1.Prokaryotic expression of antibiotic peptide Cecropin A gene and identification of expression products

BACKGROUND: Cecropins are a kind of micromolecule protein with antibacterial activity. Eukaryotic cell-expressed or artificially synthesized Cecropins is characterized by low efficiency and high cost. OBJECTIVE: To clone and express an antibacterial peptide gene of Musca domestica Cecropin A, and to identify recombinant expression product. METHODS: Mature Musca domestica Cecropin A encoding nucleotide sequence was searched from the GenBank and amplified by RT-PCR. The gene of Musca domestica Cecropin A was cloned into prokaryotic expression vector pET32a and fused with gene of Thioredoxin (Trx) and expressed in E.coli BL2l (DE3). After induction by isopropyl-β-D-thiogalactoside, the sera of the immunized rabbits were collected after rabbits were immunized with the hemolymph of housefly larvae. Recombinant protein was identified by western blot analysis and N-[Tris(hydroxymethyl)methyl]glycine-sodium dodecylsulfate-polyacrylamide gel electrophoresis.RESULTS AND CONCLUSION: After induction by isopropyl-β-D-thiogalactoside, E.coli BL21 expressed mature Cecropin. Rabbit anti- housefly larvae sera, N-[Tris(hydroxymethyl)methyl]glycine-sodium dodecylsulfate-polyacrylamide gel electrophoresis and western blot analysis results confirmed that expression products were mature Cecropin. These suggest that prokaryotic expression system can be utilized to obtain natural mature Cecropin.

MicroRNA (miRNA) regulate the expression of target mRNA according to the specific base pairing. They are involved in proliferation,differentiation and apoptosis of cells. The expression of miRNA has distinct differences between normal tissues and cancers, miRNA can be oncogenes but also suppressor genes, they play an important role in cancer. And miRNA are promising to be a new therapeutic target. In this review, we will discuss the advance in the research of miRNA in cancer.
3.Treatment of 30 Cases of Migraine by Mind-regulating Acupuncture

Objective: To observe the clinical effects of needling acupoints Shenting(GV 24),Neiguan(PC 6) and Sanyinjiao(SP 6) in the treatment of migraine. Methods: Thirty cases of Germen patients with migraine were treated mainly by Shenting(GV 24), bilateral Neiguan(PC 6) and Sanyinjiao(SP 6), as well as bilateral Hegu(LI 4) and Taichong(LR 3), Touwei(ST 8),Yangbai(GB 14), Cuanzhu(BL 2), Sizhukong(TE 23), Shuaigu(GB 8) and Sibai(ST 2) in the affected side of head, auricular Sympathetic (MA-AH 7), Er Shenmen (MA-TF 1), Heart (MA-IC) and Endocrine (MA-IC 3); another 28 cases were treated by routine acupuncture. The clinical results and immediate pain-relieving effects were observed. Results: The total effective rate was 90.0% in treatment group and 75.0% in control group, χ2= 4.57, P < 0.05; the 24-hour pain-relieving effects were obviously better in treatment group than that in control group, χ2 = 10.11, P< 0.01. Conclusion: Needling Shenting(GV 24), Neiguan (PC 6) and Sanyinjiao(SP 6) has better effects than routine acupuncture in the treatment of migraine.
4.Investigation on the Freshmen’Status of the Preventive Vaccination Against Hepatitis B in A University of Ankang City in 2 0 0 3~2 0 1 4

Objective To understand the freshmen’s status of hepatitis B virus infection,immunological effect after vaccina-tion against hepatitis B from a university in Ankang city,to provide a scientific basis for university’s prevention from hepati-tis B.Methods Analyzed the results of five markers of hepatitis B virus infection and alanine aminotransferase (ALT)of 25 895 freshmen in a college in Ankang for resent 12 years,and observed the HBsAb serocoversion in serum 1 year after vacci-nation against hepatitis B and after strengthened immunity.Results The HBeAg positive rate of the freshmen was 4.07%, among which the male was 5.58% (281/5 035),and the female was 3.14% (258/8 214).The difference had significance of statistic (P<0.05).The college students’HBeAg positive rate from rural area and from urban area were 4.77% and 2.93%respectively,the difference had significance of statistic (P<0.05).There was no significant statistic difference in immune effect of hepatitis B vaccine for different sex (χ2=5.80,P<0.05).Their HBsAb positive rate was 95.15% after injected with 10μg hepatitis B and 5μg hepatitis B vaccine,which had significance of statistic (P<0.05).Conclusion The hepatitis B virus infection rate and the hepatitis B protective antibody of the college freshmen in Ankang district was lower than that of the whole country and had a decreased tendency.It showed the male’s infection rate of hepatitis B virus was higher than the female,the rural was higher than the urban,and the immune effect of hepatitis B vaccine proved to be good.It should be-strengthened immunity for non-responders to ensure the immune effect if necessary.
5.Effect of Immunogenicity of Neuroblastoma Cell Lines by Transduction of Adenovirus-Mediated Co-Stimulated Factor B7-1 and Granulocyte-Macrophage Colony-Stimulating Factor Genes

