Objective　To investigate the changes in the e xpressions of P53 mRNA and Bcl-2 mRNA in spermatogenic cells in rats after expo sure to ultrasound to elucidate the possible mechanisms underlying the apoptosis induced by ultrasound at genetic trascription level. Methods　Thirty-two healthy male SD rats,（30±2） days old and (70±5) grams body weight, were randomized into 4 groups (n=8) according to the time of exposure to ultrasound,including control group without exposure,gr oups of 10 minutes, 20 minutes and 30 minutes with exposure to ultrasound for 10 ,20 and 30 min,respectively. Samples were taken 24 hours after exposure to ultra sound,then 4% paraformal-dehyde fixed,paraffin-embedded and sectioned into 4 μm sections (slides were pretreated with APES and poly-lysine). P53 and Bcl-2 in situ hybridization kits were purchased from Wuhan Boshida Bio-engineering C o.Results　There was no significant difference in the expressions o f P53 mRNA or Bcl-2 mRNA in sperematogenic cells in rats between the control gr oup and 10 minutes group. The expressions of P53 mRNA in groups over 10 minutes were up-regulated and that of Bcl-2 mRNA were down-regulated. P53 mRNA in s permatogenic cells in rats was increased from (44.13±5.05)% in control group to (60.47±6.57)% in group 30 minutes. Bcl-2 mRNA was decreased f rom (40.29±9.10)% in control group to (31.01±5.67)% in group 3 0 minutes. Conclusions　Exposure to ultrasound over 10 minutes can induce up -regulatory expressions of P53 mRNA and down-regulatory expressions of Bcl-2 mRNA in spermatogenic cells in rats.

[Objective]To develop a standard closed experimental fracture model in mouse femur.[Method]A 0.45 mm stainless-steel rod was introduced into the medullary cavity and the pre-nailed femur shaft was fractured by an impact device,which resulted in a reproducible transverse fracture pattern.The fractures were examined using X-ray Micro-CT,and histological method.[Result]Transverse fractures were created in 91.67% of the experimental mice,and the fracture healing was typical secondary fracture healing.[Conclusion]The standard closed fracture model in this experiment,whose healing was typically secondary fracture healing,can be obtained easily and used in study of fracture.

Objective To evaluate the effect of combination of HANS (Han's Acupoint Nerve Stimulator) and diazepam(D) or tramadol(T) on labor pain and influence on mother and infant. Methods Two-hundred and seventy normal term pregnancy and primipara with single vertex presentation were randomly divided into six groups, HANS+D ((HANS plus diazepam 10mg, iv), HANS, D(diazepam 10 mg, iv), HANS +T (HANS plus tramadol 100mg, im), T (tramadol 100mg, im) and control group . Pain relief methods were given when the cervix dilated to 2~3 cm. Labor pain was evaluated by VAS (visual analogue scale) at different stage. Results The effective rate of analgesia for HANS+D was the highest, 90.5%, for HANS+T and HANS were 78.9% and 59.8%, respectively. The VAS of the groups combination of HANS and drugs at the time one hour after analgesia, cervix dilating 7~8 cm, second stage, were much lower than that before analgesia and that of control group (P

Objective To investigate the features of renal lesions in von Hippel-Lindau (VHL) disease and the surgical intervention. Methods The clinical and pathological data from 6 patients with renal lesions in a large Chinese VHL disease kindred were reviewed individually.One patient had multifocal renal cysts,and 5 patients had bilateral renal cell carcinomas and multifocal renal cysts. Results Among 10 kidneys in the 5 cases of bilateral renal cell carcinomas, nephron-sparing surgery (NSS) was performed in 5,radical nephrectomy (RN) in 3 and observation in 2.After operation,the pathological findings indicated that all solid lesions were renal clear cell carcinoma, with one malignant cyst being detected.The Fuhrman's nuclear grade of the RCC was grade Ⅰ in 14 tumors,grade Ⅱ in 6 tumors.The TNM stage was T 1N 0M 0 in 4 cases and T 2N 0M 0 in 1 case.During a mean follow-up of 47 months (range,9 to 113 months),all patients were alive without evidence of RCC recurrence and metastasis.Five patients had normal renal function and 1 patient experienced an increase in serum creatinine but without dialysis. Conclusions The renal lesions in VHL disease include both cysts and carcinomas,which are often multifocal and bilateral.Almost all the tumors are of clear cell type with a low stage and grade.Nephron-sparing surgery may be justified and effective in the management of VHL patients.

