1.Establishment of fingerprint of processing Gentianae scraba Bge and its mutual mode by HPLC

AIM:To establish the HPLC fingerprint of methanol-resoluble component of processing Gentiana scraba Bge collected from different habits to provide evidence for quality control and quality evaluation of Gentiana scraba Bge.METHODS:Ten batches of processing Gentiana scraba Bge were measured by RP-HPLC,and the mutual mode was made by the similarity degree evaluating system 2004 of Chromatographic Fingerprint of Traditional Chinese Medicine promulgated by Chromatographic condition,consisted of Agilent-Eclipse-XDB-C_ 18-column and gradient elution with methanol-water,the detection wavelength was at 254 nm,flow rate was at 1.0 mL/min and the column temperature was at 20 ℃.RESULTS:The mutual mode of processing Gentiana scraba Bge was set up and the similarity of fingerprints of processing Gentiana scraba Bge from different habits was gained.CONCLUSION:According to the analysis of statistics,this method is steady,accurate with good repeatability and can be used as standardization implantation and quality control of Gentiana scraba Bge.
2.Risk factors of under staging and under grading of patients undergoing radical prostatectomy

Objective To determine the relevant factors of under staging and under grading in patients undergoing radical prostatectomy and to define important predictors of under staging in this population. Methods The clinical T stages and biopsy Gleason scores were compared with the pathological T stages and prostatectomy Gleason scores in 55 patients. Results Among the 55 cases,50 were preoperatively classified as clinical stages T 1~T 2,of whom 21(42%) were postoperatively classified as pathological stages T 3~T 4.Of the 26 cases who preoperatively had the Gleason scores of 2 to 6,11(42%) had postoperatively pathological grades of 7 to 10.Logistic regression analysis revealed that PSA( P =0.0159)at diagnosis and the percentage of positive biopsies of prostate ≥50%( P =0.0013) were significant predictors for preoperative clinical staging lower than postoperative pathological staging. Conclusions The results demonstrate that patients who were classified as clinical stages T 1~T 2 with PSA≥20 ng/ml and percentage of positive biopsies≥50% may have high possibility of under staging.
3.Effect of polypeptides from ehlamys farreri on human umbilical cord stem blood cells differentiation in vitro

5.Inhibition effect of celecoxib on the experimental choroidal neovascularization

Objective To observe the inhibition effect of selective cyclooxygenase-2 inhibitor (celecoxib) on the experimental choroidal neovascularization (CNV).Methods Thirty 8-10 weeks old healthy male Brown-Norway(BN) rats were randomly divided into the control,laser and celecoxib group,with 10 rats in each group.At the dosage of 50 mg/kg,celecoxib was garaged twice per day.After 7 days,experimental CNV was induced by Krypon laser on laser group and celecoxib group.Fundus fluorescein angiography (FFA) was performed on days 3,7,14,21,30 after laser photocoagulation.On days 21 after photocoagulation,5 rats in each group were sacrificed and the relative thickness of CNV membranes,the expression of COX-2,vascular endothelial growth factor(VEGF) and matrix metalloproteinase-2 (MMP-2) Were studied by histopathologic or immunohistochemistry examination.Results On days 21 after photocoagulation,the incidence of CNV in the celecoxib group is significantly lower than that in the laser group (X~2=7.1068,P=0v0077);the relative thickness of the CNV membranes in the celecoxib group is reduced 41.38% compared to the laser group,the difference is statistically significant (t=16.7600,P=0.0000).COX-2,VEGF and MMP-2 expression in the CNV membrane of celecoxib group were significantly lower than in control group (t=5.7100,5.8400,8.0200;P=0.0000);the COX-2,VEGF and MMP-2 expressions in choroid and retina of control group were weak.Conclusion Prophylactic celecoxib can reduce the expression of VEGF and MMP-2 by inhibiting COX-2,and prevent the CNV induced by laser photocoagulation.
6.Cytokine-induced Killer Cell Activation by Dentritic Cells Loaded with the Tumor Antigen from Human Lung Cancer Cell Line A549

