1.Effects of mild hypothermia on neuronal cell apoptosis , Bcl-2 and Bax protein expression following global cerebral ischemia-reperfusion in rats

Objective To investigate the effects of mild hypothermia (32.5 - 33.5℃) on neuronal cell apoptosis , Bcl-2 and Bax protein expression in hippocampus following global cerebral ischemia-reperfusion (I/R) in rats. Methods Twenty-four male Wistar rats weighing 250-300 g were randomly divided into three groups of 8 animals : (A) control group-normothermia-sham operation; (B) normothermia-global cerebral I/R (N-I/R); (C) hypothermia-global cerebral I/R (H-I/R) . Global cerebral I/R was produced by occlusion of bilateral common carotid arteries combined with hypotension (MAP = 40mmHg) induced by exsanguinations. Global cerebral ischemia was confirmed by dilated pupils and EEG. Global cerebral ischemia was maintained for 20 min followed by 72 h reperfusion. Mild hypothermia was induced by alcohol spreyed over the rats combined with fanning and maintained at 32.5-33.51 for 3 h. At the end of 72 h reperfusion the animals were sacrificed and hippocampus was obtained for detection of neuronal cell apoptosis (TUNEL) and Bcl-2 and Bax protein expression (immuno -histochemistry technique) . 2.Selection of Meridians and Acupoints of Tuina for Spastic Paralysis post Stroke: A Bibliometric Analysis

Objective To investigate the meridians and acupoints selection in the treatment of post-stroke spastic paralysis with Tuina (Chinese massage). Methods The literatures about Tuina therapy for post-stroke spastic paralysis were retrieved. The frequency of meridians and acupoints used was counted and analyzed. Results 99 papers were collected, which involved in 226 acupoints and 1602 times of application. The selected acupoints were distributed in all the fourteen meridians and 64.5% (1033/1602, 606 on upper limb, 427 on lower limb) of them were on the limbs. The acupoints of the Yang meridians was 75.0% (1200/1602) and the specific acupoints was 71.7% (1148/ 1602). Conclusion The acupoint selected was basically focused on the local areas in Tuina for post-stroke spastic paralysis, assisted with the involved meridians and distal acupoints. The acupoints of the Yang meridians were the first option, mainly on the extremities. The specific acupoints were the major components of the prescription, especially the Five-shu acupoints and the He acupoints.
3.Inducing Human Bone Marrow Mesenchymal Stem Cells to Cardiomyo-Like Cells with Supernatant of Normal or Injured Rat Cardiomyocytes in Vitro

Objective To observe whether human bone marrow mesenchymal stem cells (hMSCs) could differentiate into cardiomyo-like cells by culturing with supernatant of normal or injured rat cardiomyocytes (CMs) in vitro. Methods hMSCs were cultured with supernatant from normal or injured rat CMs for 27~30 d. The morphologies of induced hMSCs were observed with inverted microscope and the special cardio-markers cTnI and Desmin were identified with immunocytochemisry. Results A few cells cultured with supernatant from normal CMs enlarged and expressed cTnI, but little Desmin. While more cells cultured with supernatant from injured CMs enlarged and expressed cTnI, and parts of them expressed Desmin. The incidence of cTnI or Desmin positive cells were significantly different between these two groups (P<0.01). Conclusion Supernatant from both normal and injured CMs can induce hMSCs into cardio-like cells in vitro, and that from injured CMs is more effectively.
4.Effects of atorvastatin on treatment of patients with non-alcoholic fatty liver disease