Objective To identify the risk factors of adverse pregnancy outcomes in expectant management of pregnant women with early onset severe pre-eclampsia (EOSP). Methods Totally, 136 gravidas, who were diagnosed as ESOP and received expectant management from January 2007 to June 2008 in Beijing Obstetrics and Gynecology Hospital, were selected and divided into two groups; the favorable pregnancy outcome group (control, n=101)and the adverse pregnancy outcome group (n=35).The general clinical information, pregnancy outcomes, routine urine test, hemodynamic data, routine blood test, liver and renal function test on admission were collected and the risk factors for adverse outcomes were retrospectively analyzed.Results (1)General clinical information; more women complained of preeclamptic symptoms on admission in the adverse outcome group than in the control group (35.6% vs.57.1 %,P< 0.05).No significant differences was found between the two groups in the maternal age, times of previous pregnancies, prevalence of concurrent complications, pre-pregnant body mass index (BMI),proportion of women who had regular antenatal checks(P > 0.05).(2) Pregnant outcomes; the average duration of expectant management in the control group were similar to the adverse outcomes group [(6.5 ± 8.2) days vs.(6.8 ±10.0) days, P > 0.05].The main complications in the adverse outcome group included placental abruption (n=13), heart failure and pulmonary edema (n=10),hemolysis, elevated liver enzymes and low platelet syndrome (HELIP syndrome, n=5),and no eclampsia was reported.However, none of these complications was reported from the control group.(3)Blood pressure and proteinuria; the gestation ages at the onset of EOSP and at delivery in the control group were earlier than those of the adverse outcome group [(31.3 ± 3.4) weeks vs.(33.0 ± 4.9) weeks, (32.1 ± 3.0) weeks vs.(34.0 ± 3.6) weeks, P< 0.05],the systolic blood pressure and urinary protein and the proportion of women with urinary protein of (+ + +)were also much higher in the adverse outcome group (all P<0.05).(4) Hemodynamics and routine blood tests; the blood viscosity in the control group was obviously lower than that of the adverse outcome group (P< 0.05 ).But there was no significant difference in the cardiac output, cardiac index, peripheral resistance and vascular compliance between the two groups (P >0.05).The adverse outcome group showed lower platelet(PLT) level and higher red blood cell(RBC) count and hematocrit compared with those of the control(all P<0.01).(5)Liver and renal function; the alanine aminotransferase (ALT), aspartate aminotransferase (AST), lactate dehydrogenase (LDH),blood urea nitrogen(BUN) in the adverse outcome group were significantly higher than those of the control group (all P<0.05), but the plasma level of total protein (TP),albumin (Alb), uric acid (UA) and creatinine (Cr) were similar between the two groups(P>0.05).(6) Risk factor analysis: RBC count (OR =3.68, 95% CI: 1.90-7.13 ),PLT count (OR=0.99,95% CI:0.98-1.00) and the gestations at delivery (OR=0.87, 95% CI: 0.80-0.94) were the risk factors of adverse pregnancy outcomes during the expectant management of EOSP.Conclusion Elevated RBC count, reduced PLT count and earlier delivery weeks are the risk factors of adverse pregnancy outcomes during the expectant management of EOSP.

AIM: To study the characteristic of liver X receptor alpha (LXRα), its target gene expression and cholesterol efflux in human macrophages treated with atorvastatin. METHODS: Human monocyte-derived macrophages were collected and cultured. Macrophages were treated with or without atorvastatin. Apolipoprotein A-I mediated human monocyte-derived macrophage cholesterol efflux was detected by liquid scintillation counting method. Reverse transcription-polymerase chain reaction (RT-PCR) was used to evaluate the mRNA expression of LXRα and some of its target genes ABCA1, SREBP2, CETP, PLTP, apoE, MMP-9 and MIP-1α. The protein expression of LXRα, ABCA1, MMP-9 and MIP-1α was determined by Western blotting. RESULTS: Pre-incubation of human monocyte-derived macrophages with atorvastatin dose dependently (1-2 μmol/L) stimulated cholesterol efflux mediated by apolipoprotein A-I. Atorvastatin also increased the mRNA expression of LXRα, ABCA1, SREBP2, CETP, PLTP, and protein expression of LXRα, ABCA1, but decreased the expression of MMP-9 and MIP-1α at both mRNA and protein levels. CONCLUSION: Atorvastatin enhances the cholesterol efflux, upregulates LXR and some genes associated with cholesterol metabolism and inhibits inflammatory responses in macrophages, indicating that statins may affect the formation of foam cells by activating LXR signaling pathway.

Objective To investigate the value of external counterpulsation and cervical traction therapy on patients with vertebral artery insufficiency syndrome. Methods Based on the criteria of clinical diagnosis, 90 patients with vertebral artery insufficiency syndrome were randomized into three groups. The group A was treated with external counterpulsation (30 cases), the group B with cervical traction (30 cases)and the group C with cervical traction plus external counterpulsation (30 cases). Results There was better therapeutic effects in group C as compared with groups A and B (P0.05). Conclusion External counterpulsation and traction therapy are effective in the treatment of the vertebral artery insufficiency syndrome.

