1.Application of fluorescence probe marked by quantum dots to detect early submandibular lymph node metastasis in a nude mouse model.
West China Journal of Stomatology 2014;32(3):273-277
OBJECTIVEA fluorescence probe marked with a specific angle protein antibody by using quantum dots QDs605-CK (AE1/AE3) was utilized to detect metastasis and micrometastasis rates of early submandibular lymph node metastatic carcinoma in a nude mouse model. The method was compared with traditional immunohistochemistry (IHC) staining and hematine-eosin (HE) staining to explore a new method for early diagnosis and treatment of tongue squamous cell carcinoma.
METHODSHuman tongue Tca8113 cells were cultured and inoculated in 18 nude mice tongue body tissues (except the midline) to establish submandibular lymph node metastasis in tumor-bearing nude mice models. The nude mice were sacrificed, and the mandibular lymph node was dissected after injecting tumor cells for six weeks. One lymph node was divided into two parts: one was used to create a serial section embedded by paraffin then detected in HE staining and IHC staining; the other was immediately frozen in liquid nitrogen and detected using the fluorescence probe marked with a specific angle protein antibody by using quantum dots. Lymph node metastasis and micrometastases rates were calculated.
RESULTSThe rates of metastasis in the submandibular lymph node via quantum dot immunofluorescence staining, IHC staining, and HE staining showed positive values of 66.7%, 61.1%, and 27.8%, respectively. The micrometastases rates were 38.9% and 33.3% via quantum dot immunofluorescence staining and IHC staining, respectively. The positive rates of detecting lymph nodes metastasis with quantum dot labeled keratin CK (AEl/AE3) immunofluorescence and keratin CK (AE1/AE3) IHC staining were higher than that of conventional HE staining ('=6.379, P<0.05). The two methods showed no statistical difference (chi2 = 0.120, P > 0.05).
CONCLUSIONQuantum dots fluorescent probe marked with the angle protein antibody CK (AE1/ AE3) can precisely locate the submandibular lymph node metastasis tumor cell of the nude mice tongue squamous cell carcinoma, and the emitted red fluorescence showed strong specificity, high resolution, and a clear background. The method could be utilized to detect submandibular lymph node metastasis and micrometastasis of tongue squamous cell carcinoma in nude mice.

Objective To translate the English version of Comprehensive Frailty Assessment Instrument (CFAI) into Chinese and evalu-ate its reliability and validity. Methods CFAI was translated into Chinese with the Brislin's translation mode. From May to July, 2015, 200 community aged people in Shijiazhuang, China were assessed with the Chinese version of CFAI and World Health Organization Quality of Life-BREF (WHOQOL-BREF), and 30 of them were assessed with CFAI again after a week. The content validity was rated by six experts of gereology. The total scores of CFAI and WHOQOL-BREF were analyzed with Pearson's correlation, as well as the scores of CFAI test and retest. The CFAI were analyzed with Factor Analysis. The Cronbach's α was tested. Results There were 23 items in 4 dimensions. The content validity index was 0.782. Six factors were extracted by Factor Analysis and the cumulated variance was 64.05%. The total score of CFAI correlated with the score of WHOQOL-BREF (r=-0.764, P<0.001). The Cronbach's α was 0.704~0.897 in dimensions (P<0.001), and the r= 0.604~0.941 (P<0.001) between test and retest. Conclusion The Chinese version of CFAI is reliable and valid, that can be applied to evaluate frailty in community aged people.

Objective To elucidate the relationship between Child-Pugh grading and the degree of hep- atic cirrhosis.Method Six patients each with hepatic cirrhosis in Child-Pugh grade A and B were selected. The livers were observed with gross examinations during operation, and the liver specimens were observed with gross ex- aminations and also checked with light and electron microscopies. Results With naked eyes,it was quite difficult to find differences in size, shape and between different Child-Pugh grades. With hght microscopy, hepatic fibrosis of Ⅰ-Ⅱ degrees was commomly seen in Child-Pugh grade A and necrosis of hepatic cells was slight, while fibrosis of Ⅲ-Ⅴ degrees was usually found in grade B and necrosis was severe. With electron microscopy the collagen fib- rils in Disse's spaces were white cloud-like in grade A,while they were band-like in grade B.Of swelling and de- generation of organellae of hepatic and Kupffer cells, they were worse in grade B than in grade A.Conclusion The parameters in Child-Pugh grading are not complex, but they can be used to evaluate the degree of lesions. There is a positive relationship between them.

Objective To investigate the clinical role of a combination therapy of percutaneous radiofrequency ablation(RFA) and transcatheter arterial chemoembolization(TACE) in large hepatic tumors. Methods Out of 62 patients with hepatocellular carcinomas confirmed by pathology, 21 patients received a combination therapy of TACE and RFA(combination group), 22 patients TACE therapy alone, and the rest 19 patients RFA therapy alone. A total of 106 tumors with a mean diameter of ( 5.9? 0.7) cm(ranged from 5.0 to 8.1 cm) were detected, and the largest tumor was selected for observation in a patient with multiple lesions. There was no significant difference in mean age, tumor size and liver function grade among the three groups. Results Tumor complete necrosis accounted for 80.9%in combination group, which was significantly higher than that of TACE group and RFA group ( 27.2%, 47.4%,P 0.05). Mean survival duration of combination group was 25.6 months, significantly higher than that of TACE group( 14.9 months)(P0.05). Conclusions Compared with TACE or RFA therapy alone, the combination therapy improves tumor complete necrosis rate and prolongs the patients′ survival duration.
