1.Appropriate concentration of ropivacaine for differential sensory and motor block of brachial plexus

Objective To determine the appropriate concentration of ropivacaine for differential sensory and motor block of brachial plexus.Methods Ninety ASA Ⅰ - Ⅲ patients aged 16-75 yr weighing 40-85 kg undergoing upper extremity operation under axillary brachial plexus block were randomly divided into 3 groups according to the concentration of ropivacaine used for the block(n = 30 each): group A 0.15% ropivacaine;group B 0.10% ropivacaine and group C 0.05% ropivacaine.Axillary brachial plexus block was performed using ultrasound guidance and electric nerve stimulation.Sensory and motor block were assessed and recorded at 10,30,60and 240 min after local anesthetic injection(T1-4).The rate of adequate sensory block,the rate of differential sensory and motor block(the areas innervated by radial,median and ulnar nerves were numb but the patients could still move their elbow,wrist and fingers)and effectiveness of the block(excellent - completely no pain;good slight pain,iv fentanyl was needed;poor -rescue brachial plexus block was needed or general anesthesia was induced).Operation time and duration of analgesia and success rate of the block were recorded.Results 0.15%ropivacaine produced excellent or good block and no failure in group A.The rate of differential sensory and motor block was significantly higher in group B(0.10% ropivacaine)than in group A.The effect of block with 0.05% ropivacaine was unsatisfactory in group C.Conclusion Axillary brachial plexus block with 0.10% ropivacaine can induce differential sensory and motor block in the majority of patients.
2.Application of placental mesenchymal stem cells

BACKGROUND:Placental mesenchymal stem cels are becoming a new source of seed cels because of wide range of sources, low immunogenicity and not involving ethical issues.
OBJECTIVE:To elaborate the sources, biological characteristics and latest application of placental mesenchymal stem cels.
METHODS:Literature search was performed in PubMed, ScienceDirect, OvidSP, CNKI databases for relevant literatures published from 2003 to 2015. The key words were “placenta, mesenchymal stem cels, placenta mesenchymal stem cels, cel transplantation, application mechanism” in Chinese and English, respectively. Then, 57 papers were further analyzed and reviewed in line with the theme.
RESULTS AND CONCLUSION:Placental mesenchymal stem cels have been isolated and cultured successfuly, and confirmed to have multi-differentiation potential. A large number of placental mesenchymal stem cels have been used in the experimental animal and clinical researches, and they have a great potential in bone tissue engineering, revascularizaion and nerve repair. However, the specific mechanism underlying the application of placental mesenchymal stem cels is not clear. In order to ensure the safety and effectiveness, there are stil many problems to be further studied before placental mesenchymal stem cels are widely used in clinic.
3.The Application of Countercurrent Chromatography with Aqueous Two Phase System in the Separation of Proteins

Aqueous two phase system(ATPS) provides a gentle, non-denaturing separation environment for proteins, enzymes. While high-speed countercurrent chromatography (HSCCC) is a liquid-liquid partition chromatography which uses centrifugal force to hold the stationary phase and facilities the mobile phase partitioning through the stationary phase, it can produce high separation efficiency with large sample loading capacity. However, the ordinary HSCCC apparatus (Type J ) fails to retention a satisfactory stationary phase of ATPS because of its high viscosity and low interfacial tension. Nevertheless, the later designed cross-axis planetary centrifuge system(X-CPC) can produce a greater lateral force field and enhances significantly the retention of the ATPS stationary phase. A review of the application of these CCC techniques with ATPS in the separation of proteins was given. Meanwhile, new techniques such as pH-peak focusing CCC and dye-ligand affinity CCC and some new CCC column design for improvement of separation efficiency and retention of ATPS stationary phase are introduced.
4.Synergistic protective effect of picroside Ⅱ and NGF on PC12 cells against oxidative stress induced by H2O2

