1.A new steroidal glycoside from Lygodium japonicum

To investigate the chemical constituents from the aerial parts of Lygodium japonicum. Methods:Various chromatographic techniques were employed for isolation and purification of the constituents. The structures were elucidated by chemical evidence and spectral methods. Results:A new stigmasterol glycoside,(24R)-stigmastan-3β,5α,6β-triol 3-O-β-D-glucopyranoside (1),together with three known phenolic glycosides:6-O-p-coumaroyl-D-glucopyranose (2),6-O-caffeoyl-D-glucopyranose (3),1-O-(E)-caffeoyl-β-D-gentiobiose (4) were obtained and identified by spectroscopic methods. Conclusion:All compounds were isolated from Lygodiaceae for the first time.
2.A new ginkgolide from Ginkgo biloba

Aim: To study new chemical constituents of the leaves of Ginkgo biloba L. Methods: Isolation and purification were carried out by several chromatographic methods. The structures of the compounds were elucidated by detailed analysis of their UV, IR, MS, ~1H NMR and ~(13)C NMR spectra Results: A new ginkgolide, ginkgolide N( 1, 7, 10-trihydroxy-3,14- dehydroginkgolide, Ⅱ), along with a known compound, was isolated from the leaves of G. biloba. The structure of the known one was elucidated as ginkgolide L(Ⅰ). Conclusion: Compound Ⅱ was a new compound. The complete spectroscopic data of compound Ⅰ were reported for the first time.
3.Detection of pigment epithelium derived factor gene mutation in a human malignant melanoma cell line A375

Objective To detect the mutations of pigment epithelium derived factor (PEDF) gene in a human malignant melanoma cell line A375. Methods A375 cells and control melanocytes obtained from circumcised prepuce were cultured, genomic DNA was extracted from these cells. All eight exons of PEDF gene were scanned by single strand conformation polymorphism analysis ofpolymerase chain reaction products (PCR-SSCP) in both A375 cells and control melanocytes. DNA sequencing was performed for the PCR products separated into electrophoretic bands with altered mobility. Results Altered mobility was observed with SSCP analysis in amplicons of exon 3, 4, 5, 6 and 7, with the most obvious alteration occurred in exon 5 and 6. DNA sequencing revealed mutations in both exon 5 and 6. The common type of mutations was single base-deletion in exon 5 and single base-substitution in exon 6. Conclusion Mutations of PEDF gene may contribute to the development of human malignant melanoma.
4.Determination of acteoside isomer in Total Cistanches Glycosides Capsules by HPLC

Objective:To establish a determination of acteoside isomer in Total Cistanchis Glycosides Capsules by HPLC.Methods: HPLC analysis was carried out on C 18 ODS column with acetonitrile——1.5% acetic acid as the mobile phase. The linear gradient of acetonitrile was from 11.5% to 20%(0-25min). The detection wavelength was 335nm. Results: The method was linear in the range of 0.2~1.0?g, r =0.9996. The average recovery was 98.85%, RSD was 1.51%( n =9). Conclusion:This method is simple, accurate, highly sensitive and reproducible. It may be used for the determination of acteoside isomer in Total Cistanchis Glycosides Capsules.

6.Narrow-band ultraviolet-B(311 nm)regulates dendrite formation in cultured B16 melanoma cells

Objective To investigate the effect ofnarrow-band ultraviolet-B(NB-UVB)(311 nm)on dendrite formation in B16 melanoma cells.Methods B16 melanoma cells were irradiated with various doses of NB-UVB(0,25,50,100,200,300 mJ/cm2).Atier additional culture of varying durations,irradiated cells were harvested and subjected to the observation of morphological changes and cell cytoskeleton F-actin microfilaments by phase contrast microscopy and laser scanning confocal microscopy(LSCM).respectively,and to the detection of cell proliferation bv MTT colorimetric assay.Pull down assay was performed to detect the activity of GTP-RhoAA and GTP-Rac1 in B16 cells before and after UVB irradiation.Results Twenty-four hours after irradiation with UVB of 100 mJ/cm2.an increase was observed in the cell body of B16 cells which appeared in sphericity,as well as in the number of dendrites(P<0.01)which showed a branch-like appearance.compared with non-irradiated cells which had 2-3 dendrites and obscure branches.LSCM revealed that F-actin microfilaments in B16 cells were well organized with clear textures before irradiation;after irradiation wim NB-UVB of 100 mJ/cm2.stress fibers were disassembled and disrupted and the texture became unclear,which was observed as early as 30 minutes and became more and more evident,and at 6 hours the stress fibers displayed a clumping appearance with obscure textures.Following the irradiation with NB-UVB of 100 mJ/cm2,the expression level of GTP-Rac 1 protein increased at l 5 minutes,and.at 30minutes,reached 2 times of that observed in nonirradiated cells,then decreased a liale,but still remained elevated at 60 minutes and 120 minutes,compared to unirradiated cells;meanwhile.the level of GTP-RhoA dropped a little at 30 minutes,then gradually increased and,at 120 minutes.reached 1.6 times of that observed in unirradiated cells.Conclusion Narrow-band UVB(311 nm)can promote dendrite formation.likely via regulating the expression of GTP-Racl and GTP-RhoA in B16 melanoma cells.
7.The surgical outcome of sacral decompression and lumbopelvic fixation for H-shaped sacral fracture and correlation factors analysis

