1.Optimization of Fermentation Medium with Lactose as an Inducer for High Molecular Weight Recombinant Spider Silk Protein

Based on M9 culture medium,the concentration of ingredients of culture medium was optimized for the fermentation of pNSR32/BL21(DE3),the engineering bacterial with spider silk protein,and lactose as an inducer.The condition of optimum culture medium was obtained for the expression of the high molecular weight recombinant spider silk protein by using orthogonal and individual factor experimental design.The result was showed that the optimum culture medium was consisted of 0.3% glycerol,3% yeast,0.75% tryptone,0.05%(NH_4)_2SO_4 and a little inorganic salt_.It was confirmed that the optimum culture medium will benefit the growth of bacterial and expression of recombinant spider silk protein.The production level of propose protein will attain at 20% of the total proteins in the fermentation.
3.Evaluation of community health service institutions responsiveness in Henan Province:Key in-formant survey

Objective:To understand the responsiveness of community health service institutions in Henan Province. Methods:This research collected information through questionnaire investigations. The object of the investigations is key informants familiar with community health service institutions. Various dimensions of responsiveness within Henan province community health service institutions are determined through a multiple-input pecking order graph. The fuzzy comprehen-sive evaluation method is used to evaluate the level of responsiveness. Responsiveness inequality index is used to measure the distribution of reactions. Results:A total of 431 valid key informants were surveyed. The various dimensions of the questionnaire of Cronbach alpha coefficient was greater than 0. 623;the total scale of the Cronbach alpha coefficient was 0. 850;the correlation coefficient of each dimension of test-retest reliability were greater than 0. 907. The levels of responsive-ness and distribution index were 8. 33 and 0. 2744, respectively;each dimension by order of importance:timely attention>dignity>infrastructure>communication>confidentiality >social support >selective >autonomy. Approximately 50% of key informants considered that vulnerable groups in community health service institutions were sometimes or often treated unfairly. Conclusion:Key informants considered the responsiveness level of overall higher community health service institu-tions in Henan Province was high, and the distribution of responsiveness was relative equilibrium. However, there is a large shortage of infrastructure, privacy protection, doctor-patient communication, etc.
4.Recombinant adenovirus vector-delivered report gene expression in rats kidney through different ways

Objective To investigate expression of recombinant adenovirus(rAV) vector-mediated report gene in kidney, and to seek for the best passway of rAV vector-mediated gene transfer in vivo.Methods Thirty Sprague-Dawley(SD) rats were divided into 5 groups randomly. rAV-GFP(1013viral particales?L-1) was injected through different ways. 2, 7 and 14 d after injection, GFP expression in left kidney, right kidney, liver and lung was evaluated by fluorescence microscope and computer software. Results GFP gene expressed in kidey and reached peak 7 d after injection expression. Conclusion rAV vector-mediated report gene can be transducted in kidney. The rAV-mediated transgene expression in kidney is a potential strategy in the treatment of renal diseases.
5.Effects of Drug Combined with Psychological Therapy on Mood Disorders of Patients with Epilepsy

Objective To investigate the effects of mirtazapine ccombined with psychological treatment on depression of patients with epilepsy.Methods 102 patients with epilepsy and complicated with depression were divided into the control group and trial group with 51 cases in each group.The control group received conventional mirtazapine and the trial group received mirtazapine combined with psychological therapy.The course was six weeks.The follow-up was performed six months after treatment and all patients were evaluated with Hamilton Depression Scale(HAMD)and Clinical Global Impression(CGI-SI).The side effects were evaluated with Treatment Emergent Symptom Scale(TESS).Results After the treatment of six weeks,the HAMD and CGI-SI scores of patients of the two groups significantly increased(P<0.05)and the scores of the trial group increased more significantly than that of the cintrol group(P<0.05)six months later.Conclusion The effect of mirtazapine combined with psychological therapy on depression of patients with epilepsy is obvious.
6.Clinical study of high-dose ambroxol for treatment of acute exacerbations in chronic obstructive pulmonary disease

Objective To evaluate the efficacy and safety of high-dose ambroxol in treating acute exacerbations in chronic obstructive pulmonary disease (AECOPD).Methods The trial design was random,open,and parallel control.126 patients with AECOPD were involved.High-dose group received ambroxol 150 mg 3 times per day for 7 days (n=46),while conventional-dose group given ambroxol 30 mg 3 times per day for 7 days (n=40) and control group was given a routine treatment (antibiotic therapy,oxygen inhalation and bronchodilators) (n=40).Results The effective rates of high-dose group and conventional-dose group (95.6 %,77.5 %) were significantly higher compared with control group (55.0%) (x2 =19.81,4.53,both P<0.05).The average duration of hospitalization was significantly lower in high-dose group (10.78 ±0.95)d and conventional-dose group (11.75±0.84)d comparing with control group (13.15±0.89)d (q=11.88,7.24,both P<0.05).The clinical efficacy in high dose group was much better.Conclusions High-dose ambroxol is a safe and efficient therapy in the treatment of acute exacerbations in chronic obstructive pulmonary disease.
7.Effect of Budesonide on the IL-16 in Sputum from Patients With Asthma

Objective To inestigate the effect of inhaled steroid hormone on the IL-16 in sputum from paitents with asthma, and to explore the mechanism of anti-inflammation of steroid. Method Budesonid inhalant were administered(400?g twice a day) persistent 4 weeks for 12 subjects with mild to moderate asthma, The percentage of eosinophils and IL-16 in induced sputum were determined in every subject .Result After the course , the concentration of IL-16 in sputum was (41 5?5 2)ng/L, significantly lower than that before the therapy which was (58 2?7 2)ng/L,P

9.Effects of compound Yi-Zhi on D-galactose-induced learning and memory deficits in mice1

AIM: To explore the effects of Compound Yi-Zhi (YZC) on learning and memory capacity and free radical metabolism in D-galactose induced mice dementia model. METHODS: The mice dementia model was induced by a daily D-galactose 0.15 g?kg -1 sc for 45 d and after 5 d D-galactose injection, the mice were treated with three doses of YZC once a day for 40 d. In order to find out the influence of YZC on the models learning and memory capability and free radical metabolism system, Y-maze test was introduced for the former and superoxide dismutase (SOD) and malondialdehyde (MDA) tests for the latter. RESULTS: Compared with the D-galactose model group, three doses of YZC administration ( 0.108, 0.217, 0.433 g?kg -1) were all shown to significantly reduced the times of learning/memory in Y-maze test from 31.35? 8.97/ 17.12? 5.57 to 19.67? 8.07/ 11.27? 6.65, to 14.00? 6.84/ 8.29? 5.95, and to 11.41? 5.99/ 6.24? 4.97, respectively. For SOD, the administrations of 0.108 and 0.433 g?kg -1 YZC could increase the activity of SOD from 17.69? 3.14 to 26.94? 4.46, and to 21.33? 3.63 nU?mg -1 pro. While for MDA, all the three doses of YZC could decrease the level of MDA from 4.08? 0.88 to 2.82? 0.75, to 2.10? 0.42, and to 2.35? 0.39 nmol?mg -1 pro in the mice brain tissue, respectively. CONCLUSION: YZC can improve the D-galactose induced learning and memory dysfunction, and the action may be related to the improvement of antioxidase activity in the model mice brains.
