Mitofusin 2 (Mfn2) plays an important role in the process of mitochondrial fusion, and is involved in regulating mitochondrial function and morphological changes. Studies have found that Mfn2 has a tumor suppressor effect in a variety of malignant tumors and their cell lines, including cervical cancer, hepatocellular cancer, pancreatic cancer, breast cancer, and bladder cancer. The difference in the expression of Mfn2 in malignant tumor tissues suggests that Mfn2 may play an important role in the proliferation, apoptosis, invasion and migration of malignant tumor cells. As a cancer-related gene, Mfn2 provides new ideas for gene therapy of malignant tumors.

Objective To study the effect and mechanism of Plantago depressa Willd. extract on rats with chronic nonbacterial prostatitis. Methods Sixty rats were divided into six groups randomly: normal control group, model control group, cernilton group, high-, medium- and low- dose Plantago depressa Willd. extract group, ten rats in each group. Chronic nonbacterial prostatitis rat model was established in the animals except of normal control group. Rats in the cernilton group were administrated with cernilton solution( 100 mg . kg-1 ) . High, middle and low dose Plantago depressa Willd. extract groups were given 150, 300 and 600 mg . kg-1 of Plantago depressa Willd. extract, respectively. The rats in the normal control and model control group were treated with the same volume of purified water.The administration was performed once every day, and lasted for three weeks.Effects of Plantago depressa Willd.extract on prostate index (PI), prostate specific antigen (PSA), tumor necrosis factorα ( TNF-α) , interleukin-1β ( IL-1β) , cycloxygenase 2 ( COX-2) , prostaglandin E2( PEG2 ) , transforming growth factor-β1( TGF-β1 ) , and connective tissue growth factor ( CTGF ) levels and pathological changes of prostate tissue in rats were observed. Results PI and PSA levels of the normal control group were(0.8±0.3)mg.g-1 and(106.5±10.6)pg.mL-1, respectively;those of the model control group were(2.2±0.2)mg.g-1 and(319.4±23.4)pg.mL-1 respectively;Those of the cernilton group were(1.6±0.3)mg.g-1 and(179.5±13.7)pg.mL-1 respectively;Those of the low-, medium- and high- dose Plantago depressa Willd.extract groups were(1.8±0.4),(1.3±0.3),(0.8±0.3)mg.g-1 and(263.4±28.6),(178.5±21.5), (143.5±12.9)pg.mL-1, respectively.Compared with the model control group, PI and PSA were significantly decreased(P<0.01) , TNF-α, IL-1β, COX-2, PEG2 , TGF-β1 and CTGF levels in the prostate tissues were decreased ( P<0. 01 ) , and inflammatory cell infiltration and fibrosis of prostate tissue was significantly alleviated in the model control group. Conclusion This study confirms Plantago depressa Willd.extract exerts effective therapeutical effect on chronic nonbacterial prostatitis in rats.

Objective To analyse effect of nifedipine sustained tablet on hemorheology in the treatment of patients with hypertension complicated with coronary disease.Methods 70 cases with hypertension complicated with coronary disease February 2014 to February 2015, were randomly divided into observation group and control group.35 cases in each group.Observation group received nifedipine treatment, control group were treated with conventional therapy with aspirin, recorded and analyzed related indexes and clinical efficacy of two groups patients after treatment.Results After treatment, patients plasma viscosity and hematocrit values were (1.59 ±0.12)mPa? s, (0.42 ±0.02)%, higher than control group (1.69 ±0.13) mPa ? s, (0.46 ±0.03)%(P<0.05); after treatment whole blood viscosity in observation group (high shear) and whole blood viscosity (low shear) values were (4.95 ±0.41),(9.87 ±1.01) mPa? s, lower than control group (5.89 ±0.43),(10.76 ±0.89) mPa? s(P<0.05); After observation group total effective rate was 91.43%, higher than 71.43%( P<0.05 ) .Conclusion Effect of nifedipine sustained-release tablets in patients with hypertension complicated with coronary disease is sure, precise, and could be compared to conventional treatment alone.It has no significant adverse reactions, can effectively improve hemorheology in patients with hypertension, hypertension complicated with coronary disease.

Protein fulfilling the their roles, one of important ways is through protein-protein interaction. In functional genomic era, identifying all of protein-protein interaction in proteome and mapping the protein interactions that have been attracting many scientists' attention , of which large-scale yeast two-hybrid system is one strategy of most widely used. In recent two years, ambitious projects have launched to examine all of the protein-protein interaction in Saccharomyces cer-evisiae using large-scale yeast two-hybrid system. Nevertheless, huge protein network is larger than that we predict and single yeast two-hybrid system cannot solve all the problems, which need be complemented by other wags.

Myelodysplastic syndrome (MDS) is characterized by dysplastic and ineffective hematopoiesis that can result from aberrant expansion and activation of myeloid-derived suppressor cells (MDSC) within the bone marrow microenvironment. The proliferation and activation of MDSC lead to the dysfunction and depletion of natural killer cells and CD8 + T cells, and the recruitment of inflammatory cells and factors leads to the further accumulation of genetic abnormalities in MDS patients, leading to the progression of MDS. The accumulation of inflammatory cytokines in the tumor environment induces the expression of programmed death receptor 1 (PD-1) in hematopoietic stem cells and hematopoietic progenitor cells and the overexpression of programmed death receptor ligand 1 (PD-L1) in MDSC, and the interaction of PD-1/PD-L1 leads to the apoptosis of MDS hematopoietic progenitor cells and ineffective hematopoiesis. The experiments and clinical studies targeting MDSC have confirmed that correcting or reversing the bone marrow microenvironment of immune disorders in MDS is a therapeutic strategy to restore effective hematopoietic function.

