1.Effects of WNT5B over-expression on viability and apoptosis of human breast cancer cell line MCF-7

AIM: To detect the expression of WNT5B in normal breast epithelial cells and different breast cancer cell lines, and to investigate the effects of WNT5B over-expression on the viability and apoptosis of human breast cell line MCF-7.METHODS: The mRNA expression of WNT5B was detected by RT-PCR in different breast cancer cells. MCF-7 cells were transfected with plasmid pcDNA3.1 /WNT5B or pcDNA3.1, and the expression of WNT5B at mRNA and protein levels was examined in the 2 groups by real-time PCR and Western blotting, respectively.Subsequently, the chan-ges of cell viability and cell apoptosis were analyzed by CCK-8 assay and flow cytometry, respectively.RESULTS: The ex-pression of WNT5B in the breast cancer cell lines was lower than that in MCF10A cells.The WNT5B expression in the MCF-7 cells in experimental group was significantly higher than that in vector group (P <0.05).However, the cell viabili-ty in experimental group decreased significantly as compared with vector group (P <0.05).The number of the cells in S-phase obviously increased, while the percentage of the cells in G1-phase and G2 /M-phase decreased compared with vector group.The number of apoptotic cells in WNT5B group was significantly higher than that in vector group.CONCLUSION:The expression of WNT5B is decreased in breast cancer cells.WNT5B over-expression significantly inhibits the cell growth and promotes the cell apoptosis in breast cancer MCF7 cells.

Background and purpose:A chemokine receptor,CXCR4,and its endogenous ligand,stromal cell-derived factor-1(SDF-1),have been recognized to be involved in the invasion and metastasis of cancer.Inhibition of CXCR4 may be a new therapeutic target.We studied whether shRNA-CXCR4 could inhibit the CXCR4 gene expression and the proliferation in MCF-7,MDA-MB-231 and MDA-MB-435s breast cancer cells.Methods:Through knockdown CXCR4 by shRNA,the CXCR4 mRNA expression was detected by RT-PCR and the CXCR4 protein expression was mesured by Western blot.The proliferation of breast cancer cells was evaluated by MTT and flow cytometry.Results:The CXCR4mRNA and its protein expression decreased significantly in MCF-7(the average level of CXCR4 mRNA and protein expression were 0.089,0.177 in PG-CXCR4 group,compared with 0.327 and 0.313 for mRNA expression,0.911,0.874 for protein expression in control and PG-HK group),MDA-MB-231(the average level of CXCR4 mRNA and protein expression were 0.152 and 0.153 in PG-CXCR4 group,compared with 0.40 and 0.45 for mRNA expression,0.829 and 0.878 for protein expression in control and PG-HK group)and MDA-MB-435s(the average level of CXCR4 mRNA and protein expression were 0.198 and 0.173 in PG-CXCR4 group,compared with 0.69 and 0.77 for mRNA expression,0.877 and 0.906 for protein expression in control and PG-HK group)breast cancer cells(P
3.Association between Airborne Pollen Concentrations and Human Respiratory Diseases

Objective To investigate the relationship between airborne pollen concentrations and the number of medical consultations in out-patient and emergency for respiratory diseases in Haidian district,Beijing. Methods To collect the data of airborne pollen concentrations,atmospheric concentrations of total suspended particles(TSP),the related meteorological factors (average temperature,maximum temperature,minimum temperature,average air pressure,precipitation and relative humidity) and the data of patients who were diagnosed with respiratory diseases including acute pharyngitis,acute laryngitis,acute bronchitis,anaphylactic rhinitis and bronchial asthma from database on medical insurance,from April to September in 2000 to 2002 in Haidian district,Beijing. The partial correlation coefficients between airborne pollen concentrations and the number of medical consultations for respiratory diseases in spring (from April 1st to June 16th),and in summer-autumn (from July 15th to September 30th) were calculated after controlling TSP concentrations and the six related meteorological factors. Results The airborne pollen concentrations in these three years showed two obvious peaks in spring and summer-autumn,and each of the five respiratory diseases has its specific change character. 4.Protection of ginsenoside on cultured cells from virus

The protection of ginseno-side (GNS ) isolated from leaves and stems on cultured cell from virus were observed. The results showed that GNS, especially the Rb1 monomer could suppress replication of virus such as HSV -Ⅰ , HSV-Ⅱ ,ADV- Ⅲ and VSV, and protect the cultured cells from virus infec-tion. A cream containing GNS was used in the treatment of herpes lebialis infected by HSV- I and 27 of 31 (87. 1%) cases were cured.

