1.Effect of Electroacupuncture on Motilin and Somatostatin inRats with Diabetes Gastroparesis

Objective To investigate the effect of electroacupuncture on motilin (MOT) and somatostatin (SS) in rats with diabetes gastroparesis (DGP). Methods One hundred clean healthy SD rats with negative urine glucose were chosen. After they were fed for three days and into a stable state, 30 rats were randomly selected as a control group. The other 70 rats were used as an animal model of insulin-dependent diabetes made with streptozotocin. At 10 weeks after model making, rats as successful models were randomly allocated to model and treatment groups, 30 rats each. The model group received no treatment and the treatment group, electroacupuncture at points Zusanli, Zhongwan and Weishu. Serum and antral MOT and SS were measured in every group before and after treatment. Results There were statistically significant differences in body weight and gastrointestinal propulsion rate between the treatment or model group of rats and the control group (P<0.01), and between the treatment and model groups after treatment (P<0.01). There was a statistically significant difference in blood glucose value between the treatment or model group of rats and the control group after model making and at 10 weeks and the end of treatment (P<0.01). There was a statistically significant difference in blood glucose value between at the end and 10 weeks of treatment in the treatment group (P<0.01). There were statistically significant differences in serum and antral MOT and SS contents between the treatment or model group and the control group (P<0.01), and between the treatment and model groups after treatment (P<0.01). Conclusions Electrocupuncture can markedly increase serum SS and antral MOT and decrease serum MOT and antral SS to ameliorate high blood motilin and somatostatin in DGP rats.
2.Development and application of the method for the detection of human IL-6 mRNA

Objective:To develop and apply a method for real-time quantifying IL-6 mRNA.Methods:By Ficoll-Hypaque density gradient centrifugation, human peripheral blood mononuclear cells(PBMC) were isolated from whole blood treated with ethylenediaminetetraacetic acid(EDTA). Total RNA extracted from the PBMC in trizol solution was reverse transcribed to cDNA, which used as templates for fluorecent real-time quantitative PCR amplification of IL-6. PCR system consists of IL-6 primer, SBY Green Ⅰ and so on.Results:The method is wide linear range, sensitive, specific, reproducible and simple.Conclusion:The method can accurately quantify IL-6 mRNA.

4.The characteristics of the gastroesophageal junction in gastroesophageal reflux disease under high-resolution esophageal manometry

Objective To analyze the characteristics of gastroesophageal junction(GEJ) in gastroesophageal reflux disease (GERD) using high resolution manometry (HRM),and the features in different GERD subtypes,reflux esophagitis (RE) and non-erosive gastrooesophageal reflux disease (NERD).Methods Sixty GERD patients were divided into two subtypes,RE (n =33) and NERD(n =27),based on the results of endoscopy.All patients received HRM,and were divided into three subtypes,type Ⅰ,type Ⅱ and type Ⅲ according to the GEJ pressure morphology.HRM results among different subtypes were investigated.Results In 60 GERD patients,the GEJ was divided into three subtypes,including 26 cases of type Ⅰ (RE =14,NERD =12),29 cases of type Ⅱ (RE =14,NERD =15) and 5 cases of type Ⅲ (RE =5).The characteristics of GEJ morphologic subtypes were analyzed.The total length of lower esophageal sphincter(LES) of three GEJ subtypes(type Ⅰ,type Ⅱ and type Ⅲ) were (3.35 ± 0.56) cm,(3.29 ± 0.87) cm and(2.43 ± 0.22)cm,respectively.The LES intra-abdominal length of three GEJ subtypes were(2.41 ± 0.68) cm,(3.07 ± 4.59) cm and (0.00 ± 0.00) cm,respectively.The LES rest pressure of three GEJ subtypes were (22.00 ± 8.00) mmHg,(16.46 ± 8.60) mmHg and (11.07 ± 5.80) mmHg,respectively.The total length of LES were significantly different among three subtypes (P < 0.05).The LES rest pressure were not significantly different among three subtypes (P > 0.05).Among type Ⅰ and type Ⅱ patients,there were no significant difference in LES-CD distance,total length of LES,LES intra-abdominal length and LES rest pressure between RE group and NERD group.Conclusion HRM can clearly distinguish the GEJ subtypes.To some extent,esophageal motility characteristics of the patients with different subtypes GEJ is various.However,it still needs further studies with larger sample size to confirm.
5.Expressions and significances of RUNX3 and CyclinD1 in pancreatic carcinoma

