals were sacrificed at 6 h after iv LPS administration. Their hearts were removed for determination of the expression of O-GlcNAc and HSP70 in myocardium. Results Intravenous LPS significantly up-regulated the expression of O-GlcNAc and HSP70. Glutamine pretreatment further increased the expression of O-GlcNAc and HSP70. The glutamine pretreatment induced increase in O-GlcNAc and HSP70 expression was abolished by concomitant administration of alloxan. Conclusion O-GlcNAc modification may be involved in glutamine-induced myocardial HSP70 expression in endotoxic shock.
3.Treatment of Infective Ulcer Patients with Yang Syndrome by External Use of Shengji Ointment: a Multicenter Randomized Controlled Trial.

OBJECTIVETo evaluate the safety and effectiveness of Shengji Ointment (SO) (by external use) in treating infective ulcer patients with yang syndrome (YS).
METHODSA multicenter, stratified, randomized controlled clinical study was used in this study. A random digit table was formed in the ratio of 3:1,440 infective ulcer patients of YS from 5 centers were recruited, 330 in the treatment group (external use of SO, exchange once every 24 h) and 110 in the control group (external use of Votalin, exchange once every 24 h). The therapeutic course consisted of 21 days. The ulcer area, wound depth, pain degree were observed before treatment at day 3, 7, 14, and 21, and safety indices monitored at the same time.
RESULTSThe ulcer area was significantly reduced in the treatment group after 21-day medication, obviously superior to that of the control group (P = 0.002). Ulcer areas at different time points were analyzed. From the 7th day of medication the reduced area was obviously less in the treatment than in the control group (P = 0.002). Besides, the longer the more obvious difference. Compared with the control group, the healing rate of ulcer area, the pain disappearance rate, and the healing rate of ulcer depth were all obviously improved in the treatment group (all P < 0.01). No adverse event occurred in the two groups.
CONCLUSIONSExternal use of SO was safe and effective in treating infective ulcer patients with YS. It could accelerate wound healing with better analgesic effect.

4.Association of OPRMI and CYP3A gene polymorphisms with fentanyl analgesic effect on abdominal hysterec-tomy

Objective To discuss the influence of μ-opioid receptor ( OPRM1) and CYP3A gene polymor-phism on analgesic effect of fentanyl for abdominal hysterectomy patients .Methods 198 cases of gynecologic anes-thesia patients who were treated by elective abdominal hysterectomy surgery ,were selected in the hospital .The rela-tionship between the fentanyl consumption of intravenous analgesia and OPRMI and CYP 3A gene polymorphisms was detected by using polymerase chain reaction-restriction fragment length polymorphism detection .Results In 198 pa-tients,OPRM1 genotyping was 186 cases,the other 10 patients failed to typing were excluded ,including 89 cases of type A/A,type A/G 76 cases,type G/G 21 cases,OPRM1 the frequency of A118G allele was 31.7%.No statistically significant differences were found in mean VAS score of CYP 3A4*1/*1,CYP3A4*1/*1G,CYP3A4*1G/*1G instantly after operation in the three groups and 24h postoperation.By using analysis of variance with body mass ,age and intraoperative volume as a covariate factors after first 24h fentanyl consumption ,the difference was statistically sig-nificant among the three groups (P<0.05),CYP3A4*1G/*1G group was significantly lower than that in CYP3A4*1/*1G group and CYP3A4*1/*1 group,there was no significant difference between CYP 3A4*1/*1G group and CYP3A4*1/*1 group (P>0.05).In addition,because the OPRM1 A118G interacts with CYP3A4*1G, reducedthe quantity of expression of opioid receptor carrying CYP 3A4*1 and OPRM1 A118G/G,and thus more fent-anyl was needed postoperation to achieve the same effect .Conclusion It provided a theoretical basis and reference for clinical application of personalized medicine by analyzing the gynecological patients μopioid receptor gene A118G and CYP3A4*1G polymorphism.
5.Identification of the regulatory effect of evodiamine on functional regulation gene expressing in murine dendritic cell by microarrays analysis

