1.Effect ofβ-defensin 2 gene silencing on the inflammatory response induced by Haemophilus influenzae type B combined vaccine immunization in rats

Objective To investigate the effect of β‐defensin 2 (rBD‐2 ) silencing on the inflammatory response induced by Haemophilus influenzae type B vaccine immunization in rats .Methods A total of 144 SPF Sprague Dawley (SD) rats were divided into four groups .Group 1 were immunized with Haemophilus influenzae type B vaccine .Group 2 were immunized with Haemophilus influenzae type B vaccine and administered with lentivirus Lv‐shRNA‐rBD‐2 by intratracheal instillation .Group 3 were administered with the lentivirus Lv‐shRNA‐rBD‐2 by intratracheal instillation .Group 4 were blank controls .Each group was set with 12 rats . The serum was collected at 12 ,24 and 72 hours after immunization ,and tumor necrosis factor (TNF)‐α,interleukine (IL)‐1β and IL‐10 were assessed by enzyme‐linked immuno sorbent assay (ELISA ) .Expression levels of rBD‐2 in lung tissues of rats were measured by western blotting . Results The recombinant virus was successfully produced by a lipofectmaine transfection method .At 12 ,24 ,and 72 h of immunization ,serum levels of TNF‐αof rats in group 1 were (82 .0 ± 8 .0) ,(155 .0 ± 18 .2) ,and (272 .0 ± 32 .5) pg/mL ,respectively ,which were all higher than those in group 4 ([55 .0 ± 6 .2] ,[52 .0 ± 5 .8] ,and [56 .0 ± 4 .8] pg/mL ,respectively ;t=16 .034 ,P=0 .043 ;t=12 .411 ,P=0 .035 ;t=10 .530 ,P=0 .018 ,respectively) .Serum levels of TNF‐αat 12 ,24 ,and 72 h of immunization in group 2 were (66 .0 ± 8 .2) ,(90 .0 ± 12 .6) ,and (108 .0 ± 13 .6) pg/mL ,respectively ,which were all significantly lower than group 1 (t=12 .115 ,P=0 .039 ;t=12 .830 , P=0 .033 ;t=15 .522 ,P=0 .012 ,respectively) .At each time point ,serum levels of IL‐1βand IL‐10 in group 1 were all significantly higher than those in group 4 (IL‐1β:t=18 .032 , P=0 .048 ;t=15 .824 , P=0 .039 ;t=13 .518 , P=0 .021 ,respectively ;IL‐10 :t=15 .410 , P=0 .045 ;t=14 .294 , P=0 .032 ;t=13 .375 ,P=0 .013 ,respectively) .Serum levels of IL‐1βand IL‐10 at each time points in group 2 were all significantly lower than those in group 1 (IL‐1β:t=19 .012 , P=0 .043;t=16 .991 , P=0 .034;t=14 .862 , P=0 .027 ,respectively ;IL‐10 :t=15 .134 , P=0 .048 ;t=15 .264 , P=0 .036 ;t=11 .408 , P=0 .024 ,respectively) .Seventy‐two hours after immunization of SD rats ,the relative content of rBD‐2 protein in lung tissue of rats significantly increased .Protein level in group 1 was significantly higher than that in group 4 (t=10 .582 ,P=0 .035) ,while protein level in group 2 was significantly lower than that in group 1 (t=13 .250 ,P=0 .027) .Conclusions rBD‐2 gene silencing can improve the inflammatory response induced by Haemophilus inf luenzae type B combined vaccine in rats.
2.CLINICAL STUDY OF ESSENTIAL HYPERTENSION WITH DRUG INTERVENTION

133 patients with mild to moderate essential hypertension were treated with capt opril, nifedipine, propranolol and hydrochlorothiazide respectively. The effects of these drugs on plasma renin activity(PRA),hemodynamic parameters and response to antihypertensive drugs were reported.It was shown that nifedipine is the best drug for decreasing blood pressure and improving hemodynamic parameters. Effectiveness in the group with nifedipine was 93.62%, it was more effective than captopril group(74.29%), propranolol group(47. 37%)and hydrochlorothiazide group(87.5%).Our study have not shown any correlation between antihypertensive effect and PRA levels in nifedipine group.Our data indicated that nifedipine is generallly effective and well tolerated, causing a significant lowering of blood pressure without severe side effects. It may be recommended as a Firststep antihypertensive agent for patients with mild to moderate hypertension
3.The application of polypropylene mesh & plug in hernia repair.