Objective To evaluate the efficacy and safety of atorvastatin in treatment of patients with non-alcoholic fatty liver disease(NAFLD).Methods Of 72 patients with NAFLD6,8 patients with elevated alanine transaminase(ALT)and cholesterol levels at baseline had completed the study.The patients were randomly divided into treatment trial group(n=38)and control group(n=30,ithout any treatment).The patients in trial group were orally received 10 or 20 mg of atorvastatin daily according to basal serum cholesterol levels being≤or>6.5 mmol/L,respectively.The parameters including body mass index,serum cholesterol level and ALT level were tested and liver density was assessed by echography at baseline and 6 months after treatment.Results There was a significant difference in syndrome scores between baseline and 6 months after treatment[(11.05±1.29)vs.(6.08±0.87)]in trial group(t=1.96,P<0.05).Six months after treatment,11 patients in trial group were presented with normal ALT levels.whereas only 2 patients in control group had normal ALT levels (P<0.05).There was a significant differenee in cholesterol level between baseline and 6 months after treatment[(6.6±0.54)ramol/L vs.(5.4±0.5)mmol/L]in trial group(t=1.72,P<0.05).In addition,there was no differenee in body mass index between baseline and 6 months after treatment both in rial group[(26.8±2.9)kg/m~2 vs.(26.4±2.8)kg/m~2]and control group[(26.5±2.3)kg/m~2 vs.(26.4±2.2)kg/m2~](P>0.05).There was no significant difference in liver density of two groups before and after treatment(P>0.05).No side effect was reported.Conclusions Serum ALT and cholesterol levels are reduced significantly in all patients treated with atorvastatin.It is effective and safe for atorvastatin in treating NAFLD patients with hypercholesterolemia.
5.Assessment of drug sensitivity of the prostate cancer cells based on real-time cell analysis system

Objective To develop a more convenient and stable method for assessment of drug sensitivity of prostate cancer based on real-time cell analysis system as a reference for clinical treatment.Methods Human prostate cancer VCaP, DU145, PC-3, PC-3M-2B4 and PC-3M-IE8 cells were chosen to detect the sensitivity to three drugs, docetaxel, cabazitaxel and abiraterone acetate.Serial dilutions of the three drugs were used to treat the cell culture for 24 hours.The drug-induced effects on the cell lines after an incubation of 24 hours were recorded by the real-time cell analysis system to determine the half maximal inhibitory concentration (IC50).Results Docetaxel showd IC50 of 8.81 nmol/L, 11.61 nmol/L, 1.78 nmol/L, 1.44 nmol/L, 8.69 nmol/L for VCaP, DU145, PC-3, PC-3M-2B4, PC-3M-IE8 cells, respectively.Cabazitaxel showed IC50 of 3.73 nmol/L, 3.96 nmol/L, 10.41 nmol/L, 5.43 nmol/L, and 7.37 nmol/L, respectively, for the five cell lines.Abiraterone acetate showed IC50 of 8.34 μmol/L, 8.60 μmol/L, 24.20 μmol/L, 8.59 μmol/L, and 13.21 μmol/L for the five cell lines.Conclusions PC-3M-2B4 and DU145, VCaP and PC-3 cells can be used as control for docetaxel, cabazitaxel and abiraterone acetate to establish cell models for the drug screening in vitro and to provide reference for clinical applications.
6.Drug sensitivity assessment of pancreatic cancer cells by real-time cell analysis

Objective To assess the drug sensitivity of pancreatic cancer cells based on real-time cell analysis and provide a reference for individualized diagnosis and treatment of pancreatic cancer.Methods Three human pancreatic cancer cells lines SW1900, Capan-2 and PANC-1 were selected and treated with gemcitabine hydrochloride and tegafur gimeracil oteracil potassium capsules, respectively.After 24 hours of culture, the cells were treated with the two drugs in gradient concentration.The cell growth curves before and after the drug administration was monitored using a real-time cells analyzer and the growth inhibition rates (IC50) of the drugs of the pancreatic cancer cells were calculated.At the same time, the cells in the cell culture plate were treated with the drug, and acridine orange/ethidium bromide (AO/EB) staining and laser scanning confocal microscopy were performed to observe the changes of cells after the drug administration.Results 72 hours after the drug administration, IC50 values for the three cell lines were different.The IC50 values of gemcitabine hydrochloride for SW1900, Capan-2 and PANC-1 cells were 1.69 μmol/L, 10.05 μmol/L and 12.74 μmol/L, respectively.The IC50 values of tegafur capsule for SW1900, Capan-2 and PANC-1 cells were 180.29 μmol/L, 765.70 μmol/L and 95.57μmol/L, respectively.AO/EB staining confirmed the reliability of IC50.Conclusions SW1900 and Capan-2 cells can be used as the control for gemcitabine hydrochloride and tegafur gimeracil oteracil potassium capsules to establish cell models for drug screening in vitro, which provides a reference for the application of the technology in anticancer drugs screening.