Objective:To explore the expression levels of MHCⅡ molecules and its regulator genes CⅡTA on bleomycin-induced pulmonary fibrosis in rats,and to investigate the underlying immunologic mechanisms of pulmonary fibrosis.Methods:The rats were treated with either a single intratracheal bleomycin injection(fibrosis group)or a normal saline injection(control group).The pathologic changes of lung tissues stained with HE and Masson were observed,and the contents of hydroxyproline were detected on the 7th and 28th day respectively after bleomycin administration.The expression of MHCⅡ molecules in the lung tissues was evaluated with immunohistochemistry techniques,and the percentage of MHCⅡ+ cells was measured.The amounts of total CⅡTA and typeⅠ,Ⅲ and Ⅳ CⅡTA mRNA of lung tissues were measured by real-time PCR using Taqman probe.Results:(1)The percentage of MHCⅡ+ cells in lung tissues increased significantly in fibrosis group compared with that of control group on the 7th day and the 28th day[(0.10?0.03)vs(0.06?0.02),P0.05];In fibrosis group,type Ⅳ CⅡTA mRNA was 667.3% [(2.98?0.92)vs(0.39?0.15),P

Objective Toinvestigatetheeffectofpowerelectricalstimulation(PES)incombination withtask-specifictrainingonlowerextremitymotorfunctioninstrokepatientswithfootdrop.Methods Fifty patients with poststroke foot drop were enrolled retrospectively. They were divided into either an experimental group or a control group (n=25 in each group). The control group received routine rehabilitation treatment. On this basis,the experimental group was given PES in combination with task-oriented training. Both groups of patients were trained 2 times a day,once for 40 minutes,5 days a week for 6 weeks. The ankle active range of motion,plantar flexor muscle tension,and tibialis anterior muscle strength were determined before and after treatment. The balance and lower extremity motor function of the patients were assessed by using Berg balance scale score,Fugl-Meyer lower extremity score,modified Barthel index,and Holden walk grading. At thesametime,thewalkingspeedandsteplengthbeforeandaftertreatmentwerecompared.Results Six weeks after treatment,the ankle activity,plantar flexors tension,and tibialis anterior muscle strength scores in patients of the experimental group were improved compared with before treatment,and each indicator was significant better than the control group (t=6. 261,-6. 163,and 2. 968,respectively;all P<0. 05). Berg balance scale,walking speed and step length scores were also improved as compared with before treatment, and each indicator was better than the control group (t=10. 733,9. 074,and 9. 013,respectively;all P<0.01). The lower limb motion scores,modified Barthel indexes,and Holden walk grading scores were improved compared with before treatment,and each indicator was significantly better than the control group (t=3.261,7.573,and4.010,respectively;allP<0.05).Conclusion Usingpowerelectricalstimulation in combination with task-oriented training may effectively improve the lower extremity motor function in stroke patients with foot drop.

BACKGROUND:Vascular endothelial growth factor (VEGF) is a crucial factor for regulating bone marrow mesenchymal stem cells in microenvironment, which can promote endothelial differentiation of bone marrow mesenchymal stem cells. But there is no report about VEGF effect on regulating the proliferation and osteogenic differentiation of bone marrow mesenchymal stem cells. OBJECTIVE:To investigate the regulatory role of VEGF in the proliferation and osteogenic differentiation of bone marrow mesenchymal stem cells. METHODS:The bone marrow mesenchymal stem cells were isolated and cultured from mouse long bones. cellCounting Kit-8 was used to test the proliferation of bone marrow mesenchymal stem cells cultured with different concentrations of recombinant VEGF proteins. The appropriate concentration of VEGF recombinant protein was selected to test their effects on the osteogenic differentiation of bone marrow mesenchymal stem cells. The mRNA and protein levels of Osterix, Runx2, alkaline phosphatase, osteocalcin and heme oxygenase-1 in bone marrow mesenchymal stem cells under induction of VEGF were detected by molecular biology methods. RESULTS AND CONCLUSION:(1) VEGF promoted the proliferation of bone marrow mesenchymal stem cells dose-dependently, and 100μg/L was the optimum concentration. (2) VEGF promoted the expression of Osterix, Runx2, alkaline phosphatase and osteocalcin in bone marrow mesenchymal stem cells under osteogenic induction. Similar results were obtained by alizarin red staining and quantification of numbers of calcium nodules. (3) VEGF induced the expression of heme oxygenase-1 in bone marrow mesenchymal stem cells at mRNA and protein levels. These findings indicate that VEGF can induce the expression of heme oxygenase-1 i in bone marrow mesenchymal stem cells, and promote the proliferation and osteogenic differentiation of bone marrow mesenchymal stem cells.
