1.Study on changes of ROS, GSH content, and GSH - PX activity of serum in persons under different state of exercise

ObjectiveTo probe the alterated characteristic of reactive oxygen species, reduce glutathione and glutathione peroxidase (GSH - PX) in serum of persons. Method Under the controlled load by treadmill, correlative indexes were determined during resting state, sub - maximalload and exhaustive exercise. ResultROS level of serum increased significantly during sub- maximal load compaired with that of restingstate. The ROS level during exhaustive exercise was higher than that of sub - maximal load( P <0. 01 ) . GSH content and GSH - PX activity de-creased significantly( P <0. 001 ). GSH content and GSH - PX reactivity decreased significantly( P <0. 001 ) during sub - maxiaml load and in-creased after exhaustive exercise which was still lower that of resting state. Conclusion ROS increased with the enhancement of exercise intensityand the time. GSH and GSH - PX showed a significant decrease in content and activity respectively followed by apparent enhancement.

Objective To explore the CD4 + CD25 + Foxp3 + regulatory T cell percentage and plasma levels of soluble CD25 molecules in peripheral blood of septic patients and their clinical value through prospective study. Method A total of 37 septic patients and 15 non-infectious SIRS patients, who conformed to the criteria of SIRS and sepsis which proposed by American College of Chest Physicians/Society of Critical Care Medicine Consensus Conference ( ACCP/SCCM ) in 1997, were collected in ICU of Ruijin Hospital ( Shanghai Jiaotong University) from February 2009 to February 2010. Twenty-four health people were from Medical Center of Ruijin Hospital, who were excluded infection and (or) autoimmune diseases. There were 26 male and 11 female in sepsis group, average age ( 61.67 ± 11.87 ) years old; 8 male and 7 female in SIRS group, average age (67.06 ± 12.57)years old; 14 male and 10 female in health control, average age (56.54 ± 6.37 )years old. All selected patrents were excluded the autoimmune diseases and (or) patients within recent (30 days) had used or now used immunosuppressive agents. We therefore measured the Treg cell percentage in peripheral blood by Flow Cytometry and the plasma levels of IL-2sRa, IL-4, IFN-γ by ELISA. The data were analyzed by analysis of variance or nonparametric Kruskal-Wallis H test. Results ① The percentage of CD4 + CD25 + Foxp3 + regulatory T cells among septic patients, SIRS patients, and control group was: ( 66.82 ± 21.79 ) %, ( 51.79 ± 21.79 ) %, ( 56.45 ± 10. 68 ) %, respectively. septic patients showed the highest percentages of CD4 + CD25 + Foxp3 + regulatory T cell among CD4 + CD25 + T cells(P < 0.05 ). ② The plasma levels of soluble CD25 in septic patients (425. 619 ± 270.12 ) were significantly higher than SIRS patients (381. 664 ± 189.83) and the control group ( 164. 1 32 ± 56.37 ) ( P < 0.05 ). ③ The correlation analysis between the concentration of soluble CD25 molecules in plasma and the ratio of CD4 + CD25 + Foxp3 + regulatory T cells to CD4 + CD25 + T cells showed Spearman correlation coefficient =0.390, P = 0.003 ( P < 0.05 ). Conclusion: the expression of natural regulatory T cells characteristically increased in septic patients. And the levels of soluble CD25 in peripheral blood were related to the percentages of natural regulatory T cells, which simplified the assessment of the immune status in Septic patients.

Increasing detection of unruptured intracranial aneurysms,catastrophic outcomes from subarachnoid hemorrhage,and risks and cost of treatment necessitate defining objective predictive parameters of aneurysm rupture risk.However,long-term follow-up have shown the risk of intracranial aneurysm rupture is associated with its morphologic characteristics,hemodynamic factors and patient's own situation.

Objective To evaluate the clinical value of 3D-FS-SPGR and 3D-reconstructional technique in detecting the lesion of articular cartilage of knee.Methods Fifty eight knees with OA(42 cases)or trauma (9 cases) in 54 cases (12 cases were confirmed by operation) were imaged with eight different MR imaging sequences including T 1-weighted,proton density-weighted,T 2-weighted FSE,STIR,3D-FS-SPGR,2D-FS-SPGR,3D-SPGR and 3D-FS-GRE.The classification and three-dimensional reconstruction of cartilage in four locations of knee were performed on different MR sequences.The detecting rate of cartilage lesion were calculated.The results in partly cases were analysed compared to operation. 7.Clinical Observation on the Relationship between Helicobacter Pylori and Peptic Ulcer and Chronic Gastritis

Patients with Helicobacter pylori (Hp) positive peptic ulcer were randomly treated with Hp-killing or Hp-nonkilling anti-ulcer drugs, and 19 patients with healed peptic ulcer were followed up to determine the relapse rate of peptic ulcer. It was found that there was a close relationship between Hp infection and the existence of active chronic gastritis, and the clearance of Hp infection could not alleviate the activity of chronic gastritis or influence the 3-week healing rate of peptic ulcer. The relapse rate of the Hp-positive ulcer patients was higher than those Hp-negative cases.

Objective To investigate the effect of pancreatic kininogenase on the expression of matrix metalloproteinases-2 2(MMP-2), transfer growth factor-?_ 1 (TGF-?_ 1 ) and ventriculer remodeling in spontaneously hypertensive rats (SHR). Methods Twenty-four male 15 weeks SHR were randomly divided into three groups: SHR group, pancreatic kininogenase treatment group(PK: 7.2 U/kg?d), captopril treatment group(Cap: 10 mg/kg?d)(n=8 in each), 8 Wister Kyoto were served as control. Objective To explore the effects of surface functionalized multi-walled carbon nanotubes (FMWCNTs) on the cytotoxicity of human peripheral blood mononuclear cell (PBMC).Methods Five different types of MWCNTs (hydroxylated,carboxylated,aminated,nickel-plated and pristine MWCNTs (P-MWCNTs)) with the same diameter and length were evaluated the dispersion and characterizations in physiological salt solution by transmission electron microscopy.PBMC were isolated by density gradient centrifugation from human peripheral blood,and 5 types of MWCNTs were ultrasonically dispersed in serum-containing medium respectively.After incubation with PBMC for 12,24,48 or 72 h,cytotoxicity was detected by CCK-8 kits.Results All the MWCNTs had well dispersion,especially the F-MWCNTs.Cytotoxicity results showed that all types of MWCNTs could induced PBMC death,and presented dose-dependence manner and a certain degree of time-dependence manner.Compared with the P-MWCNTs,F-MWCNTs changed cytotoxicity statistically,with the hydroxylated,carboxylated,aminated MWCNTs weakened,aminated MWCNTs significant (P<0.05),nevertheless the nickel-plated MWCNTs increased.Compared with the P-MWCNTs (25 μg/ml),cell viability of PBMC after 24 and 48 h incubation with the same dose of nickelplated MWCNTs both decreased,and the differences was statistically significant (P<0.01,P<0.05).Conclusions The functional group modification affects not only the MWCNTs dispersion in medium,but also the cytotoxicity of the MWCNTs on PBMC.
