1.Construction of specifically expressed vascular endothelial growth factor_(165) gene in retina

Objective To construct specifically expressed vascular endothelial growth factor (VEGF) 165 gene in retina. Methods Rho promoter, specifically expressed in retina, was amplified by polymerase chain reaction (PCR) from the genomic DNA of a BLAB/C rat, then it was cut with restriction enzymes and cloned into the plasmid pcDNA 3.1+-VEGF 165 to form recombinant plasmid pcDNA 3.1+-rho-VEGF 165. The correct recombinant plasmid pcDNA 3.1+-rho-VEGF 165 was identified by restriction enzymes and PCR, and was transferred by jetPEI into cultured human navel vein endothelial cells and human retinal pigment epithelial (RPE) cells. The expression of VEGF protein in human navel vein endothelial and RPE cells was detected by immunocytochemical staining and protraction of the growth curve of the cells. Results In human RPE cells, the expression of VEGF protein was more in recombinant plasmid pcDNA 3.1+-rho-VEGF 165 than that in plasmid pcDNA 3.1+-VEGF 165; in human navel vein endothelial cells, no obvious difference of the expression of VEGF protein between recombinant plasmid pcDNA 3.1+-rho-VEGF 165 and plasmid pcDNA 3.1+-VEGF 165 was found. Conclusions The construction of pcDNA 3.1+-rho-VEGF 165 carrier may provide the basic material for the study of the nosogenesis of VEGF in retinal neovascularization, and establish the foundation to set up the model of transgenic mice with VEGF specific expressing in retina.

Predictive factors of neoadjuvant chemotherapy play an important role in selecting sensitive chemotherapy regimen and avoiding unsuitable regimen for breast cancer patients. At present, estrogen receptor (ER), progestin receptor (PR) and HER-2 status are the main predictive factors. Other factors such as Ki-67 also seem to be promising for optimizing neoadjuvant chemotherapy.

4.Imbalance between proliferation and apoptosis of cholangiocytes and its relation to cholangiopathies

Cholangiocytes are the epithelial cells that line the intrahepatic biliary tree and also the target of cholangiopathies. Despite the heterogeneous etiology ,most of cholangiopathies share the common pathological process which is the imbalance between proliferation and apoptosis of cholangiocytes. The article is a review about the characteristics and mechanism of proliferation and apoptosis of cholangiocytes and their relationship to cholangiopathies.
5.Progression of immunomodulatory molecules in the change of can-cer chemotherapy sensitivity

Intrinsic or acquired chemo-resistance is the main reason for chemotherapy failure, and thus finding ways to reverse chemo-resistance has become an interesting topic for research. Studies have revealed that immunomodulatory molecules are involved in cancer chemo-resistance. Hence, interventions that target immunomodulatory molecules to reverse chemo-resistance have attracted a great deal of concern from domestic and foreign scholars. Immunomodulatory molecules, such as PD-L1, B7-H3, HMGB1, TRAIL, MyD88, and Cytokines (TNF-α, IFN-α, IL-6), have been proven to take part in regulating immune function and tumor drug-resistance characteristics, thereby providing new ideas to the reversal of tumor chemo-resistance. This artide reviews the progression of immuno-modulatory molecules with the change in cancer chemotherapy sensitivity to provide a theoretical basis for the application of new thera-peutic regimen of bio-chemotherapy.

8.Treatment of 58 Cases of Knee Osteoarthritis with Acupuncture and Moxibustion Theray

Of all 58 cases of knee osteoarthritis treated by acupuncture and moxibustion, 42 cases were recovered,(accounting for 72.5%), 14 cases were obviously improved (accounting for 24.1%), and 2 cases were ineffective (accounting for 3.4%). The total effective rate was 96.6%. This result demonstrated that acupuncture and moxibnstiou therapy has the good functions of dredging meridians and collaterals, activating qi and blood circulation, dispelling cold and dampness, and removing swollen to stop pain.

10.Clinical Study of 106 Myasthenia Gravis in Children

Objective To explore the clinical characteristics of myasthenia gravis(MG)in children.Methods One hundred and six children with MG in Children′s Hospital of Chongqing Medical University from Apr. 2002 to Apr. 2007 were concluded in the study.The data of laboratory examinations,including virus antibody,autoantibody test,immunity function,chest X-ray or chest CT,MRI,ECG and repetitive nerve stimulation, and treatment of cholinesterase inhibitor,glucocorticoids and high-dose immunoglobulin were reviewed.Results Among 106 children,male-to-female ratio was 1 to 1.16,male 49 cases,female 57 cases.The course of disease was 3 days to 11 years.The age of onset of all 106 children with MG varied from 10 months to 14.3 years old and the mean age was (4.74?3.32) years old.The most common age of onset was less than 3 years old(55.7%).There were 41 cases newly diagnosed children and 65 cases received irregular treatment or relapsed after treatment.The ocular muscular symptoms were mostely found at the onset(84.0%);brainstem type was involved at the onset(2.8%),generalized type was involved at the onset(13.2%);58 out of 106 cases(54.7%) had upper respiratory tract infection before the onset with MG; 41 out of 52 cases with MG had abnormal immune function or auto antibody-positive,myocardial enzymes increased in 45 cases increased in varying degrees,thymoma were not found by use of sternum or CT or MRI in 69 cases,repetitive nerve stimulation of 7 out of 11 cases were abnormal; 17 cases (16.0%) of the children received single pyridostigmine treatment,10 cases (9.4%) received co-adrenal hormone and before steroid given IVIG therapy respectively were satisfied with the efficacy,and gravis symptoms were improved;61.3% of the cases had a relapse who underwent irregular treatment or relapse after treatment,before relapse 37 cases (56.9%) received single cholinesterase inhibitor treatment, 28 cases (43.1%)for co-adrenal hormone, after adrenocortical hormone and pyridostigmine treatment.The prognosis of 72 patients(67.9%) with MG was usually good; the causes of relapse resulted from infection (30 cases, 46.2%), irregular drug use (22 cases,33.8%), and drug withdrawal and infection (13 cases, 20.0%).Conclusions As early as age of onset,the disease easily repeated,to be early co-adrenal hormone therapy group is the children′s clinical features of MG.