Objective:Guanxinshutong on cerebral ischemia reperfusion ( I/R) in rats learning and memory ability and brain Bcl-2,Bax protein expression to explore the protection mechanisms of Guanxinshutong on cerebral I /R injury in brain tissue.Methods:Healthy Wistar rats for the study were randomly divided into sham operation group ,I/R model group,high dose Guanxinshutong (1 g/kg) and low dose Guanxinshutong (0.5 g/kg) group,n=15.cerebral I/R model was estabhished by using the left commom cartid artery ligation method,modeling each group were fed daily before saline ,saline,Guanxinshutong (1 g/kg) and Guanxinshutong (0.5 g/kg), continuous 7 days;3 d after water maze training while continuing administration ,continuous 7 d,the eight day ,Morris water maze test;after the test,the rats were sacrificed blood and brain tissue ,serum by ELISA of Bcl-2 and Bax protein levels change;using HE staining of rat brain pathology;using immunohistochemistry assay in rat brain organization of Bcl-2 and Bax protein levels changes.Results:Compared with the sham group ,Guanxinshutong high-dose group test latency slightly longer timers after platform and platform quadrant dwell decreased slightly ,serum and brain tissue slightly elevated levels of Bax protein ,Bcl-2 protein levels decreased slightly but not significantly different ( P>0.05 ) ,low-dose group Guanxinshutong through latency test in rats ,after the number of platforms and platform quadrant dwell time decreased , serum and brain tissue elevated levels of Bax protein , Bcl-2 protein content decreased significantly different(P<0.01),cerebral I/R model group was significantly prolonged latency test ,after the number of platforms and platform quadrant dwell time significantly reduced serum and brain tissue was significantly increased Bax protein ,Bcl-2 protein content decreased with significant differences ( P<0.01 ).Conclusion: Guanxinshutong can significantly improve spatial memory , and by reducing Bax protein content and increased Bcl-2 protein,inhibition of cerebral I/R injury of apoptosis,and thus the brain tissue I/R has a protective effect.

Objective To assess the efficacy of laryngeal mask airway Supreme (LMA Supreme) used in patients undergoing gynecological laparoscopic surgery. Methods Eighty ASA Ⅰ or Ⅱ patients, aged 40-64 yr,weighing 50-70 kg, undergoing elective gynecological laparoscopic surgery, were randomized into 2 groups ( n = 40 each): LMA Supreme group (group S) and LMA ProSeal group (group P). Mallampati test was performed before operation in both groups. The patients were classified as Ⅰ or Ⅱ . LMA Supreme and LMA ProSeal were inserted in group S and P respectively after induction of anesthesia with midazolam 0.05 mg/kg, propofol 2 mg/kg, fentanyl 3 μg/kg and vecuronium 0.1 mg/kg. A gastric tube was inserted through the drain tube of the LMA. The number of attempt, success rate, LMA placement time, success rate of gastric tube placement, cuff volume, peak airway pressure, and airway seal pressure, BP, HR, ECG, SpO2, PET CO2 and complications were recorded. The duration of surgery and pneumoperitoneum, and emergence time were also recorded. The fiberoptic bronchoscope scores were assessed after successful LMA placement. Results There were no significant differences in the duration of operation and pneumoperitoneum, emergence time between the two groups (P ＞ 0.05). The BP, HR,SpO2, PET CO2 and peak pressure were within the normal range during operation in both groups. The first attempt success rates of LMA placement and gastric tube placement were significantly higher, the LMA placement time was significantly shorter, and the fiberoptic bronchoscopic scores were significantly higher in group S than in group P ( P ＜ 0.05). Conclusion LMA Supreme is helpful for ventilation during operation with less complications and can be used effectively for gynecological laparoscopic surgery.

Objective To investigate the sensitivity of the real-time fluorescence PCR technique and the bacterial culture for detecting the colonization of group B Streptococcus(GBS)in late pregnant women.Methods 2 specimens were collected from preg-nant women genital tract-rectal secretions swabs,one specimen for conducting the bacterial culture and another for conducting the real-time PCR technique to detect genital GBS.The accuracy and rapidness were compared between the two methods.308 cases of pregnant women were divided into the GBS positive group and the GBS negative group according to the detection results of the real time real-time fluorescence PCR technique.The relation between the occurrence of premature rupture of membranes with GBS was investigated by the comparative analysis.Results Among 308 pregnant women with GBS detection,18 cases were positive by the ordinary bacterial culture with the positive rate of 5.8%(18/308),while 28 cases were positive by the real-time fluorescent PCR with the positive rate of 9.4%(29/308).In the GBS positive group detected by PCR,the premature rupture of membranes occurred in 9 cases with the positive rate of 31%,while in the GBS negative group detected by PCR,which occurred in 33 cases with the pos-itive rate of 11.83%.Conclusion This survey shows that the positive detection rate of the real-time fluorescent PCR technique is significantly higher than that of the bacterial culture method,the application of this detection technique for detecting GBS provides the basis for rapidly diagnosing GBS and conducting the prophylactic use of antibacterial drugs more accurately and more effectively.

Objective:To analyze two different kinds of mouse model with immune thrombocytopenic purpura for the immune mechanisms.Methods:Immune thrombocytopenic purpura was induced by either injection of polyclonal antibody or immunized inoculation with rat platelets.Results:The two kinds of model all induced decrease of platelets,while immunization with platelets of rat induced alloantibodies in the mice.Conclusion:Immunization with platelets of rat can mimic the production process of autoantibodies in human and lead to immune thrombocytopenic purpura in mice.
