1.Stem cell therapy for ischemic heart disease

BACKGROUND:Stem cel transplantation is a promising strategy for treatment of ischemic heart diseases, which has obtained great achievements in recent years. OBJECTIVE: To analyze the clinical trends of stem cel therapy for ischemic heart diseases reported from 2001 to 2015 in Medline, CNKI, Wanfang, ClinicalTrials.gov and Chinese Clinical Trial Register. METHODS:The relevant articles addressing stem cel therapy for ischemic heart diseases were retrieved using the keywords of “stem cel transplantation” and “ischemic heart disease” in Chinese and English in Medline, CNKI, Wanfang, ClinicalTrials.gov and Chinese Clinical Trial Register folowed by statistical analysis. The retrieval time was from 2001 to 2015, including 2001-2005, 2006-2010 and 2011-2015. RESULTS AND CONCLUSION: In Medline database, there were 219 clinical studies about stem cel therapy for ischemic heart disease published from 2001 to 2015, and the number of retrieved articles was most from 2006 to 2010, and decreased from 2011 to 2015. In CNKI and Wanfang databases, the number of relevant articles which had a similar trend with that in the Medline database decreased remarkably after 2011. In ClinicalTrials.gov, there were 63 clinical trials about stem cel therapy for ischemic heart disease, most of which were registered from 2006 to 2015 and came from Europe and North America. In Chinese Clinical Trial Register, there was no clinical trial about stem cel therapy for ischemic heart disease, which may result from the strict management in China.
2.Summary of rectal cancer anastomotic fistula treatment experience

Objective To investigate the cause of postoperative colorectal cancer anastomotic fistula,the treatment and prevention measures.Methods 2007 -2014,The department of General surgery in Qingdao Municipal hospitals for abdominal colorectal cancer resection(Dixon surgery)were 208 cases,including 13 cases of anastomotic leakage occurred.Retrospective summarized the cause,treatment and preventive measures of abdominal colorectal cancer anastomotic fistula.Results All of 13 patients were discharged,and had no adhesions obstruction,anastomotic stricture,pelvic abscess and other symptoms following -up 6 months after discharge.Conclusion Grasping preopera-tive risk factors of abdominal colorectal cancer for anastomotic fistula,actively preoperative preparation,and master of fine anatomical and surgical point are the main points of the prevention of anastomotic leakage.
3.Assessing with the MMPI-2 for Stress Disorders

Objective: To investigate psychopathology and diagnostic efficiency with MMPI-2 in patients with stress disorder (SD). Methods: MMPI-2s were administered to twenty-nine patients with acute stress disorder (ASD), fourteen patients with posttraumatic stress disorder (PTSD) and forty-four well-adjusted trauma survivors. Results: Patients with ASD obtained significantly higher scores on F, Fb, Fp, Pa, Pt, Sc and Ma and lower on K, S than controls. PTSD patients scored significantly higher on F, Fb, Fp, Hs, D, Hy, Pa, Pt and Sc and lower on S than controls. There were significant differences in scores of Hs, D and Hy between patients with ASD and with PTSD. M678 code type was found as the mean profile for SD patients. Discriminate analysis indicated that Pa and Sc were the most important variables and it successfully classified 83.9% and 72.0% respectively in training sample and cross-validation sample. Conclusion: Patients with SD and trauma survivors show significantly different psychopathology and there were also differences between patients with ASD and with PTSD. MMPI-2 scales especially Pa and Sc were sensitive to posttraumatic stress symptoms.

5.Effect of ligustrazine on cerebral injury in LPS-induced septic shock rats

AIM: To investigate the effect of ligustrazine (Lig) on cerebral injury in LPS-induced septic shock rats and to explore the underlying mechanism.METHODS: Wistar rats (n =48) were randomly divided into control group, LPS group and LPS +Lig treatment group.The rats in LPS group were randomly divided into 2 subgroups at time points of 6 h and 12 h.After ligustrazine treatment, the venous blood was collected by removal of eyeballs to detect the con-centration of neuron-specific enolase (NSE) using ELISA.The nitric oxide (NO) concentration in the homogenate of brain tissues was examined.The apoptosis in the hippocampus was analyzed by TUNEL staining.The protein expression of Bax and Bcl-2 was determined by Western blot.RESULTS: Ligustrazine inhibited the elevation of NSE and NO concentrations in LPS-induced septic shock rats.Furthermore, ligustrazine administration also attenuated LPS-induced increase in Bax ex-pression and decrease in Bcl-2 expression.CONCLUSION: Ligustrazine decreases the concentration of NSE and NO, and attenuates cerebral injury in LPS-induced septic shock rats.These effects may be related to the regulation of Bax and Bcl-2 expression.
6.Toxic effects of manganese ion eyedrops to ocular anterior segment