Objective To investigate the expressions of RUNX3 and CyclinDl in pancreatic carcinoma and their significance. Methods Expressions of RUNX3, CyclinD1 in 47 cases with pancreatic carcinoma, 18 cases with cystadenoma of pancreas and 12 normal pancreas cases were detected by immunohistochemistry, and the relationship between their expressions and clinicopathological parameters was analyzed. Results The positive expression rates of RUNX3 in pancreatic carcinoma, cystadenoma of pancreas, normal pancreas cases were 57.4% (27/47), 94.4% (17/18), 100% (12/12); the positive expression rates of CyclinD1 in pancreatic carcinoma, cystadenoma of pancreas, normal pancreas cases were 72.3% (34/47), 44.4%(8/18), 8.3% (1/12). RUNX3 expression was not related to the age and sex, but it was negatively associated with clinical staging, lymph node metastasis, the differentiation degree (P <0.05 ). CyclinD1 expression was not related to the age and sex, but it was positively associated with clinical staging, lymph node metastasis, the differentiation degree (P <0.05 ). The expression of RUNX3 and CyclinD1 was negatively associated (r = - 0.375, P = 0.009). Conclusions The expression of RUNX3 is decreased in pancreatic carcinoma. The expression of CyclinD1 is increased in pancreatic carcinoma. They may play an important role in the carcinogenesis and progression of pancreatic carcinoma.
6.Clinical Study of the treatment of cervical spondylosts with percutaneous laser disc decompression combined with ozone injecaon

Objective To study the clinical effect of percutaneous laser disc decompression(PLDD) combined with ozone injection in the treatment of cervical spondylosis.Methods 183 cases of cervical spondylosis were randomly divided into two groups.One group of 80 cases with 118 cervical discs herniation were treated with PLDDonly.while the other group of 103 cases with 142 cervical discs herniation were treated with PLDD combined with ozone injectlon.The patients were followed up for 3 to18 months(mean 7 months).The clinical effects were assessed according to the Japanese Orthopaedic Association scoring system(JOA score)and the improvement rates of the two groups were compared.Results 167 cases were followed up.The average point of the group treated with PLDD is 15.2 and the improvement rate is 74.7%.The average point of the group treated with PLDD combined with ozone injection is 16.3 and the improvement rate is 91.3%.The difference between two groups has statistical significance(P<0.05).Conclusion The treatment of cervical spondylosis with PLDD combined with ozone injection is safe.painless and minimally invasive.The clinical effect is impmved tremendously and much better than monotherapy.
7.Correlating estrogen receptor-alpha Pvull genetic polymorphism with bone mass index in premenopausal Han Chinese women from Nanchang region

BACKGROUND: Obesity is serious complex disease determined by multiple genetic factors. Body mass index (BMI) is a common phenotype for obesity. Although some candidate genes have been shown to be related with BMI variation, the underlying substantial relationship remains poorly understood.OBJECTIVE: To investigate whether estrogen receptor-α (ER-α) Pvull genetic polymorphism is associated with BMI variation.METHODS: In total 390 premenopausal women from a local population of Nanchang City in Jiangxi Province People's Hospital.Body weight and height were measured. All participants were genotyped at the ER-α Pvull locus using polymerase chain reaction-restriction fragment length polymorphism.RESULTS AND CONCLUSION: The ER-α Pvull genotype frequencies of PP, Pp and pp were 0.133, 0.485 and 0.382,respectively. The distribution of ER-Pvull genotypes was in accordance with Hardy-Weinberg equilibrium (HWE) (P > 0.05).There was no significant correlation between ER-α Pvull genetic polymorphism and BMI variation (P = 0.338). These findings indicate that ER-α Pvull genetic polymorphism does not greatly affect BMI variation of premenopausal women in Nanchang region.