Objective To study the expression of chemokine receptor-4(CXCR4) in human breast cancer and its correlation with the prognostic factors.Methods Forty-four samples of breast cancer were collected from Sep.2004 to Jan.2006.The expressions of CXCR4 mRNA and protein were evaluated by RT-PCR and Western blotting respectively.The expressions of CXCR4,estrogen receptor(ER),progesterone receptor(PR) and C-erbB-2 were examined by immunohistochemical methods.The clinicopathological features,such as ages,menstrual status,lymph node metastasis,tumor size and histological grading were recorded and analyzed.The relationship between CXCR4 and the clinicopathological features was analyzed.Results The immunohistochemical staining showed that CXCR4 expressed mainly in cytoplasm and little in nuclei.CXCR4 mRNA and protein expressed differently in 44 samples of breast cancer,and the expressive level increased with the increase in metastatic lymph nodes number.Compared with axillary lymph nodes metastasis negative group,the expression of CXCR4 mRNA and protein increased significantly in ≥4 and 1-3 nodes metastasis groups(P0.05).Conclusion CXCR4 might play an important role in promoting the metastasis of breast cancer.

8.Surgical Site Infection Analysis for Orthopedics Patients

OBJECTIVE To analyzed the conditions and tendency of surgical site infection(SSI) occurring in orthopedics patients at 11 hospitals in Beijing from 2002 to 2005.METHODS By sorting the case history database in the computer,the needed case and relative information were first selected.The conditions and tendency that SSI occurring were described.RESULTS The ratio of SSI in orthopedics was 0.14%,from which 0.09% were in type Ⅰ section,0.65% in type Ⅱ section and 1.78% in type Ⅲ section.CONCLUSIONS This paper shows that the different types of surgical section induce different SSI rate.
9.Transurethra Resection of the Prostate for High-Risk Benign Prostatic Hyperplasia

Objective To evaluate the safety and efficacy of transurethral resection of the prostate(TURP)for high-risk benign prostatic hyperplasia(BPH).Methods Form January 2001 to July 2007,we performed TURP on 303 patients with high-risk BPH.The patients aged form 65 to 89 years with a mean of 76.3.Among them,151 were complicated with cardiovascular diseases,67 had pulmonary diseases,37 had diabetes mellitus,26 had brain disease,16 showed renal inadequacy,and 6 showed abnormal liver function.Results The procedure was completed successfully in all of the 303 cases without death,dysuria,or severe complications.Compared to the examinations carried out before the procedure,the IPSS and QOL scores,and RU decreased significantly,while the Qmax increased 3 months after the prosedure[IPSS:(19.6?7.9)points vs.(6.4?3.3)points,t=26.838,P=0.000;QOL:(4.7?1.5)points vs.(2.3?1.1)points,t=22.459,P=0.000;Qmax:(4.1?2.6)ml/s vs.(13.9?7.1)ml/s,t=-22.561,P=0.000;and RU:(139.0?32.5)ml vs(13.5?8.6)ml,t=55.143,P=0.000].ConclusionsTURP is a safet and effective procedure for high-risk BPH with a low rate of postoperative complications.Proper peri-operative individual management and operation skills are important for the success of the procedure.
10.Correlation of the nutritional status with liver function and clinical outcomes in surgically treated liver cancer patients

Objective To investigate the relationship of the nutritional status with liver function and clinical outcomes of liver cancer patients treated with surgery.Methods Altogether 112 hospitalized patients undergoing surgical treatments for liver cancer were enrolled from October 2011 to October 2013.Their general clinical data were collected,including creatinine-height index (CHI),arm circumference,grip strenghth,albumin,prealbumin,and transferrin.The nutritional status was assessed using Patient-Generated Subjective Global Assessment (PG-SGA).The liver function was assessed with Child-Pugh classification.Postoperative infectious complications and the hospital stays were recorded to assess the clinical outcomes.The correlation between nutritional status and liver function,and that between nutritional status and clinical outcomes were analyzed.Results Among the 112 patients,70 (62.5%) were in normal nutritional status,34 (30.4%) were with moderate malnutrition,and 8 (7.1%) were with severe malnutrition according to PG-SGA scores.PG-SGA assessment showed strong consistence with CHI nutritional assessment (κ =0.760,P =0.000),and moderate consistence with arm circumference assessment (κ =0.564,P =0.000),and grip strength assessment (κ =0.523,P =0.000).The live function classified by Child-Pugh was found highly correlated with PG-SGA assessment (rs =0.829,P =0.000).Postoperative infectious complications and hospital stays were both positively correlated with PG-SGA assessment (r =0.349,P =0.000 ; r =0.624,P =0.000).Conclusions PD-SGA combining with CHI can be used for the nutritional status assessment of liver cancer patients undergoing surgical treatments.The nutritional status of the patients has positive correlation with live function,infectious complications,and postoperative hospital stays.
