2.Effect of Combined Rehabilitative Training on Stroke in Convalescent Stage

ObjectiveTo observe the effect of combined rehabilitative training on stroke patients in convalescent stage.Methods52 stroke patients with hemiplegia were divided into rehabilitation group (n=26, receiving combined rehabilitative training) and control group (n=26, not receiving combined rehabilitative training). Scores of functional independence measure (FIM) and Fugl-Meyer assessment (FMA) of all patients were assessed when admission and discharge.ResultsFor patients of rehabilitation group, FMA scores when discharge was not significant difference compared with admission, but FIM scores was significantly higher than that of control group (P<0.01).ConclusionCombined rehabilitative training can improve function and living quality of stroke patients in convalescent stage.

4.The effect on leukocytes in peripheral blood and C-reactive protein levels after eradication of Helicobacter pylori

Objective To investigate the systemic effects of Helicobacter pylori (Hp) infection by comparing differential counts of leukocytes in peripheral blood and C-reactive protein (CRP) levels before and after eradication of Hp. Method A total of 158 Hp-positive patients underwent eradication by standard one-week triple therapy were divided into Hp eradicated group(n = 108) and Hp non-eradicated group(n = 50),the populations of peripheral blood leukocytes and CRP levels before and 0 (just after therapy),1 and 12 months after eradication were retrospectively analysed. Results In the eradicated group,blood leukecytes,neutrophils and monocytes decreased significantly after eradication,and the CRP levels were significantly lower than the levels before treatment (P < 0.05 ),but there was no significant change in eosinophils,basophils and lymphecytes (P > 0.05).In the non-eradicated group,there was no significant change in any studied pa-rameter (P > 0.05). Conclusion These findings suggest that Hp infection increase blood leukocytes,neu-trophil and monocyte counts in the peripheral blood and CRP levels,which indicates a significant role of Hp infection in systemic disorders.
5.Up-regulation of miR-15b and miR-16and inhibition of VEGF expression in HLF cells after hyperoxia explosure

Objective To investigate the effect of miR-15b and miR-16 on the expression of vascular endothelial growth factor (VEGF) protein in human embryonic lung fibroblast (HLF) cells under hyperoxia.Methods The expression level of miR-15b and miR-16 was up-regulated and down-regulated in HLF cells by transfection technology, respectively.The expression of VEGF protein in HLF cells was assessed by Western blot.Furthermore, under hyperoxia exposure in vitro, the expression of miR-15b, miR-16 and VEGF protein in HLF cells was analyzed.Results Up-regulation of miR-15b and miR-16 suppressed VEGF protein expression, while down-regulated miR-15b and miR-16 promoted VEGF protein expression.In addition, hyperoxia exposure induced up-regulation of miR-15b and miR-16, but down-regulation of VEGF protein in HLF cells.Conclusions Hyperoxia exposure may up-regulate the expression level of miR-15b and miR-16, but suppress VEGF protein expression.These may contribute to the development of bronchopulmonary dysplasia.
6.EXPRESSION OF GDNF,GFAP AND NGFRp75 BY OLFACTORY ENSHEATHING CELLS IN ADULT RAT AND GOLDEN HAMSTER

Objective To investigate the expression of GDNF in the olfactory ensheathing glia cells of the adult rats and golden hamster for studying the mechanisms of the OECs in the CNS regeneration. Methods The immunohistochemical staining was performed on the slides of adult olfactory bulb from 3 rats and 3 golden hamsters.According to the primary antibody,tissue slides were divided into 3 groups:rabbit polyclonal GDNF,the rabbit polyclonal GFAP and rabbit polyclonal NGFRp75(Santa Cruz Biotechnology)respectively,and followed by the rabbit ABC immunoreactive staining system. Results It revealed that GDNF was expressed by cells in the first two lines of olfactory bulb from rat and golden hamster.The GFAP and NGFRp75 were also expressed in the same position from other two groups of slides respectively.Conclusion The olfactory ensheathing cell could secret GDNF in the progress of mediating neuronal survival and axonal elongation.
7.Effect of Therapy Combination with Rehabilitation Approach and Acupuncture on Cerebral Trauma during Recovery Stage

Objective To observe the effect of therapy combined with rehabilitation approach and acupuncture on cerebral trauma during the recovery stage. Methods 48 cases were divided into two proups: 24 cases in treatment group who applied the rehabilitation combined with acupuncture, the other 24 cases in control group who applied acupuncture alone. They were evaluated with Functional Independent Measure (FIM) 3 months after treatment. Results The scores of FIM were significantly difference between these two groups (P<0.05). Conclusion The therapy combined with rehabilitation approach and acupuncture can improve the recovery of cerebral trauma.
8.Correlation of synovial fluid and articular cartilage osteopontin with disease severity in knee osteoarthritis