Background A preliminary study determined an application of manganese ion eyedrops in tracing observation of visual system in vivo,but whether manganese ion has toxic effects on ocular anterior segment tissues is still unclear.Objective This study was to investigate the toxic effects of different concentrations of manganese ion eyedrops on ocular anterior segment tissues.Methods Thirty-two clean New-Zealand white rabbits were divided into 1.00 mol/L MnCl2 group,0.50 mol/L MnCl2 group,0.25 mol/L MnCl2 group and normal control group according to random number table.The experimental eye was randomly chosen from each rabbit for eyedrops application.The manganese ion eyedrops at concentration of 1.00 mol/L,0.50 mol/L or 0.25 mol/L was topically administered in any lateral eye of each rabbit in the 1.00 mol/L MnCl2 group,0.50 mol/L MnCl2 group,0.25 mol/L MnCl2 group,and normal saline solution was used in the same way in the normal control group.In 2 hours,1 day and 3,7,14,21,28 days after eyedrops application,the irritative response of ocular surface was examined under the slit lamp microscope,and corneal damage was evaluated by corneal fluorescence staining,and corneal endothelial cell density (ECD) and corneal thickness were assessed using corneal specular microscope.The eyeball samples were prepared in 3,7,14 and 28 days after eyedrops application and hematoxylin and eosin staining was performed for histopathological examination of conjunctiva,cornea,trabecular meshwork and ciliary body tissue.The use and care of experimental animals complied with the Statement of ARVO.Results Severe hyperemia and edema in palpebral conjunctiva appeared in the 1.00 mol/L MnCl2 group and 0.50 mol/L MnCl2 group,and the symptoms reached a maximum level 1 day after eyedrop application and disappeared 7 days after eyedrops application in the 1.00 mol/L MnCl2 group.These symptoms were milder in the 0.50 mol/L MnCl2 group in comparison with the 1.00 mol/L MnCl2 group.There were no ocular irritative response in the 0.25 mol/L MnCl2 group.Significant differences were found in the corneal fluorescence scores among the groups 2 hours,1 day,3 and 7 days after eyedrop application (X2 =17.350,19.200,12.200,10.140,all at P<0.05),with the highest score in the 1.00 mol/L MnCl2 group.The ECD was not significantly different among the groups and different time points (Fgroup =0.38,P>0.05;Ftime =1.79,P>0.05).The corneal thickness value was significantly different among the groups and different time points (Fgroup =18.22,P<0.05;Ftime =116.75,P < 0.05).The corneal thickness value was elevated during 2 hours to 3 days after administration of eyedrops in the 1.00 mol/L MnCl2 group and closed to normal from 7 days to 28 days.The corneal thickness value was higher from 2 hours to 14 days than that in 28 days after administration of eyedrops in the 1.00 moL/L MnCl2 group,and the corneal thickness value was higher in 2 hours to 1 day than that in 28 days after administration of eyedrops in the 0.50 mol/L MnCl2 group (all at P<0.05).Inflammatory cell infiltration and stromal edema were exhibited in conjunctival,trabecular meshwork and ciliary body tissue in the 1.00 mol/L MnCl2 group 3 days and 7 days after eyedrops application,and only inflammatory cell infiltration in conjunctival tissue in the 0.50 mol/L MnCl2 group 3 days after eyedrops application.No obvious abnormity was seen in the 0.25 mol/L MnCl2 group.Conclusions 1.00 mol/L manganese ion eyedrops has a remarkable toxic effect on ocular anterior segment tissue in rabbit.With the lowing of drug concentrations,the toxic effects were reduced.The manganese-enhanced MRI using 0.25 mol/L manganese ion eyedrops is feasibility bacause of its non-toxicity to ocular tissues.
7.Investigation on heart rate turbulence in patients with decreased left ventricular diastolic function

Objective To assess the characteristics of heart rate turbulencer (HRT) in elderly patients with decreased left ventricular diastolie function.Methods 40 patients were divided into two groups:20 patients with enlarged left atrium and 20 patients without enlarged left atrium.20 healthy people were selected as controls. Turbu1ence onset (TO) and turbulence slope (TS) were measured,and correlation was analyzed between TO,TS and the E/A,index of Macruz and heart rate variability (HRV).Results TO was higher (P<0.05) and TS was lower in patients (P<0.01),TO was higber in patients with enlarged left atrium than in with out enlarged lef atrium people (P<0.05) and TS was lower (P<0.01).TO was positively (P<0.01) and TS Was negatively (P<0.05),correlated with the index of Macruz.TO Was negatively (P<0.05) and TS was positively (P<0.05) related to SDNN and SDANN.Conclusion HRT can be used as a potential risk predictor in decreased left ventricular diastolic function patients.
8.Degradation kinetics of chlorogenic acid, cryptochlorogenic acid, and neochlorogenic acid at neutral and alkaline pH values.

