1.Clinical value of serum BNP,TNF-α,MMP-9 and IL-6 detection in CHF patients with different heart function grades

Objective To discuss the clinical value of serum brain natriuretic peptide (BNP) ,tumor necrosis factor‐α(TNF‐α) , matrix metalloproteinase‐9(MMP‐9) and interleukin‐6(IL‐6) detection in chronic heart failure (CHF) patients with different heart function grades and their correlation with CHF .Methods Totally 67 cases of CHF in our hospital from Jan .2013 to Jun .2015 were selected as the observation group and the NYHA classification was performed :gradeⅠ in 15 cases ,gradeⅡ in 21 cases ,gradeⅢ in 23 cases and grade Ⅳ in 9 cases .Other 30 individuals undergoing the healthy physical examination were chosen as the control group at the same period .The venous blood samples were collected for detecting serum BNP ,TNF‐α,MMP‐9 and IL‐6 .LVEDD ,LVESD and LVEF were measured by echocardiography .Then the obtained data were analyzed by using the SPSS 21 .0 software .Results The serum BNP ,TNF‐α,MMP‐9 and IL‐6 levels in the observation group and various cardiac function groups were higher ,the differences between them were statistically significant (P< 0 .05);LVEF in the observation group and various cardiac function groups were obviously declined ,while LVEDD and LVESD were obviously increased ,the differences were statistically significant (P<0 .05) .The linear regression was adopted to analyze the correlation between serum BNP ,TNF‐α,MMP‐9 and IL‐6 with the left ventricular echocardiographic parameters in the CHF patients ,the results showed that serum BNP ,TNF‐α,MMP‐9 and IL‐6 levels were negatively correlated with LVEF (P<0 .05) ,while positively correlated with LVEDD and LVESD (P<0 .05) .Conclusion The serum BNP ,TNF‐α,MMP‐9 and IL‐6 levels are the good indexes for diagnosing CHF ,moreover can accurately reflect different heart function status in the CHF patients with different cardiac function grades ,which are closely related to the occurrence and de‐velopment process of CHF .
2.Expression of MT-1 and MT-2 Genes in Liver of Chronic Exposure to Inorganic Arsenate Mice

Objective To investigate the effect of MT-1 and MT-2 in liver damage in chronic inorganic arsenate exposure mice and to explore the mechanism of arsenic-induced liver damage. Methods The male Kunming mice were randomly divided into control and exposed groups. The control group was given ordinary feed and tap water. The exposed group was given ordinary feed and 300 mg/L of sodium arsenite solution by drinking water. After 10 months of treatment, the activity of serum alanine aminotransferase (ALT), aspartate aminotransferase(AST) and globulin (Glb) content were determined. The total RNA was extracted by the TRIzol-phenol-chlorofor-method from the liver tissue. The quantity of the RNA was determined by spectrophotometry and its purity was judged at a ratio of A260/A280. Then real-time PCR(RT-PCR) was used to measure the mRNA expression of MT-1 and MT-2. 3.Expression and distribution of COX-2 in human gastric cancer cells

Objective To detect the expression of cyclooxygenase(COX) in human gastric cancer cell lines and its subcellular location of the isoforms. Methods Immunohistochemistry、RTPCR combined with laser scanning cofocal microscopy (LSCM) were used to investigate expression and distribution of COX. Results Positive staining of COX2 and COX1 protein was seen in human gastric cancer cell line MKN45、SGC7901 and AGS. However, the COX2 staining was absent and COX1 staining was weak in MGC803; though their mRNA in all the four cell lines. When compared to COX1,COX2 showed a stronger signal at both protein and mRNA levels of the gastric cancer cell lines, which was confirmed by double labeling and LSCM. The quantitative analysis of fluorescein intensity indicated the pixel intensity peak of COX2 reached 50~70, while COX1 only 10. LSCM also showed that COX2 was both cytoplasmic and nuclear envelope staining but COX1 only in cytoplasm. Conclusions In human gastric cancer, there is stronger expression of COX2 than COX1, and different distribution of the two isoforms implies their distinct roles in cell function.
4.Mechanism of Antiasthma by Inhaled Arsenic Trioxide