AIM: To study the synergistic protective effect of picrosideII and NGF for the oxidative stress on PC12 cells induced by hydrogen peroxide (H2O2). METHODS: The fluorescent probe 6-carboxy-2',7'-dichlorodihydrofluorescein (CDCFH) was used to assess the intracellular reactiveoxygen species (ROS) level, and MTT assay, morphological observation as well aslactate dehydrogenase (LDH) leakage were conducted to measure cellular injury. RESULTS: The H2O2-induced cytotoxicity was significantly attenuated in the presence of picroside II (25 μg/mL) and NGF (2 ng/mL). Cultures with this combined treatment possessed decreased level of ROS while increased cell survival, as compared to that of picroside II or NGF alone-treated cells. Accordingly, it was concluded that their synergistic protective activities against oxidative stress in vitro were demonstrated in various aspects including reversing morphological changes, enhancingthe ability of cell proliferation and ROS scavenging. CONCLUSION: Such action supports the therapeutic potential of picroside II and NGF in treating nervous disorders based on their synergistic effect.
5.The dose rate response characterization of four kinds of dosimeters for linear accelerator

Objective To explore the dose rate response characterization of four kinds of dosimeters for clinical application.Methods Within the range of 100-600 cGy/min,the dose rate responses of the PTW 0.6 cm3 ion chamber,0.015 cm3 ion chamber,Matrixx Evolution 2D diode array and MapCHECK 2D diode array under the same measuring conditions were measured.The dose rate response of the PTW 0.6 cm3 ion chamber under different energy and working voltage were analyzed.Results All ionization chambe.r types of measured equipment showed certain dose rate dependence for 6 MV X-rays.All the differences were below 1%.The dose rate dependence disappeared for 15 MV X-rays.The 2D diode array had strong dose rate dependence and the response difference was about 2%.Conclusions It is necessary to test and analyze the dose rate response of the measured equipment in treatment technology with dose rate varying,in order to ensure the precision of daily calibration and dose verification.
6.Mental disorder and its risk factors in first degree relatives of patients with type 2 diabetes mellitus

Objective To investigate the incidence and the risk factors of anxiety or depression and in the first degree relatives (FDRs) of patients with type 2 diabetes mellitus ( T2DM ). Methods A total of 102 T2DM patients who visited our hospital from May to July 2010 and their FDRs completed SAS,SDS,and life satisfaction evaluation. The medical history of T2DM patients was collected and compared with their FDRs with or without mental disorders. The correlations of risk factors with anxiety or depression were analyzed, t test or X2 test were used for statistical analysis and a P value less than 0. 05 was considered statistically significant. Results The incidence of anxiety or depression of FDRs was 31.4% and 17.6%,respectively. The life satisfaction score of FDRs with mental disorders was significantly decreased ( t = 4. 26 and 4. 09, P<0. 05 ). Fasting glucose level of T2DM patients with anxious or depressive FDRs was significantly increased ( t = -2. 48 and -2. 15, P<0. 05 ). Higher medical cost and multiple treatment strategies were positively correlated with SAS and SDS score of the FDRs. Conclusion Higher incidence of mental disorders and lower satisfaction scores could be found in the FDRs of T2DM patients. Better control of blood glucose and reduced treatment cost and strategies may be useful in improving metal condition and life satisfaction of the FDRs of T2DM patients.
7.Application of low tube voltage and low volume contrast medium protocol on 128-slice dual-source CT in children with tetralogy of Fallot

Objective To investigate the effectiveness and feasibility of cardiovascular computed tomography angiography(CTA) in 128-slice DSCT with low tube voltage and low dosage contrast media in children with tetralogy of Fallot (TOF). Methods Forty patients with TOF were randomly divided into group A and group B by random number table method, patients in group A received a conventional scan with 80 kVp and contrast media of 1.2 ml/kg, patients in group B, 70 kVp and contrast media of 1.0 ml/kg were used. The injection time of the two groups were both fixed for 12 s. CT attenuation, image noise and signal-to-noise ratio (SNR) of ascending aorta, the main pulmonary artery, left ventricle and right ventricle were quantified. Radiation dose and volume of the contrast medium were recorded. Subjective image quality was assessed by two radiologists in consensus. The Student's t test was performed to analyse the differences between the two groups regarding CT attenuation, image noise, SNR, radiation dose and volume of the contrast medium. The image quality scores between the two groups were compared by using the Mann-Whitney U test. Results No significant difference was found in the attenuation, noise, SNR between the two groups in the same evaluated anatomic regions and no significant difference was found in the image quality. Effective dose (ED) was(0.17±0.05),(0.13±0.04)mSv respectively, there was significant reduction in group B than that in group A (t=2.48, P=0.019). The consumed iodine amount was(10.00±1.84),(8.29± 1.45) ml respectively, there was significant reduction between the two groups (t=2.89, P=0.007). Conclusions In children with TOF, the cardiovascular CTA with diagnostic quality can be adequately acquired with low tube voltage (70 kVp) and low concentration contrast media (1.0 ml/kg), there is significant reduction in radiation dose and contrast medium amount.
8.Screening of host proteins interacting with GP, VP30, NP,and L proteins of Ebola virus by yeast two-hybrid system

