1.Isolation and culturing of highly purified glandular epithelial cells from human endometrial specimens

Objective: To develop an improved method for the primary isolation and culture of highly purified endometrial glandular epithelial cells in vitro.Methods: Normal endometrial tissues were isolated and cultured in vitro by double collagenase digestion and selective wall-pasting.Results: Highly purified endometrial epithelial cells were successfully isolated and cultured.Conclusion: Epithelial and interstitial cells of the endometrium can be successfully isolated with high purity by double collagenase digestion and selective wall-pasting.
2.Evaluation of posterior chamber implantable collamer lens with a central hole for myopic correction

Objective To evaluate the effectiveness,safety,predictability and stability of a new posterior chamber implantable collamer lens with central hole to correct myopia.Methods 111 eyes of 58 patients were evaluated,including uncorrected (UCVA) and best corrected (BCVA) visual acuities,spherical equivalent(SE),cylindrical equivalent(CE),contrast sensitivity,intraocular pressure evaluated preoperatively.75 eyes of 41 patients were implanted with ICL V4c(ICL group) while 36 eyes of 21 patients were implanted with Toric ICL V4c(TICL group).Postoperative observation and adverse events were evaluated over 3 months follow-up.Results 3 months after surgery,the mean UCVA in ICL group was 1.0 or better in 58.67%.The UCVA of 61 eyes (95.31%) was more than BCVA before operation.The mean SE was (-0.26±1.16) D,66.67% of 50 eyes were within ±0.50 D.The safety and efficacy indices were 1.37 and 1.31,respectively.The mean UCVA in TICL group was 1.0 or better in 86.11%.The UCVA of 32 eyes (96.97%) was more than BCVA before operation.The mean SE was (0.08±0.84) D,63.89% of 23 eyes were within ±0.50 D.And the mean CE was (-0.34±0.54) D,66.67% of 24 eyes were within±0.50D.The safety and efficacy indices were 1.32 and 1.30,respectively.The postoperative contrast sensitivity values in two groups were found significantly higher than those of the preoperative eyes.The IOP remained stable over time.Conclusion Implantation of the posterior chamber ICL with a central hole is effective,safe,predictable,and stable for the correction of high myopia.
3.Therapeutic trauma-induced repigmentation of vitiligo lesions: mechanisms and relevant clinical studies

It has long been noted that dermabrasion, plum-blossom needling or micro-needling, and even ablative fractional CO 2 laser can effectively induce repigmentation of vitiligo lesions resistant to conventional ultraviolet B phototherapy. In addition to trauma-induced increase in the transdermal absorption of drugs, these treatments also initiate wound repair and activate melanocytes in hair follicles or epidermis. Basal layer keratinocytes, vascular endothelial cells and fibroblasts in the wound margins can secrete chemokine CXCL12 to recruit melanocytes or melanocyte stem cells expressing chemokine receptors CXCR4/CXCR7 in the hair follicle bulge or around the skin lesions to move towards the vitiliginous area. This review summarizes progress in repigmentation of vitiligo lesions induced by therapeutic skin trauma.

5.Expression of the hBD-2 in human airway epithelium induced by IL-1? in vitro and its molecular mechanism

Objective To investigate induction of the expression of human ?-defensin-2 (hBD-2) mRNA and the changes in activation of I-?B? and NF-?B by IL-1? in the human airway epithelium. Methods Primary human respiratory tract epithelium were stimulated with IL-1? and and/or PDTC. The expression of hBD-2 mRNA was detected by reverse transcription polymerase chain reaction (RT-PCR). The I-?B? protein level in the cytoplasm was determined by Western blot, and the nuclear factor-kappaB (NF-?B) binding activity was analyzed by electrophoretic mobility shift assays. Results The hBD-2 mRNA expression could be detected after 1.5 hours of IL-1? stimulation at concentration of 1?g/L, and the expression of HBD-2 was in a dose dependent manner. Stimulation with IL-1? resulted in a reduction of I-?B? protein levels. The supershifts assays indicated that the p65-p50 heterodimer complexes of NF-?B were involved in the activation of NF-?B. Conclusions I-?B? can induce the expression of HBD-2 mRNA in a dose dependent manner; the p65-p50 heterodimer complexes of NF-?B play an important role in the regulation of hBD-2 gene expression in response to IL-1?.
6.On shear-induced platelet aggregation and its therapy