Objective To approach the advantages and summarize the experience of mesh & plug tension-free hernia repair. MethodsThe mesh & plug tension-free hernia repair was administered in 102 patients of groin hernia with the BardR Mesh & PerfixR plug offered by Bard Company in U. S. A. The operation time,postoperative pain,patient's ability recovery, complications and recurrence rate were followed up. ResultsThe average operation time was 31.5minutes. The patients could move 4~6 hours after operation. Postoperative pain lasted about 2~3 days. One had retention of urine and two had wound subcutaneous hydrops. No wound infection. Nine had feeling of foreign matter in the groin region. Only one had recurrence in 89 patients who were followed-up. ConclusionBardR Mesh & PerfixR Plug has good tissue-compatibility,no rejection,and capability of anti-infection. It is an ideal material for hernia repair. The mesh & plug tension-free hernia repair is easy to be administered, with minor lesion, quicker recovery and lower recurrence. And the operation indication can be widened. It is a most advanced method in treating groin hernia.
4.Effect of Celecoxib on the Expression of Cyclooxygenase-2 in Atherosclerotic Tissues in Rabbits

OBJECTIVE: To observe the expression of cyclooxygenase-2(COX-2) in atherosclerotic tissue in rabbits and to discuss the association between COX-2 and atherosclerosis (AS) and evaluate the effect of cyclooxygenase-2(COX-2) inhibitor celecoxib (cele) on atherosclerosis progression in atherosclerotic model rabbits. METHODS: Eighteen New Zealand rabbits were randomly divided into three groups: normal control group, high cholesterol (HC) group, and celecoxib(20 mg?kg?d-1) group. After feeding for 14 weeks, the aortic atherosclerotic lesions and the ratio of aortic intima to media in rabbits were measured. RT-PCR, immunohistochemistry and Western Blot were used for the detection of expression of COX-2 mRNA and protein. 5.Comparative study of fluorescent distribution within the rat cochlea after three viral vectors inoculation in round window membrane

OBJECTIVE To determine the most effective viral vector in infecting the spiral ganglion cells and cochlear nerve fibers among three viral vectors that was inoculated via round window membrane. METHODS Three types vectors carrying GFP gene including Lentivirus, Adenovirus-5 and Adenoassociated virus-2 were infused into the perilymphatic space through the round window membrane with micro injector in rat respectively. The distribution and intensity of green fluorescence were estimated by fluorescent microscope at 3days, 7days and 14days after gene transfer. RESULTS Multiple types of cochlear cells were infected by three different viral vectors. In contrast to the other vectors, the adenovirus-5 vector effectively transfected spiral ganglion cells and cochlea nerve fibers with high expression level for 2 weeks. CONCLUSION Distribution of GFP gene expression mediated by three kinds of viral vector was different obviously in rat cochlea, only adenovirus-5 successfully targets cochlear spiral ganglion cells. These findings are of value in gene therapy for cochlear spiral ganglion cell degeneration.
6.Construction and expression of mouse-human chimeric anti-human melanoma antibody

Objective:To construct eukaryotic expression vector for anti-human melanoma chimeric antibody,and express the antibody in eukaryotic cells.Methods:Variable regions of the murine antibody were cloned from the anti-human melanoma hybridoma cell line HB8459,HB8760,Mel3,and was inserted them into the chimeric antibody eukaryotic expression vector pMH-CA.The constructed vector was transfected into 293T,the chimeric antibody was expressed,as detected by RT-PCR,ELISA,and Western blot.Results:Eukaryotic expression vectors of the chimeric antibody against human melanoma were successfully constructed,and expressed in eukaryotic cells.The expression of chimeric gene was confirmed by RT-PCR,specificity of the chimeric antibody was proved by ELISA and Western blot.Conclusion:Anti-human melanoma chimeric antibody was expressed successfully.

10. Evolutionary characteristics of polymerase PA, PB1, and PB2 genes of novel influenza virus A/H1N1 in 2009 pandemic

Objective: To elucidate the evolutionary characteristics of polymerase PA, PB1, and PB2 genes of the novel influenza virus A/H1N1 in 2009 pandemic. Methods: The sequences of the PA, PB1, and PB2 genes of the novel H1N1 strains in 2009 pandemic, and the reference sequences of human, swine, and avian influenza viruses isolated during different years at different locations were retrieved from NCBI. Molecular Evolutionary Genetics Analysis version 4.0 (MEGA4.0) software was employed to align and blunt nucleotide sequences, construct phylogenetic tree, deduce and align PB2 protein sequences, and the results were compared between the novel A/H1N1 and each of the reference strains. Results: The sequences of the PA, PB1, and PB2 genes of 2009 novel A/H1N1 strains isolated from different locations shared a high homology and clustered in a unique new clade, and were close to the swine influenza viruses. The PA, PB1, and PB2 genes of the novel H1N1 viruses had a high similarity with the corresponding sequences of a human H1N1 strain isolated in Iowa State of USA in 2005 (A/Iowa/CEID23/2005/H1N1). Alignments of the deduced protein sequences showed that the 627th amino acid of PB2 of the novel H1N1 strains and A/Iowa/CEID23/2005/H1N1 were glutamic acid (Glu), which was the same as that in the avian influenza virus in Iowa State of USA in 2005 (DQ889682), and was different from those of the reference sequences of human A/H1N1 strains isolated from 1918 to 2008, which were lysine (Lys). Conclusion: The 2009 novel A/H1N1 virus might be originated from the human A/H1N1 strains isolated in 2005 in Iowa State of America (A/Iowa/CEID23/2005/H1N1), and the polymerase gene of the novel H1N1 virus might re-assort with avian A influenza virus.
