Objective To observe the morphology, cell viability and secretion function of catecholamine of the alginate-polylysine-alginate (APA) microencapsulated bovine chromaffin cells (BCCs) before and after cryopreserving with liquid nitrogen. Methods The APA microencapsulated BCCs were cryopreserved with dimethyl sulfoxide as cryopreservative agent by slow cooling and rapid rewarming for revivification. The change of cell function was observed by detecting the cell viability and secretion of catecholamine. Results As compared with the precryopreseving cells, the morphology and cell viability of the resuscitated APA microencapsulated BCCs showed no significant change. The catecholamine secretion volume of BCCs remained 80% of that by the precryopreserving cells. Conclusion It demonstrates that the resuscitated cryopreserved APA microencapsulated BCCs still remained good morphology, cell viability and secretion function of catecholamine.

To evaluate the efficacy and safety of limbal relaxing incision ( LRl) for correcting corneal astigmatism during implantable collamer lens ( lCL) surgery.METHODS:A total of 185 eyes of 105 patients with high myopia and corneal keratometric astigmatism were included in the study. lCL surgery with concomitant relaxing incision was performed in 105 eyes of 60 patients in LRls group ( Group A) . Eighty eyes of 45 patients only underwent lCL surgery were in control group ( Group B) . All patients undergone ophthalmic examination that included uncorrected visual acuity ( UCVA ) , best -corrected visual acuity ( BCVA ) , Pentacam analysis system to observe the changes of corneal astigmatism before and 1wk, 1 and 3mo after surgery.RESULTS: Respectively comparing UCVA between two groups in 1 and 3mo postoperatively, the P values were considered statistically significant ( P < 0. 05 ). But, respectively comparing BCVA between two groups in 1wk, 1 and 3mo postoperatively, the P values were considered no statistically significant ( P > 0. 05 ). Preoperative corneal astigmatism was 1. 52 ± 0. 55D in group A and 1. 48 ± 0. 57D in group B, there was no statistically significant difference (P>0. 05). One week postoperatively, the astigmatism was 0. 55 ± 0. 41D in group A and 1. 20 ± 0. 48D in group B. One month postoperatively, the astigmatism was 0. 60 ± 0. 38D in group A and 0. 93 ± 0. 47D. Three months postoperatively, the astigmatism was 0. 51 ± 0. 32D in group A and 0. 96 ± 0. 40D in group B. The difference between the two groups were statistically significant ( P<0. 05 ). The difference value of corned astigmatism before surgery and 1wk, 1 and 3mo after surgery had statistical significance ( P<0. 05). ln LRls group, at preoperative and postoperative time points, the average corneal astigmatism changes were also considered statistically significant difference (P<0. 05). CONCLUSlON: LRls performed during lCL surgery appeared to be an effective and safer procedure to reduce pre-existing corneal astigmatism and improve UCVA as well as the visual quality.

OBJECTIVE: To observe the effect of electro-acupuncture of Neiguan (PC 6) on mean systemic arterial blood pressure and plasmic concentrations of NO and TNFalpha in endotoxin shock rats. METHODS: The model of endotoxin shock was induced by lipopolysaccharide (1.5 mg/kg i.v.) and D-galactosamine (100 mg/kg i.p.). Aminoguanidine (100 mg/kg i.p.) and electro-acupuncture of bilateral Neiguan (PC 6) were administered. A catheter was inserted into the right subclavian artery to record the change of blood pressure and the blood was abstracted out and centrifuged to determine the NO and TNFalpha concentrations. RESULTS: Electro-acupuncture of Neiguan (PC 6) retrieved the blood pressure and reduced the plasmic NO and TNFalpha concentrations. CONCLUSION: Electro-acupuncture of Neiguan (PC 6) expresses an anti-endotoxin shock effect by repressing the plasmic NO and TNFalpha concentrations smoothly and retrieving the blood pressure stably.

Objective: To explore the effect of Plastrum Testudinis on the three subtypes of NOS (nNOS,iNOS,eNOS) expression in rats with focal cerebral ischemia. Methods:Rat model of focal cerebral ischemia was set up by inserting nylon thread into the internal carotid artery . The expressions of nNOS, iNOS and eNOS was detected by immunohistochemical method to observe the effects of Plastrum Testudinis. Results: Expressions of nNOS and iNOS in cerebral cortex and caudate-putamen of Plastrum Testudinis treatment group were significantly lower than those of model control group (P

[Objective] To observe the effect of serum containing Carapax et Plastrum Testudinis (CPT) in inducing the differentiation of mesenchymal stem cells (MSC) of adult rats into neurons. [Methods] The fifth generation of MSC were induced by the serum containing CPT (sero-CPT) and the expressions of neurofilament (NF) protein and glial fibrillary acidic protein (GFAP), the marks of neuron-like cells were detected by immunohistochemical method. [Results] The expression of NF protein was positive after the stimulation of sero-CPT and the expression reached the peak 12 hours after induction. [Conclusion] Sero-CPT can induce the differentiation of MSC into neurons in vitro.

Objective To investigate the effect of low-speed treadmill training on function of lower limbs in hemiplegic patients afterstroke. Methods 26 hemiplegic patients after stroke were divided into the treatment group (n=14) and control group (n=12). Both groups acceptedroutine rehabilitation, and the treatment group accepted low-speed treadmill training in addition. They were evaluated with Fugl-MeyerAssessment (FMA), Berg Balance Scale (BBS), 6-min walk distance, 10-m walking speed before and after treatment. Results The scoresof FMA and BBS, and the distance of 6-min walk, speed of 10-m walking improved in both groups (P<0.05), and improved more in thetreatment group than in the control group (P<0.05) after 3-mouth treatment. Conclusion Low-speed treadmill training can significantly improvewalk ability and balance function of the hemiplegic patients after stroke.

ObjectiveTo establish a novel rapid detection method based on PCR-single-strand-conformational polymorphism (PCR-SSCP) to determine mutation of streptomycin-resistance associated rpsL and rrs genes in isolates of Mycobacterium tuberculosis (MTB).MethodsStreptomycin-resistance of 112 MTB isolates was detected using the routine drug susceptibility test, and a special PCR-SSCP assay was established. The mutations of rpsL and rrs genes in streptomycin-resistant MTB isolates were detected by PCR-SSCP and PCR direct sequencing (PCR-DS) ; the results from two techniques were compared. Results All isolates had both rpsL and rrs genes.Fifty-two isolates (46.4%) were streptomycin susceptible,in which only 1 isolate showed abnormal PCR-SSCP fragments from rrs gene, and the specificity of PCR-SSCP was 98.1% (51/52).Sixty isolates (53.6%) were streptomycin-resistant,in which 46 (76.6%) and 11 ( 18.3% ) isolates presented the abnormal PCR-SSCP fragments of rpsL and rrs gene, respectively. One streptomycin-resistant isolate showed abnormal PCR-SSCP fragments from both rpsL and rrs genes. The sensitivity of PCR-SSCP was 93.3% (56/60).ConclusionThe PCR-SSCP that established in this study is a specific and sensitive method for rapid detection of the streptomycin-resistance associated mutations in rpsL and rrs genes of MTB.