Objective:cDNA microarrary technique was employed to detect the effects of evodiamine on functional regulation of gene expressing in murine marrow-derived dendritic cells.Methods:Immature dendritic cells (iDC) derived from bone marrow of BALB/c mice were induced by GM-CSF for ten days in vitro and received corresponding treatment for 24 h.Then the cell were randomized into the control groups (Ⅰ),evoamine group (Ⅱ),LPS group (Ⅲ) and evodiamine+LPS group (Ⅳ).After 24 h’s treatment, total RNA of the cells was collected,and cDNA microarrary (GEArray S Series Mouse Dendritic & Antigen Presenting Cell Gene Array:MM-604) was used to screen the functionalg correlation genes of dendritic cells.Results:There were seven genes up-regulated and two genes down-regulated two-folds above in Ⅱ/Ⅰ;thirty four genes up-regulated and twelve genes dwon-regulated two-folds above in Ⅲ/Ⅰ;forty six genes up-regulated and seven genes down-regulated two-folds above in Ⅳ/Ⅱ;twenty four genes up-regulated and two genes down-regulated two-folds above in Ⅳ/Ⅲ.The function of these genes involve the secret of the cytokines and the expression of their receptors,antigen uptake,antigen presentation,cell surface receptors and signal transduction.Conclusion:The effects of evodiamine on dendritic cell involve in multi-regulation and expression of genes,these genes contribute to the functions,differentiation and maturation of the dendritic cells.The work provides a potentiality to seek the target of medicine.
6.Effects of pioglitazone on cardiac fibroblast proliferation induced with high glucose and high insulin

Aim To study effects of pioglitazone on cardiac fibroblast proliferation induced with high glucose and high insulin.Methods The neonatal rat cardiac fibroblasts were cultured and treated with different factors.The cellular number was determined by crystal violet uptake;the DNA synthesis was assayed with thymidine intake method;the percentage of S+G_2+M in the cell cycle was determined by flow cytometry.Results Pioglitazone inhibited the cell number、DNA synthesis and the percentage of S+G_2+M in the cell cycle of cultured cardiac fibroblasts induced with high glucose and high insulin.Conclusion Pioglitazone inhibits cardiac fibroblast proliferation induced with high glucose and high insulin and the effective mechanism of pioglitazone on cardiac fibroblast proliferation needs further study.

Spinal biomechanics, especially the range of spine motion,has close connection with spinal surgery. The change of the range of motion (ROM) is an important indicator of diseases and injuries of spine, and the essential evaluating standards of effect of surgeries and therapies to spine. The analysis of ROM can be dated to the time of the invention of X-ray and even that before it. With the development of science and technology as well as the optimization of various types of calculation methods, diverse measuring methods have emerged, from imaging methods to non-imaging methods, from two-dimensional to three-dimensional, from measuring directly on the X-ray films to calculating automatically by computer. Analysis of ROM has made great progress, but there are some older methods cannot meet the needs of the times and disappear, some classical methods such as X-ray still have vitality. Combining different methods, three dimensions and more vivo spine research are the trend of analysis of ROM. And more and more researchers began to focus on vivo spine research. In this paper, the advantages and disadvantages of the methods utilized recently are presented through viewing recent literatures, providing reference and help for the movement analysis of spine.

The history of the World Federation of Acupuncture-Moxibustion Societies (WFAS) was reviewed through summarizing the background and process of the establishment of WFAS. The establishment background was explained in different aspects, named the recovery of acupuncture-Moxibustion in the world, the successive setup of world acupuncture-Moxibustion organizations, the divergences of International Association of Acupuncture-Moxibustion, striding forward of China reform and opening policy as well as the attention of the World Health Organization (WHO), etc. The establishment of WFAS was introduced on the proposal from eight countries, the important time of the development of acupuncture and moxibustion in China, 1984, divergences and consensus as well as the final phase. The official establishment of WFAS represents the global benefits of acupuncture-Moxibustion colleagues. It is the international organization of acupuncture and moxibustion, contributing to the promotion of acupuncture and moxibustion in the world.