Objective To discuss the effect of evaluation on research performance of a upper first-class hospital in Guangxi by comprehensive index method,and to verify the scientific and practical of this method.Methods Five indexes for evaluating the research performance were selected,and different weight to each index was endowed.The original data were translated,comprehensive index method was applied to evaluate the research performance of that hospital from 2007 to 2012.Results The final sorting results were obtained through the comprehensive index method.Of the output quality from 2007 to 2012,year 2012 ranked the first,the comprehensive index value is 2.413;and year 2007 ranked the last,the value is 0.138.Conclusions The value was high relative,which accorded with the fact,comprehensive index method can evaluate the research performance precisely and rationally.

Objective To develop a model that could roundly show the phenotypes of human alzheimer disease (AD), the triple-transgenic rat model harboring APP(Swe), PS1dE9, and TAU transgenes was established in view of the advantage of rat as an important animal model on the research of nerve system .Methods APPswe/PS1dE9/TAU triple transgenic rat AD rats were generated on a SD background by co-injecting rat pronuclei with two human genes driven by the mouse prion promoter:‘Swedish’ mutant human APP (APPsw) and exon 9 mutant human presenilin-1 (PS1dE9) and human microtubule-associated protein tau gene under the control of PDGF promoter .Transgene integration was confirmed by genotyping and expression levels were evaluated by western blot ( WB ) of brain homogenates .The pathological changes were detected by human Abeta, TAU and Phospho-PHF-TAU immunohistochemistry staining (IHC).The behavioral and cognitive changes were evaluated by Morris water maze .Results One transgenic rat lines with high human APP ( Swe ) , PS1dE9, and TAU transgenic expression was selected from three transgenic founders .Compared with the wild type rat , the transgenic rat showed significant learning and memory impairments in the Morris water maze at 6 months of age .The triple transgenic rat manifested hyperphosphorylated tau and obvious aggregation of amyloid -β( Aβ) in the brain cortex and hippocampus.Conclusion APPswe/PS1dE9/TAU triple transgenic rat AD model was established .The triple transgenic AD rat fills a critical need for a next-generation animal model to enable basic and translational AD research .
9.Study on the strategy of effective application of informationization teaching resources

Informationization teaching resources contain a large number of information so rea-sonable and effective application of the resources is of great significance to the teaching. The article tries to explore and work out some teaching strategies for effective application of informationization teaching resources through action research combined with literature research , questionnaire survey method , interview and other methods. The research process: the original teaching strategies of infor-mationization teaching resources were implemented , implicated and verified in the first round action research; the problems in the first round were summarized and analyzed and the original teaching strategies were adjusted and optimized in the next round action research; finally, the teaching effec-tiveness between the two rounds action research was evaluated and analyzed by the “classroom teach-ing effectiveness criteria based on the use of information technology teaching resources”. The results show that effective application of informationization teaching resources is of great help to the teaching effect. It can enhance the learners' interest in learning and improve their academic performance. Ap-propriate and rational teaching strategy should be provided flexibly according to the learning back-ground and characteristics of students as well as learning contents.

Objective To observe the expression and location of TF and TFPI in femoral artery atherosclerotic plaque.Methods We detected the expressions and locations of TF and TFPI in femoral artery atherosclerotic plaque by immunohistochemical and double-stain immunohistochemical method.We detected TF mRNA and TFPI mRNA expressions in atherosclerotic plaque by RT-PCR,with the normal umbilical artery as a control.ResultsThe normal umbilical artery contained little TF,TFPI and their mRNA in the adventitia.A great deal of TF,TFPI and their mRNA were found in the tunica intima of the femoral artery atherosclerotic plaque.Conclusion Expression of TF,TFPI and their mRNA of all types of cells and stroma in the proliferative tunica intima.