The degradation kinetics of chlorogenic acid (5-CQA), cryptochlorogenic acid (4-CQA), and neochlorogenic acid (3-CQA) in aqueous solution at 37 degrees C and different pH values (7.05, 7.96, 9.25) were investigated in the present work. The results indicated that 3-, 4- and 5-CQA tended to remain stable in acidic pH circumstance, and unstable in neutral and alkaline pH circumstance. With the increase of the alkalinity, the degradation of 3-, 4- and 5-CQA was increased leading to a less amount of total CQA and was satisfactorily described by the Weibull equation. Meanwhile, caffeic acid was not detected after the degradation of CQA. Moreover, the degradation of 3-CQA and 5-CQA tended to be converted to 4-CQA, and the degradation of 4-CQA tended to be converted to 3-CQA rather than 5-CQA. The comparison of the degradation kinetics parameters of 3-, 4- and 5-CQA at neutral and alkaline pH values showed that the orders of the rate constant (k) values were 4-CQA > 3-CQA > 5-CQA, while the orders of the degradation half life (t½) values were 4-CQA < 3-CQA < 5-CQA, indicating the orders of the stabilities of 3-, 4- and 5-CQA at 37 degrees C and neutral and alkaline pH values were 4-CQA < 3-CQA < 5-CQA.

9.Role of PI3K/Akt signal pathway in BMSCs migration induced by serum of rats with severely burn

Objective To study the effects of severely burned rats serum on migration of BMSCs and mechanism.Methods Severely burned rats model was established,and the preparation of severely burned rats serum.Experimental groups:normal train-ing group(containing 10% fetal bovine serum,group C),burn serum group(containing 10% burns in the rat serum,group B),burn serum+blockers(10% burns in the rat serum+final concentration of 10 μmol/L PI3K signaling pathway inhibitor LY294002 train-ing,group B+LY).Activity of cells was examined with MTT;migration of cells was examined with Transwell chambers testing;protein expression of p-AKT/AKT was determined with Western blot;microtubule structure of cells was examined with immuno-fluorescence.Results Compared with group C,group B burn serum treatment after 24 h,BMSCs activity(P <0.01),p-AKT levels (P <0.05),increased migration quantity(P <0.001);cell microtubule structures appear rupture,after adding inhibitor,compared with group B,group B+LY BMSCs activity(P <0.01),to reduce the number of migration(P <0.001),p-lower AKT(P <0.05), cell microtubule structure similar to the normal group.Conclusion Severely burned rats serum can promote BMSCs migration,may burn serum cytokine activation of PI3K/AKT signal pathway,resulting in cell microtubule structure change,promote the migration of BMSCs.
10.Expression of COX-2 and VEGF-C in papillary thyroid carcinoma and their relationship to cervical lymph metastases

Objective:To investigate the expression of cyclooxygenase-2 (COX-2) and vascular endothelial growth factor-C (VEGF-C) in papillary thyroid carcinoma and their relationship to cervical lymph metastases.Method:In this study, the expressions of COX-2 and VEGF-C were examined by immunohistochemistry in papillary thyroid carcinoma tissues of 40 patients, and analysis was performed on the correlation of cervical lymph metastases with COX-2 and VEGF-C expression.Result:Positive expressions of COX-2 and VEGF-C were 70.0%(28/40)and 75.0%(30/40)respectively in papillary thyroid carcinoma. The positive rates of COX-2 and VEGF-C expression were 80.8%(21/26)and 84.6%(22/26)respectively in patients with cervical lymph metastases, and 50.0%(7/14)and 57.1%(8/14)respectively in patients without cervical lymph metastases, with a statistically significant difference between two groups(P<0.05, all). COX-2 was positively correlated to VEGF-C expression in papillary thyroid carcinoma(r=0.378, P<0.05).Conclusion:The results suggest that COX-2 and VEGF-C were highly expressed in papillary thyroid carcinoma, with possible interaction of their expressions, and may play a critical role in the cervical lymph metastases of papillary thyroid carcinoma.
