1.Effects of curdione on cognitive function and expression of inflammatoryfactors in the hippocampus after partial hepatectomy in aged mice

Objective To explore the effects of curdione on cognitive function and expression of inflammatory factors in the hippocampus after partial hepatectomy in aged mice.Methods The animals were divided into control group, sham group, operation group, and high-, medium-and low-dose curdione groups.The model of partial hepatectomy in aged mice was established according to the method reported in literature.The levels of SOD, MDA, CAT and GSH-Px in the hippocampus were detected by enzyme-linked immunosorbent assay (ELISA), while the expressions of NF-κB, IL-1β, IL-6 and TNF-α were measured by Western blot.Results The escape latency was significantly increased (P<0.01), and the number of platform-site crossovers was significantly decreased in operation group (P<0.05).Compared with those in operation group, curdione significantly decreased escape latency and increased platform-site crossovers (P<0.05, P<0.05).The levels of SOD, CAT and GSH-Px in the hippocampus were significantly decreased (P<0.05) and the content of MDA was significantly increased after partial hepatectomy (P<0.01), and these were reversed by curdione (P<0.05, P<0.05, respectively).The expressions of NF-κB, IL-1β, IL-6 and TNF-α protein were significantly inhibited by curdione (P<0.05).Conclusion Curdione can significantly improve cognitive dysfunction after partial hepatectomy in aged mice and the mechanism may be related to its inhibition of oxidative stress, inhibition of NF-κB activation and reduction of inflammatory cytokines IL-1β, IL-6 and TNF-α in the hippocampus.

6.Combined Effects of Fluoride and Aluminum on Liver Function of Mice

Objective To study the combined toxic effects of fluoride and aluminum on the liver of mouse and the probable mechanism. Methods Kunming mice were randomly divided into nine groups and treated with fluoride and aluminum through drinking water:control(tap water)group, 50 mg/L fluoride group, 150 mg/L fluoride group, 200 mg/L aluminum group, 600 mg/L aluminum group, 50 mg/L fluoride+200 mg/L aluminum group, 50 mg/L fluoride+600 mg/L aluminum group, 150 mg/L fluoride+200 mg/L aluminum group, 150 mg/L fluoride+600 mg/L aluminum group. After 24 weeks of treatment, the activity of ALT, AST, the content of TBIL, DBIL, IBIL, ALB, Ca, P and Mg in the serum was determined. Results Different doses of single fluoride or fluoride+aluminum led to significant different changes in the liver function(single fluoride:F=2.61, P0.05). Compared with the control group, the Ca level in the serum decreased in the exposed groups except 150 mg/L fluoride+600 mg/L aluminum group(P0.05). Conclusion The combination of fluoride and aluminum at certain levels has a toxic effect on the liver, but the combination of fluoride and aluminum at high doses shows an antagonistic effect.

9.Induction of apoptosis by Panax quinquefolium effective parts (PQEP) on K562 cells

Aim This research was performed to study the anticancer effects of Panax quinquefolium effective parts (PQEP) in human chronic myeloid leukemia (K562). Method After K562 cells were routinely cultured,MTT assay was performed for cytotoxicity test. Cytotoxicity of PQEP (6.25~400 mg?L -1) in K562 cells was increased in a dose-and time-dependent manner. To explore the mechanism of cytotoxicity, we used several measures of apoptosis to determine whether these processes were involved in PQEP-induced leukemic cell death.Results PQEP induced the cell shrinkage, cell membrane blebbing, chromosomes condensation and DNA fragment. In addition, the flow cytometric analysis revealed PQEP (10~400 mg?L -1) dose-dependently increased apoptotic cells with hypodiploid DNA contents. Conclusion These results indicate that PQEP can control leukemic K562 cells through apoptosis and may have a possibility of potential anticancer activities.
