Neurodegenerative diseases are chronic lesions caused by neuronal degeneration and secondary demyelination.Recent studies have shown that a DNA/RNA binding protein named fused in sarcoma/translocated in liposarcoma protein (FUS/TLS) is associated with neurodegenerative diseases,such as amyotrophic lateral sclerosis (ALS),frontotemporal dementia (FTD) and polyglutamine diseases.Mutations or dysfunction in FUS/TLS are responsible for the abnormal processes in the related gene transcription,RNA processing,transportation and translation and are highly correlated with the development of neurodegenerative disease.Discovery and the research of FUS/TLS not only contribute to the understanding of neurodegenerative disease at RNA level,but also provide a new way for the prevention and treatment of neurodegenerative diseases.

4.Expression of microRNA in plasma of gallbladder cancer and its clinical significance

Objective To investigate the expression of microRNA in plasma of gallbladder cancer and its clinical significance.Methods miRNA microarray was performed in seven gallbladder cancer tissues and seven adjacent normal gallbladder tissues to identify differentially expressed miRNA.The results were validated in plasmas of 40 gallbladder cancers and 20 health people with real-time polymerase chain reaction (RT-PCR).Results Eleven gallbladder cancer-related miRNAs were identified with miRNA microarray analysis.Among them,miRNA-21,miRNA-370,miRNA-187,miRNA-122,and miRNA-202 were up-regulated,and let-7a,miRNA-200b,miRNA-143,miRNA-31,miRNA-335,and miRNA-551 were down-regulated in gallbladder cancers.Moreover,let-7a,miRNA-21,miRNA-187,miRNA-143,miRNA-202,and miRNA-335 were validated with RT-PCR,which were consistent with miRNA microarray results with significant difference between gallbladder cancers and health people (P < 0.05).Furthermore,miRNA-187,miRNA-143,and miRNA-202 were related to clinical and pathological features(P <0.05).Conclusions These data indicate that The differentially expressed plasma miRNAs are related to gallbladder carcinogenesis and might be the novel biomarkers of gallbladder cancers.
5.Clinical value of transvaginal ultrasound combined with abdominal ultrasound in the diagnosis of gynecologic acute abdomen

Objective To discuss value of transvaginal ultrasound combined with abdominal ultrasound in the diagnosis of gynecologic acute abdomen.Methods 102 cases with gynecologic acute abdomen who were treated in our department were selected,they got through transvaginal ultrasound combined with abdominal ultrasound.The diagnosis result and pathologic diagnosis,simple ultrasound diagnosis result were compared.Results Compared with transvaginal ultrasound,abdominal ultrasound diagnosis coincidence rate obviously decreased,the difference was statistically significant (t =5.17,P <0.05 ),ultrasound combination diagnosis coincidence rate all obviously increased,the difference was statistically significant (t =11.25,P <0.01 ).The highest ultrasound combination diagnosis coincidence rate was heterotopic pregnancy (100.00%),the lowest was perforation uterus (75.00%). Average ultrasound combination diagnosis coincidence rate in this research was 86.78%,misdiagnosis rate was 1.96%,and missed diagnosis rate was 3.92%.Conclusion Accuracy rate of transvaginal ultrasound combined with abdominal ultrasound pathologic diagnosis results,have these following advantages,such as high accuracy rate,rapid diagnosis,suitable for gynecologic acute abdomen diagnosis.

To investigate the relationship between the expression of cyclooxygenase-2 (COX-2) and menstrual cycle, the regulatory effects of 17-beta-estradiol (E(2)) and medroxyprogesterone acetate (MPA) on the expression of COX-2 in cervical cancer Hela cells were examined. Cervical cancer specimens were obtained from 47 pre-menopausal patients. The phase of menstrual cycle was determined by case history and HE staining of uterine endometrium. COX-2 was immunohistochemically stained by SABC staining and the staining intensity was determined with computerized image analysis system. Hela cells were incubated with alcohol, E(2), E(2)+MPA, MPA for 12, 24 and 48 h respectively. The expression of COX-2 in Hela cells was detected by Western blotting and reverse transcriptase-polymerase chain reaction (RT-PCR). Our results showed that the expression of COX-2 was significantly higher during proliferative phase than secretory phase (P<0.05), but there was no difference in the positive rate between proliferative phase and secretory phase (P>0.05). Incubation with E(2) could significantly enhance the expression of COX-2 continually. On the contrary, E(2)+MPA and MPA alone could decrease the expression of COX-2 as compared with the control and E(2) group (P<0.05 and P<0.01 respectively). It is concluded that the expression of COX-2 in cervical cancer of pre-menopausal patients and Hela cells was regulated by estrogen/progestogen.

9.Cementless total hip arthroplasty for the developmental dislocation of hip in adults with high dislo-cation

Objective To evaluate the operative technique and the short-term clinical results of ce-mentless total hip arthroplasty (THA) for developmental dysplasia of the hip (DDH) in adults with high level dislocation. Methods Nine patients (11 hips) with DDH of Hartofilakidis type Ⅲ, who underwent cement-less THA between 1997 and 2002, were analyzed. Of 9 patients, 8 were females, and 1 male with an average age of 29.4 years (range, 17 to 56 years). There were unilateral DDH in 7, and bilateral in 2. The patients had no previous surgery. The preoperative average length difference between the two legs was 4.1 cm (range, 0 to 7 cm). The femoral head was dislocated upward from the normal position by 4.8 cm on average (range, 3.2 to 7.0 cm). The Zweym?ller cup was placed on the original acetabulum in all patients with more than 80% bony coverage on the cup, and no bone graft needed. In 4 hips with more than 5 cm of displacement, the reduction of prosthesis was achieved by shortening osteotomy below lesser trochanter. The preoperative average Harris score was 40.2. Results All of 9 patients were obtained follow-up of X-ray films and clinical evaluation. The follow-up ranged from 6 to 32 months with a mean of 18.2 months, the osteotomy of 4 hips were bony healed at 3 months postoperatively showed by X-ray films. The postoperative average difference of the two leg length reduced to an average 1.1 cm (range, 0 to 3 cm), and the postoperative average Harris score increased to 90. Femoral nerve impairment occurred in 1 case, and recovered 4 months later. There were no loosening, dislocation and infection at follow-up. Conclusion Based on the short-term results of the cementless total hip arthroplasty for the developmental dislocation of the hip in adults with high level dislocation, it is proven to be a good technique, and the satisfied clinical results would be obtained.

Purpose:To study the secretion and gene expression of Angiogenesis factors in the patients with CML and study the effect of Angiogenesis in the occurrence and development of CML. Methods:Concentration of VEGF in peripheral blood was determined by using ELISA. Myeloid tissue was extracted from all CML cases and ITP patients to detect MVD by using CD34 labelling. At the same time the level of VEGF,b-FGF were detected by using RT-PCR in both peripheral blood and myeloid cells.Results:Our results showed that the concentration of VEGF was obviously higher in the peripheral blood of CML patients(177.53?153.45 pg/ml) than in those of control group(73.12?19.82 pg/ml). The gene expression of VEGF and b-FGF were both higher than those of control group. The difference has statistical significance(P