Objective To examine osteopontin (OPN) levels in both synovial fluid and articular cartilage of patients with primary knee osteoarthritis (OA) and to investigate their relationship with severity of the disease. Methods Fifty patients with knee OA and 10 healthy controls were enrolled in this study.There were 15 males and 35 females with an average age of 61.8±7.4 years in OA group. The control group included 4 males and 6 females with an average age of 63.2±6.0 years. Mankin score were taken to determine the disease severity of the affected knee. The radiographic grading of OA in the knee was performed using the Kellgren-Lawrence criteria. OPN levels in synovial fluid were measured using enzyme-linked immunosorbent assay. OPN levels in articular cartilage were assessed by immunohistochemical methods. Results Compared to healthy controls, the knee OA patients had higher OPN concentration in synovial fluid ([4519.60±1830.37] pg/ml vs. [1179.70±303.39] pg/ml) and articular cartilage([0.60±0.06] vs. [0.43 ±0.07]). In addition, synovial fluid OPN levels showed a positive correlation with articular cartilage OPN levels (r=0.411,P=0.003). Subsequent analysis showed that the OPN levels in synovial fluid significantly had been correlated with severity of disease using Kellgren-Lawrence criteria (r=0.581, P< 0.001). Furthermore,the levels of OPN in the articular cartilage also were correlated with disease severity using Mankin score (r=0.675, P< 0.001).Conclusion The data suggest that OPN in synovial fluid and articular cartilage is related to progressive joint damage and could be a predictive biomarker respect to disease severity and progression in knee OA.
9.Peroxisome proliferator activated receptor gamma ligand suppresses hepatic fibrosis in rats

Objective To study the effect of peroxisome proliferator-activated receptor garama (PPARγ) ligand on hepatic fibrosis in rats. Methods Forty Wistar rats were randomly divided into two groups: the control group(20 rats) ,in which liver cirrhosis was induced by adding 0. 3‰ thioacetamide in the fodder for 6 months, and rosiglitazone group(20 rats) in which 200 ppm of rosiglitazone in combination with 0. 3‰ thioacetamide was added in the fodder. Liver tissue's mRNA expression of PPARγ, TGF-β1 ,type Ⅰ pro-collagen and α-smooth muscle actin(α-SMA) was detected by RT-PCR. The protein expression of PPARγ, TGF-β1 ,type Ⅰ collagen and α-SMA was detected by Western blot. The expression of collagenof liver histological section was evaluated by Van Gieson (VG) staining. Results The expression ofPPARγ at mRNA level significantly increased in rosiglitazone group compared with those in the control group ( t = 6. 93, P < 0. 01 ), while the expression of TGF-β1 ( t = 3. 89, P < 0. 01 ) and type Ⅰ pro-collagen ( t =5.67,P <0. 01 ) were lower than that in the control group. The protein expression of PPARγ, TGF-β1 and type Ⅰ collagen was in similar tendence with that of mRNA expression. The expression of α-SMA decreased significantly in rosiglitazone group compared with that in the control group (t = 3. 12,P < 0.01 ). The collagen stainings of liver histological section in rosiglitazone group was lower than those in the control group (t = 3.47, P < 0. 01 ). Conclusion PPARγ ligand inhibits the production of collagen in fibrofic livers in rats and prevents hepatic fibrosis in vivo.
10.THE STUDY OF DEGRADATION OF ANTHRACENE,PHENANTHRENE AND PYRENE BY EFFECTIVE FLAVOBACTERIUM

Two effective flavobacterium strains FCN1 and FCN2 were isolated from the sludge which had been contaminated by coke-plant waste water for a long time.Before isolation,the naphthalene was used to cultivate such sludge for seven weeks as the sole carbon source and its concentration adding to the cultivating system was increased up to 50mg/L gradually.The polycyclic aromatic hydrocarbon degrading characteristics of our isolates themselves and with inorganic ions was studied respectively.The testing results show that our two isolates can transform and degrade anthracene,phenanthrene and pyrene.After 10 hours reaction,the removing rates of anthracene,phenanthrene and pyrene by FCN1 were 84%,69%,80% and by FCN2 were 76%,40%,71% respectively.After 106 hours,the removing rates of TOC caused by anthracene,phenanthrene and pyrene were 70%,54%,69% by FCN1 and 63%,50%,46% by FCN2.The Fe 3+ and Mg 2+ can accelerate the degrading reaction of FCN1.
