Journal of Shanghai Jiaotong University(Medical Science) 2006;0(04):-
Objective To clone and compare RNase HIIa and RNase HIIb of Chlamydia pneumoniae AR39(CpRNaseHIIa and CpRNaseHIIb). Methods Genes of CpRNase HIIs were amplified with the designed primers. Then, the recombinant plasmids were constructed, and CpRNase HIIs were expressed and purified with pET expression system. The 5′-32P-labeled RNA/DNA substrate and DNA with oligoribonucleotides substrates were prepared to identify characterization of CpRNase HIIs. Results Ribonuclease H activity of both CpRNase HIIa and CpRNase HIIb could cleave oligoribonucleotides strand, generating break with 3′-OH and 5′-phosphate ends. The results showed that there was different biochemical characterization between them. Conclusion CpRNase HIIa and CpRNase HIIb have different functions in C.pneumoniae.

Journal of Chinese Physician 2000;0(11):-
Objective To analyze the clinical characteristics of digestive system dysfunctions after cardiovascular surgery with cardiopulmonary bypass (CPB). Methods The clinical data of 2349 consecutive cases undergoing cardiovascular surgery with CPB from Jan 1996 to Dec 2002 were retrospectively reviewed. The risk factors, clinical features, diagnostic methods, treatment and prognosis of digestive system dysfuctions were summarized. Results The incidence of digestive system dysfunctions was 1.4% (n=33), and the most common events were paralytic ileus (33.3%, n=11) and gastrointestinal bleeding (27.3%, n=9). Liver function insufficiency and ischemic bowel disease were the most common causes of deaths. 26(78.8%) patients underwent medicine treatment, 7(21.2%) patients accepted surgical interventions, and 5 patients (15.2%) died. Conclusion Digestive system dysfunctions following cardiovascular surgery were uncommon but had a high mortality. Advanced ages, history of gastrointestinal disease and perioperative hemodynamic unstability may be the clinical risk factors of digestive system dysfuctions. Early diagnosis and prompt treatment are essential for the outcome of the patients.

Microbiology 1992;0(05):-
Bacterial biodiversities of microbial mat and sediments, which were sampled from thermal springs of Tengchong Rehai in Yunnan, were preliminarily studied with PCR-denaturing gradient gel electrophoresis (PCR-DGGE). Directly extracted total DNA from environmental samples amplified by PCR with two sets of bacteria-specific primers. The PCR products, which include the V 8 and V 9 high-variable regions respectively, were analyzed by using DGGE. The DGGE profiles not only indicated the existence of higher levels of bacterial diversity, but also showed that the microbial mat and sediments have different dominant bacteria. Furthermore, the bacterial PCR-DGGE displayed clear profiles of bacterial structure selected by the key abiotic factors of the extreme environments, such as temperature and concentration of oxygen.
