5.A review on microRNAs regulation in osteoclast differentiation and function.

MicroRNAs (miRNAs) are small non-coding RNAs that contain 22 to 25 nucleotides and play important roles in post-transcriptional regulation of target genes. MiRNAs are involved in cell proliferation, differentiation, apoptosis, inflammation, and cancer development. Alveolar bone resorption is the main clinical manifestation of periodontitis. Osteoclasts are unique cells regulated by osteoblasts and inflammatory cytokines and are responsible for bone resorption in periodontitis. Recently, miRNAs have emerged as an important regulator of osteoclast differentiation and function. In this study, we review the recent research progress on the effects of miRNAs on osteoclast differentiation and function, particularly the mechanisms of miRNAs-mediated osteoclast formation and bone loss in periodontitis.

6.Influence of Tongluo Tangtai Recipe on Inflammatory Cytokines in Rats with Diabetic Peripheral Neuropathy

Objective To investigate the effect of Tongluo Tangtai Recipe(TTR)on inflammatory cytokines in rats with diabetic peripheral neuropathy(DPN). Methods Rat model of spontaneous DPN was established by feeding high fat diet. The rats were divided into 5 groups , namely model group , western medicine(metformin 100 mg·kg-1·d-1+ methycobal 0.2 mg·kg-1·d-1) group, and low-, middle- and high-dose TTR groups (TTR in the dosage of 0.72, 1.44, 2.88 g·kg-1·d-1 respectively). Rats of the normal group and model group were treated with normal saline orally, and the medication groups were given the corresponding medicine according to the experimental design. After intragastric administration for 4 weeks, sciatic motor/sensory nerve conduction velocity was detected with electrophysiological examination method, and tumor necrosis factor alpha(TNF-α), interleukin-6(IL-6), C-reactive protein(CRP), myelin basic protein(MBP) levels in the serum were examined by enzyme-linked immunosorbent assay(ELISA). Results TTR in various dosages could reduce the serum levels of MBP and inflammatory cytokines of TNF-α, IL-6 and CRP, and increase sciatic nerve conduction velocity (P<0.05 or P<0.01 compared with those in the model group), the high dose showing the strongest effect. Conclusion TTR could inhibit inflammation reaction and nerve demyelination, and improve nerve structure and conduction function, which may contribute to the preventive and therapeutic mechanism of TTR for DPN.
7.EXTRACTION AND STRUCTURAL STUDY OF SCHIZOPHYLLAN FROM SCHIZOPHYLLUM COMMUNE BY SUBMERGED CULTIVATION

The extraction method and structure of exo-polysoccharide from schizophyllum commune by submerged cultivation were studied. Proteins were removed com p letely from the polysoccharide by the sevag method following the isoelectric poi nt precipitation method. The purified schizophyllan was proved to be homogeneou s with molecular weight 4?10 4D by sephadex G-200 column chromatography, PAGE and HPLC. Its monomer was determined by hydrolysis, PC, GC and its structure wa s analyzed by IR, enzymolysis, periodate oxidation, the results showed that schi zophyllan was only composed of glucose and it was the ?-glucan consisting of ?-(1-3) and ?-(1-6) glucosidic linkages.
8.PRELIMINARY STUDY ON MONOCLONAL ANTIBODIES AGAINST CYSTICERCUS CELLULOSAE

In order to produce valuable monoclonal antibodies for immunodiagnosis of cysticerco-sis, hybridoma technique was adopted using the fusion of splenic lymphocytes from BALB/c mice immunized with Cysticercus cellulosae antigens and the myeloma cell line NS-1. Through screening and cloning, three monoclonal antibody cell lines have been established. Im- munoglobulin subclasses of 3 lines were determined to be IgGl. These monoclonal antibodies were found to be highly specific and without cross reaction with Echinococcus granulosus hy-datid cyst and Cysticercus fasciolasis. Antigen fractions specifically reacting to monoclonal antibodies were demonstrated with Western blot method. The results showed that specific reaction bands of SCW located in 25 kDa and 17 kDa regions.
9.Expression and Significance of RXR_? in Gastric Carcinoma

Objective To detect the expression of RXR? gene protein in gastric cancer, superficial gastritis, normal gastric mucosa, in order to explore the relationship between the development of gastric cancer, combined with clinical and pathological analysis of its clinical significance. Methods Using immunohistochemical detection RXR? protein in gastric cancer, superficial gastritis, gastric mucosa in the normal expression. Results RXR? protein mainly located in the nucleus of cells, there are a small number of cytoplasmic staining, a brown or yellow granular, in gastric cancer, superficial gastritis, normal gastric mucosa, RXR? protein expression rates were 25.0%, 81.1%, 96.1%; group and the normal mucosa and superficial gastritis, gastric cancer group RXR? expression rate has dropped, there were significant differences (P0.05).