Objective To investigate the effect of eosinophils(EOS)in asthmatic guinea pigs airway and explore the mechanism of antiasthma by inhaled arsenic trioxide. Methods The guinea pigs asthma models were established with ovalbumin(OVA)challenge method,2 groups received inhaled different doses of arsenic trioxide,and three control groups respectively received normal saline inhaled and high dose of arsenic trioxide by intraperitoneal injection and dexamethasone(DEX)intraperitoneally. EOS invaded and apoptosis with all of the groups were assessed after 7 days intraperitoneal injection or inhalation. Results Compared with group of inhaled normal saline, both EOS invaded and apoptosis in bronchus submuconsa were significantly different(P0.05)among groups of inhaled low dose arsenic trioxide [2.0 mg(kg?d)]and intraperitoneal injection with higher dose arsenic trioxide[5.0 kg/(kg?d)]and DEX[10 mg/(kg?d)]for these two parameters. Conclusions The mechanism of arsenic trioxide antiasthma with arsenic trioxide can decrease EOS amount in bronchial submucosa and accelerate EOS apoptosis, and relieve bronchial inflammation in asthma. For inhaled lower dose arsenic trioxide or intraperitoneal injection with high dose arsenic trioxide ,the effect of EOS is equivalent. The antiasthma effect of inhaled lower dose arsenic trioxide[2.0 mg/(kg?d)]is equivalent to intraperitoneal injection higher dose[5.0 mg/(kg?d)],and may be safe comparatively. The mechanism of antiasthma by inhaled arsenic trioxide is the same with DEX.
5.Effects of Liraglutide on plasma fibroblast growth factor-21 level in patients with type 2 diabetes mellitus

Objective To investigate the effects of Liraglutide on plasma FGF-21 level in patients with type 2 diabetes mellitus (T2DM). Methods Enzyme linked immunosorbent assay (ELISA) was used to measure plasma FGF-21 level in patients with T2DM and normal controls (NGT). Relationship between plasma FGF-21level and BMI, WHR, HOMA-IR, HOMA-IS, FFA was analyzed. After Liraglutide treatment, plasma FGF-21level was observed in patients with T2DM. Results Fasting plasma FGF-21 level was higher in T2DM patients than that in normal control group ( 1.81 ±0.32 vs. 1.32 ±0.09 μg/L, P <0.01 ). After Liraglutide treatment, WHR, BMI, FPG, 2hPBG, HbA1c and FFA in T2DM patients significantly decreased ( P <0.05 or P <0. 01 ),HOMA-IS remarkably increased (P < 0. 01 ), and FGF-21 had no change. Conclusion Liraglutide and FGF-21may have different roles in insulin resistance.
6.Postmenopausal bleeding:a clinical analysis of 210 cases

Objective To investigate the causes,diagnostic methods,prevention and cure of postmenopausal bleeding(PMB).Methods The clinical and pathological data of 210 PMB patients admitted in this hospital were retrospectively analyzed.Results The causes for PMB were benign diseases(69.5%),the most of which were infec- tious diseases(49.5%),non-organic disease(20.5%)and malignant tumor(10.0%).Compared with benign diseases and non-organic diseases,the attack age of malignant tumor was higher[(67.1?6.9)years old],menopausal dura- tion was longer(11.6?2.9)years,bleeding duration was longer[(157.6?26.4)days],and mucous membrane was obviously thickened[(14.6?3.2)mm](P0.05).Conclusion The main cause for PMB is benign diseases,followed by non-organic diseases.More attentions should be paid to the PMB patients with higher attack age,longer menopausal duration and bleeding dura- tion,and obviously thickened mucous membrane,so as to be precautions to the occurrence of malignant tumor.
7.Down-regulating HIPK2 promotes cisplatin-induced apoptosis of human kidney tubular epithelial cells