Objective To explore the specific anti-tumor effect of cytokine-induced killer cell (CIK) activation by dentritic cells (DC) loaded with the tumor antigen from human lung cancer cell line A549. Methods CIKs and DCs derived from the health donor's peripheral blood monocyte (PBMC) were isolated and induced by the cytokine in vitro. DCs were generated by culture of adherent cells from PBMC for 7days in the presence of IL-4 and GM-CSF. CIKs were generated by culture of non-adherent cells for 7 or 14 days in the presence of INF-?, IL-2, IL-1?, and mouse anti-human CD3 monoclonal antibody. The phenotype of DC and CIK was analyzed by FCM. The activity of DC was evaluated by MLR; and the cytotoxicity of CIK was assayed by MTT. The CIK expression profile of cytokine and signal transduction genes were detected by the DNA micro-array under different circumstances. Results Phenotypic analysis indicated that CD3+CD56+NKT cells were predominant in the CIK cells generated in the current study, and that DC cells expressed high levels of CD40, CD80, CD86 and HLA-DR. As demonstrated by MTT assay, the CIK cells proliferated rapidly, and the DC cells were able to induce an MLR. Both antigen-pulsed and unpulsed DC stimulated the proliferation of CIK cells, and no significant difference was found between the two kinds of DC cells. Unpulsed DC cells did not enhance cytotoxicity mediated by CIK cells even though they were able to stimulate CIK proliferation. Antigen-pulsed DC, however, stimulated CIK cells to specifically kill target cells. The specific antitumor effect was also observed in nude mice bearing tumors. Conclusion One of the major effects of antigen-pulsed DC is to activate CIK cells and to make them specific in killing tumor cells.
7.Clinical analysis of otogenic Mouret abscess: a case report.

Mouret abscess is a rare extracranial complication of suppurative otitis media. It is generally believed to be a deep neck abscess caused by inflammation leading to the rupture of the bony tip of the mastoid tip. The location of Mouret abscess is deep. The symptoms are insidious at the onset, but may eventually spread to the surrounding tissue, and even lead to mediastinal abscess, cavernous sinus thrombosis, meningitis, dyspnea and other serious complications. At present, with the popularization of antibiotics, the occurrence rate of Mouret abscess is very low, and only sporadic cases have been reported.In this paper, a case of Mouret abscess caused by cholesteatoma was analyzed to explore Mouret abscess in terms of the route of infection, clinical manifestations, imaging features, diagnosis and treatment.

9.Relationship between remuneration incentive and satisfaction of medical staff in county public hospital:Based on questionnaire survey in Heilongjiang Province

Objective: To understand the effects of remuneration incentive and their influences on job satis-faction in county public hospital. Methods: Stratified random sampling was adopted. Six county public hospitals in county level in Heilongjiang province were selected as the sample. Exploratory factor analysis was used to ana-lyze the remuneration incentives, descriptive statistical analysis and logistic regression analysis were used to ana-lyze the effects and their influences on work satisfaction. Results: remuneration incentives are divided into four dimensions that comprised a total of 16 items, which including direct material rewards, indirect material re-wards, occupational rewards and social rewards. The surgeon’s job satisfaction is higher than the physician, and is lower than the obstetrics and gynecology and pediatrics. Technician and nurses’ satisfaction is lower than the general practitioner. The medical staff who worked less than5 years has the lowest satisfaction. Authorized staff’s satisfaction is lower than off-staff. Direct material rewards and occupational rewards are related to job satisfac-tion. Conclusions:Material rewards were in low level, and non-economic incentive was insufficient. The salary incentive for highly educated talents should be strengthened.
10.Effects of peritoneal jet ventilation on peritoneal oxygenation in pigs

Objective To evaluate the effects of peritoneal jet ventilation on peritoneal oxygenation in pigs.Methods Twenty-four pigs of both sexes (12-16 weeks,35-45 kg) were randomly divided into 3 groups (n=8 each): sham operation group (group S) ; peritoneal regular frequency jet ventilation group (16 bpm)(group N) and peritoneal high-frequency jet ventilation group (150 bpm) (group H).Oral tracheal intubation was performed.The animals were mechanically ventilated (VT 8-12 ml/kg,RR 12-16 bpm,I:E 1.0:1.5,FiO2 100%) via airway.Endotracheal tubes were inserted into abdomen through the incisions in the left lower (for peritoneal jet ventilation) and right upper quadrant (for outlet of air).Arterial blood PaO2 and PaCO2 were measured before (baseline) and at 30,60,90,120,150,180,210 and 240 s of peritoneal jet ventilation.Peritoneal jet ventilation was started after the lungs being mechanically ventilated for 35 min.Peritoneal jet ventilation was terminated when SpO2 dropped to SpO2 < 90 %.The duration of safe apnea (DSA,from the moment of begging of peritoneal jet ventilation to the time when PaO2 < 60 mm Hg).Results PaO2 was significandy higher and DSA longer in group H than in groups S and N.But there was no significant difference in PaCO2 among the 3 groups.Conclusion Peritoneal high-frequency jet ventilation can significantly enhance the efficiency of peritoneal oxygenation and prolong DSA,while peritoneal regular frequency jet ventilation has no effect on peritoneal oxygenation.