It is now widely accepted that platelet aggregation plays an important role in physiological hemostasis and pathological thrombosis associated with cardiovascular and cerebrovascular diseases. Anti-platelet aggregation drug research is also a hot spot of current research. The biggest challenge of antiplatelet therapy has been the molecular overlap of the hemostasis and thrombosis, leading to a serious risk of bleeding. Recent studies have emphasized the importance of shear stress generated from blood flow, which will primarily drive platelet activation and aggregation in thrombosis. So if we can take advantage of the differences between the physiological and pathological vascular blood flow environment, the development of selective anti-platelet therapy may be a safer treatment for cardiovascular and cerebrovascular diseases. In this review, we discuss the underlying mechanisms of shear-induced platelet activation. Later, we summarize the effects and mechanisms of compounds and traditional Chinese medicine on shear-induced platelet activation. The aim is to provide a reference for the study of biological pharmacology of traditional Chinese medicine for promoting blood circulation and removing blood stasis.
7.Early bile duct to bile duct end-to-end(side) anastomosis for bile duct injuries: an analysis of 25 patients

Objective To study the use of early bile duct to bile duct end-to-end(side) anastomosis for extrahepatic bile duct injuries.Methods The clinical data of 25 patients with extrahepatic bile duct injuries with early bile duct to bile duct end-to-end(side) anastomosis were retrospectively analyzed.Results Bile duct injuries happened in laparoscopic cholecystectomy (n=17),laparoscopic exploration of common bile duct (n=1),laparotomy (n=5),and knife wound (n=2).All of the repairs were successfully carried out.Except for one patient who was lost to follow-up,the other patients were followed for over 2 years.There was no bile duct stenosis.One patient died after 5 months of repair in other hospital because of a recurrent bile duct calculus.Conclusions Early bile duct to bile duct end-to-end(side) anastomosis had a good long-term results.The operation needs to be done by a good surgeon or supervised by an experienced specialist in biliary surgery.A prolonged period of T tube stenting is a prerequisite for success.
8.Study on Antioxidant Activity of Extract of Cassia Obtusifolia

Objective To study antioxidant activity of extract of Cassia obtusifolia. Method The free radical-scavenging effect of the extract was evaluated in the flavins system; erythrocyte membrane was exposed to three radicals generation systems, the inhibition of the extract on lipid peroxidation was determined. Results The extract showed strong scavenging effect on superoxide radical and dose- dependently inhibited the increases of lipid peroxidation content in the membrane exposed to xanthine-xanthine oxidase system, H2O2 or UV light. Conclusion Antioxidant activity of the extract was obvious.

Objective To study of effects Medlar flavone on NO and NOS in H 2 O2 damaged vascular endothelial cells.Meth-ods Human Umbilical Vein Endothelial Cells(HUVEC)have been set as research object.HUVEC cells was damaged by H 2 O2 in-duced oxidative stress.Experiments were divided into normal control group,H 2 O2 injury group,VitC control group,and low,medi-um,high flavonoids protective groups(100,200,400 mg/L).MTT method was used to observe cells activity.The effects Medlar fla-vone on MDA、TNOS、iNOS and NO in H 2 O2 damaged vascular endothelial cells was observed.Results (1)MTT result demonstra-ted the IR of HUVEC cells was 38.8% in H 2 O2 injury group.Which in low,medium,high flavonoids protective groups were 34.0%,30.7%,25.5%.All were lower than H 2 O2 injury group(P <0.01).(2)Compared with normal control group,NOS activity was decreased in H 2 O2 injury cell,while TNOS,iNOS,NO was more.Compared H 2 O2 group,the change of NO,TNOS,iNOS, MDA was opposite in flavone intervention group,and which were dose dependent (P <0.01 ).Conclusion Medlar flavone has a protective effect on H 2 O2 induced HUVEC cells injury,and there may be some kind of relevance between them.