Objective To study the biological function of GP,VP30,NP and L proteins in Ebola virus(EBOV) infectious diseases,and to screen the host proteins which interact with GP,VP30,NP,and L proteins of EBOV from human liver cDNA library using yeast two-hybrid technique.Methods Recombinant PCR was used to construct bait plasmids pGBKT7-GP,pGBKT7-VP30,pGBKT7-NP and pGBKT7-L.Bait strains were combined with human liver cDNA library strains.The positive clones were analyzed by DNA sequencing and bioinformatics.A yeast recovery experiment was performed to further verify and exclude false positive results.Results We constructed bait plasmids pGBKT7-GP,pGBKT7-VP30,pGBKT7-NP and pGBKT7-L with the recombinant PCR method.Six host proteins which could interact with GP,VP30,NP,and L proteins were screened,including COMMD1,ALB,PSMD8,APOA2,CYP2E1,and HP.The yeast recovery experiment proved that COMMD1 and APOA2 might interact with NP protein.Conclusion A number of prey proteins which interact with GP,VP30,NP,and L proteins of EBOV are screened,which may provide reference for the research of EBOV.
9.Diagnostic values of normalized ADC value in benign and malignant breast lesions

Objective To evaluate the values of DWI normalized ADC value in breast MRI in the differential diagnosis of benign and malignant breast lesions.Methods The absolute value of ADC,normalized ADC value and T1 WI dynamic contrast-enhanced TIC were analyzed in 49 patients with benign and malignant breast lesions;the efficiencies of single method diagnosis and combined diagnosis were assessed.Results When b=1 000 s/mm2 ,the absolute value of ADC <1.25×10 -3 mm2/s was taken as the thresh-old value for the diagnosis of malignant breast lesions,the area under the ROC curve was 0.818,the sensitivity was 73.9% and the specificity was 84.6%;the normalized ADC value <0.55 × 10 -3 mm2/s was taken as the diagnostic threshold,the area under the ROC curve was 0.901,the sensitivity was 87.0% and the specificity was 88.5%,there was a significant difference between both values.The lesions with a wash-out type of T1 WI dynamic contrast-enhanced TIC were diagnosed as malignant lesions,the sensitivi-ty was 89.5% and the specificity was 70.0%.The absolute value of ADC and T1 WI dynamic contrast-enhanced TIC were applied combinedly,the predicted percentage correct was 81.6% for the diagnosis of breast lesions,while the normalized ADC value and T1 WI dynamic contrast-enhanced TIC were applied combinedly,the predicted percentage correct was increased,which was 89.8%. Conclusion Normalized ADC value has a higher sensitivity and specificity in differentiating benign and malignant breast lesions,its combined application with T1 WI dynamic contrast-enhanced TIC can improve the diagnostic performance of breast lesions.
10.Research progress of α-crystaIIin in regeneration of optic nerve injury

·α-crystallin is the predominant structural protein in the lens.lt is a member of small heat shock proteins ( sHSPs) which has the common functions of HSPs.lt also has anti-apoptotic activity etc.Recently, it has been proved to combine with the cellular membrane of retinal ganglion cells ( RGCs ) to enhance the survival of RGCs and the regeneration of axons, thereby partly restore visual function.But we haven’t come to a unified conclusion of the mechanism.This review is focused on structure and functions of α-crystallin, the protection function and mechanism of α-crystallin towards RGCs after the optic nerve injury.