Objective To investigate the correlation factors for surgical outcome of sacral decompression and lumbopelvic fixation in H-shaped sacral fracture and the methods to prevent and treat the complications.Methods From January 2008 to January 2016,45 patients with H-shaped sacral fracture treated by sacral decompression and lumbopelvic fixation were respectively analyzed,including 29 men and 16 women,mean age 41.2 (range,24-53 years),mean follow-up time 52.6 months (range,16-93 months).The surgical outcome was evaluated by pelvic outcome score,and correlation factors were analyzed.We analyzed whether each factor was in correlation with pelvic outcome score.Then we integrated the statistically significant indicators into Logistic regression equation to determine the related factors.Postoperative complications were all recorded.Results The average operation time was 161.2 min (range,100-220 min),average blood loss was 491.6 ml (range,370-1 000 ml),injury-surgery interval was 7.2 d (range,1-23 d).In terms of pelvic outcome score,31 (68.9%) patients had satisfactory result and 14 (31.3%) patients had unsatisfactory result.Univariate x2 analysis suggested that cauda equina injury,Roy-Camille classification,L5S1 facet injury,fracture line,kyphotic angle,injury-surgery interval,decompression approach and inserting sacral screws were correlated with pelvic outcome score,but gender,age,injury mechanism and L5 pedicle fracture had no relation with pelvic outcome score.The multivariate Logistic regression analysis showed that cauda equina injury,Roy-Camille classification,fracture line,kyphotic angle,injury-surgery interval,decompression approach and inserting sacral screws were closely related to pelvic outcome score,but L5S1 facet injury was excluded.2 patients required early surgical procedures with proper antibiotics for deep wound infection;3 patients complained pain related to hardware prominence and the pain subsided after removal of implants;1 patient got unilateral rod breakage at 3-6 months and achieved bony fusion after nine months of observation.Conclusion Sacral decompression and lumbopelvic fixation is effective in neurological recovery and early ambulation in treating H-shaped sacral fracture.Better surgical outcome is related to timely surgery,preoperative or intraoperative bone traction,sacral screws insertion,incomplete cauda equina injury,Roy-Camille type Ⅱ,fracture line penetrating S2 and kyphotic angle less than 40°.
8.Increased Circulating CXCL10 in Non-Segmental Vitiligo Concomitant with Autoimmune Thyroid Disease and Alopecia Areata

BACKGROUND: Vitiligo is a common acquired pigmentary disease caused by destruction of epidermal melanocytes in underlying autoimmune response. Few studies have been focused on the role of chemokines in non-segmental vitiligo (NSV) concomitant with autoimmune thyroid disease (AITD) and alopecia areata (AA). OBJECTIVE: The aim of this study was to determine the best serum biomarker for predictive role in the progression of vitiligo and to evaluate the influence of AA and/or AITD on vitiligo by using the biomarker. METHODS: This prospective cohort study recruited 45 NSV patients: 14 without either AITD or AA, 12 with AITD, 11 with AA, and 8 with both AITD and AA. Serum levels of CXCL1, CXCL8, CXCL9, CXCL10, CXCL12, CXCL13, and CXCL16 were analyzed by ELISA. CXCR3 mRNA expression was detected on PBMCs by RT-PCR. Improvement was evaluated using repigmentation scales. RESULTS: Serum CXCL10 levels, along with the expression of CXCR3 mRNA were higher in NSV patients with AITD or AA alone than in those without AITD or AA. Moreover, serum CXCL10 levels, along with the expression of CXCR3 mRNA were higher in NSV patients with both AITD and AA than in those with AITD or AA alone. Poorer repigmentation was observed in NSV patients with both AA and AITD than in those with AA or AITD alone. CONCLUSION: CXCL10 could be a biomarker to predict the progression of NSV. Dermatologists should pay much attention to those NSV patients concomitant with AITD and/or AA, for comorbidity might lead to more active autoimmune reaction.

9.Symptom-based traditional Chinese medicine slices relationship network and its network pharmacology study.

OBJECTIVETo discover and interpret the correlations between traditional Chinese medicine (TCM) slices and their properties such as function, symptom and channel tropism by constructing the relationship network using network pharmacology approaches.
METHODTCM slices related information was extracted from Chinese Pharmacopeia (2010 edition, volume I) by text mining, and was used to construct the TCM slices-symptom relationship network. The corresponding network analysis was also performed.
RESULTThree thousands and sixteen pair of TCM slice-symptom correlation associated with 646 TCM slices was discovered, and the constructed network unfolded the complex relationships between TCM slices. Further network analysis results indicated that the un-annotated function and channel tropism of TCM slice can be revealed by proposed symptom-based network.
CONCLUSIONNetwork pharmacology approaches can be applied in TCM research to discover and interpret the relationships between TCM slices and their properties.

10.Network pharmacology study on major active compounds of Fufang Danshen formula.

OBJECTIVETo investigate the correlations between multi-compounds of Fufang Danshen formula and their multi targets and multi diseases.
METHODLiterature knowledge of nine major active compounds from Fufang Danshen formula, including tanshinone II(A), salvianolic acid B, protocatechuic aldehyde, danshensu, cryptotanshinone, notoginsenoside R1, ginsenoside Rg1, ginsenoside Rb1 and borneol were collected from PubMed. Combined with cardiovascular related diseases and genes from OMIM database, the corresponding multi-compound-multi- target-multi-disease network was constructed and visualized by Cytoscape software.
RESULTAND CONCLUSION: Network analysis showed that the 9 compounds could modulate 42 cardiovascular associated genes (e. g. PPARG, ACE, KCNJ11, KCNQ1, ABCC8, et al), which related to 30 cardiovascular associated diseases including non-insulin-dependent diabetes mellitus, hyperinsulinemic hypoglycemia, hypertension, and coronary heart disease. These results suggested new potential indications of Fufang Danshen formula.