Objective To understand the fluorescence pattern of antinuclear antibody(ANA)and the distribution of target anti-gen and ANA′s impact on rheumatoid arthritis(RA)RA.Methods The data of rheumatoid factor(RF)and anti-cyclic citrullinated peptide antibodies(anti-CCP antibodies)and ANA of 860 RA patients during 2014-2015 were collected and divided into 2 groups according to the ANA levels.The differences of RF and anti-CCP antibodies level between 2 groups were analyzed.Analyzed the fluorescence pattern of RA merger ANA and the distribution of target antigen by statistical.Results Of the 860 RA patients,the positive rate of ANA was 37.6%,the major fluorescence pattern of RA merger ANA was speckled pattern,anti SSA positive rate was 21.7%.The RF and anti-CCP antibodies level had statistical difference between ANA positive group and ANA negative group (P <0.05).Conclusion RA merger ANA positive can show different fluorescence patterns and target antigen,the speckled pattern and anti-SSA positive are the main pattern.The severity of arthritis symptoms and the degree of bone destruction between ANA positive group and ANA negative group were difference.Anti-ANA positive has a certain promoting effect on the progression of RA disease.
9.Three-dimensional conformal hypofractionated radiotherapy for non-small-cell lung cancer

Background and purpose:The local tumor control rate of non-small-cell lung cancer treated with conventionally fractionated radiotherapy is low.Hypofractionated radiotherapy performed by conformal irradiation techniques can improne the local control rate.But further studies for appropriate fraction dose and toxicity for hypofractionation should be done.The purpose of this study was to prospectively evaluate the safety and efficacy of three-dimensional conformal hypofractionated radiotherapy for non-small-cell lung cancer(NSCLC).Methods:According to the dose-volume histogram(DVH) V_(20), patients were divided into three groups:① V_(20)≤20%,②20%30%,grade Ⅲ RP was observes in 2 of 5 patients and grade Ⅳ RP in 1 patient who died of lung function failure.No grade≥Ⅲ radiation esophagitis was observed.25 patients were evaluated with 8 complete responses,13 partial responses,3 stable diseases and 1 progressive disease.Conclusions:For three-dimensional conformal hypofractionated radiotherapy V_(20) level should be controlled below 30%,the treatment plan with V_(20)≥30% should be changed to palliative treatment.More studies are needed to confirm its efficacy.
10.Influence of pathologic factors on phenotypic transformation of renal tubular epithelial cells in vitro

Objective To study the effects of high albumin, high glucose and low bovine serum on the phenotypic transformation of renal tubular epithelial cells. Methods The normal human kidney proximal tubular eel] line (HKC) was cultured for 30 days in the presence of high albumin (1.5 g/L), high glucose(25 mmol/L) and low bovine serum(2% ) . Morphological changes were observed by electronic microscopy. Immunohistochemistry stain was used to examine the expression of cytokeratin, vimentin, a-SMA, collagen Ⅰ and TGF-pl protein. Western blot was applied to further detect the process of collagen I protein expression, and in situ hybridization was used to examined the expression of collagen Ⅰ gene. Results Renal tubular epithelial cells cultured in high albumin, high glucose and low bovine serum showed obvious morphologic changes, including elongated shape, decrease of microvilli and mitochondria, and increase of rough endoplasmic reticulum under electronic microscopy. Immunohistochemistry stain revealed the reduction of cytokeratin, and enhancement of vimentin, ?-SMA, TGF-?1 and collagen Ⅰ. Western blot demonstrated that the expression of collagen Ⅰincreased in a time-dependent manner, and in situ hybridization showed that collagen type Ⅰ mRNA increased as well. Conclusion High albumin, high glucose and low bovine serum induce phenotypic transformation of renal tubular epithelial cells into mesanchymal cells.