Objective To explore the effect of HIPK2 on apoptosis of human kidney tubular epithelial cells (HKC) induced by cisplatin.Methods Apoptosis of HKC cells was induced by cisplatin and the expression of HIPK2 was detected by RT-qPCR and Western blot.Two HIPK2 siRNAs were designed according to gene sequence of HIPK2 and cell lines with HIPK2 knockdown were established through transfecting the HIPK2 siRNAs into HKC cells by liposome.The expression of HIPK2 mRNA and protein was detected by RT-qPCR and Western blot after induced by cisplatin.Then cell apoptosis was detected by Annexin V/PI after the HIPK2-knockdown cells were treated with cisplatin.Moreover,the expression of pro-apoptotic protein bax was detected by Western blot after HIPK2 was knockdown.Results The expression of HIPK2 mRNA and protein was down-regulated obviously on the process of HKC apoptosis which induced by dose-dependent cisplatin (P<0.05).The transfection of siRNA could significantly reduce the expression of HIPK2 mRNA and protein in HKC (P<0.05),which promotes the HKC cells apoptosis induced by cisplatin.Conclusions HIPK2 can suppress the HKC cells apoptosis induced by cisplatin.

Results:Infusion apparatus can be placed in any position, the peripheral infusion tube length range without the suspension liquid device and changing the existing infusion (blood) products and procedures, which create convenient, comfortable infusion conditions without position limitation The controllable range of the infusion speed is 50ml-1000ml/h,which can monitor the infusion and the air in the pipeline, liquid flow rate, the remaining amount, According to the set alarm timely and automatically stop the transfusion. Conclusion:To improve the safety of transfusion, save human resources. and it’s dual-use, small volume, easy to carry.
9.Warming nursing to severe traumatic patients with hypothermia and nursing measures

Objective To investigate the causes of hypothermia in severe trauma patients as well as the effects of warming nursing.Methods Toally 100 severe trauma patients with hypothermia were engaged in the study during January to December 2014.Their temperature was monitored and recorded,the causes analyzed so that the warming nursing measures were done to them.Results Among the 100 patients,59 contracted hypothermia,with the rate of 59.0% before operation,28 contracted hypothermia during operation,with the incidence rate of 28.0%.The causes of hypothermia included injury,anaesthesia,exposure and fluid resuscitation.The nursing measures included pre-treatment before anaesthesia,avoidance of more exposure and intraoperative warming.Conclusions For the patients with severe trauma,the hypothermia during the operation can be caused by injury,anaesthesia,exposure and fluid resuscitation.The warming nursing can reduce the incidence of hypothermia so as to increase their survival rate.
10.Study on Transformation of Complete Atrioventricular Block in Children

Objective To observe the transformation features of children with complete atrioventricular block(CAVB), and to improve the diagnosis and treatment of this disease.Methods Seventeen children with primary diagnosis of CAVB were reviewed by retrospective and follow-up study and the clinical characteristics, treatment schemes and prognosis were evaluated.Results Four cases of congenital CAVB lived normally without obvious symptoms in the tracing period from 6 months to 9 years;1 case had the onset of Adams-Stokes syndrome induced by diarrhea;3 cases of the CAVB caused by virus myocarditis turned into sinus rhythm after comprehensive therapy;2 cases persistently presented with the CAVB Complicated with enlarged heart;1 case gave up treatment after deterioration, and one case died.Three cases of temporary CAVB after the open heart operation turned to sinus rhythm in transitory time, while other two cases presented with permanent CAVB and treated with epicardium pacemaker,among whom one had the pacemaker replaced for one time in the 8-year follow-up,and the follow-up of other cases were intermitted.Conclusions The congenital CAVB in the study group with normal QRS interphase and no obvious symptom might not require treatment but follow-up is needed. While infants with heart malformation and wide QRS wave could not endure the low ventricular rhythm are in high risk. Virus myocarditis induced CAVB children tend to present the Adams-Stokes syndrome, and require effective treatments. Partial cases of the CAVB caused by open heart operation may turn to normal sinus rhythm in 2-4 weeks after surgery. cases has persistent CAVB for over 4 weeks, or Adams-Stokes syndrome onset after the surgery demand epicardium pacemaker treatment.