ObjectiveTo observe the effect of co-stimulated factor B7-1 and granulocyte-macrophage colony-stimulating factor(GM-CSF) genes mediated by recombinant adenovirus on immunogenicity of neuroblastoma(NB)cell line SH-SY5Y.MethodsAfter NB cell line SH-SY5Y was transfected with recombinant adenovirus mediating green fluorescent protein(Ad-GFP) in different multiplicity of infection(MOI),the transfer efficiency was tested by flow cytometry(FCM).SH-SY5Y cells were transfected separately with recombinant adenovirus,recombinant adenovirus mediating B7-1 and GM-CSF genes or only B7-1 gene,and they were named as Ad,B7-GM and B7 group.The cells which were not transfected were named as SH group.The protein expression of B7-1 and GM-CSF were detected by using FCM and enzyme linked immunosorbent assay(ELISA).Peripheral mononuclear cells were separated and purified with Ficoll density gradient centrifugation.Cpm and cytokine(IFN-? and IL-2) secretory were evaluated by mixed lymphocyte culturing with different ratio(5 to 1 and 10 to 1) on target cells and effect cells.ResultsTransfer efficiency of recombinant adenovirus to SH-SY5Y was high.After transfection,B7-1 and GM-CSF genes were expressed.Proliferation of lymphocytes was stimulated and cytokine increased vigorously in B7-GM group and B7 group,and the effect in B7-GM group was stronger than that in B7 group.And the effect in ratio 5 to 1 group was stronger than that in 10 to 1 group.ConclusionsB7-1 and GM-CSF genes have synergistic effect.After transfection with recombinant adenovirus mediated B7-1 and GM-CSF genes,the immunogenicity of SH-SY5Y is greatly enhanced.
6.Different surface treatments and bond strength of fiber posts: Differences among sandblasted, hydrogen peroxide and silaned treatments

BACKGROUND: Fiber posts are currently perceived as promising alternatives to cast metal posts, as their elastic modulus are similar to that of dentin, producing a favorable stress distribution and providing more esthetic outcomes for endodontically treated teeth. Failure of a fiber post and composite resin core often occurs at the junction between the two materials. This failure process requires better characterization.OBJECTIVE: To compare the bond strength of fiber posts after three surface treatments to a composite resin. METHODS: The coronal portions of 20 mandibular premolars with a single root canal were removed and were endodontically treated. After post space preparation, the roots were divided into four groups and the fiber posts and resin cores were lured. In the three testing groups, the surfaces of fiber posts were silaned, sandblasted or etched with hydrogen peroxide. In the control group, the surfaces of fiber posts were not treated. All specimens were exposed to cyclic loadings and thermal cycles. The roots were sectioned into slices with 1 mm thickness and their bond strengths were tested with a universal material testing machine. Test piece following testing was placed under optical microscope to observe the breakage method.RESULTS AND CONCLUSION: Stickiness strength was significantly greater in the sandblasted and hydrogen peroxide groups compared with the control group (P < 0.05). No significant difference in stickiness strength was determined between the sandblasted and hydrogen peroxide groups (P > 0.05). No significant difference in stickiness strength was detected between the silaned and control groups (P > 0.05). The splicing destruction among each group was the main breakage method between materials and fiber posts. The surface treatment of fiber posts with sandblast or hydrogen peroxide significantly enhanced the bond strength of the composite resin tested. The surface treatment of fiber posts with silane did not enhance the bond strength of the composite resin.
7.Simultaneous Determination of 5-Hydroxymethylfurfural and Nine Kinds of Phenolic Compounds in Rice Wine Using High Performance Liquid Chromatography

An analytical method of reversed phase high performance liquid chromatography (RP-HPLC) was developed for simultaneous determination of 5-hydroxymethylfurfural and nine phenolic compounds (including (+)-catechin, (-)-epicatechin, chlorogenic acid, rutin, caffeic acid, protocatechuic acid, syringic acid, ferulic acid and p-coumaric acid) in 20 min.Ten components were detected and separated successfully by Diamonsil C_(18) column (150 mm x4.6 mm, 5 μm) at wavelength of 280 nm and column temperature of 42 ℃, with acetonitrile and 3% acetic acid solution as the mobile phase in gradient elution.The resultant correlation coefficients of the ten compounds were between 0.9911 and 0.9995 with detection limit from 0.2 to 0.5 mg/L, the RSD less than 2.4% and the average recoveries from 89.4% to 98.3%.These experimental results demonstrate that 5-hydroxymethylfurfural and the nine phenolic compounds in different rice wine samples can be determined with the new method for practical uses.

In recent years,nano - materials have represented the superior biocompatibiliry and the arena of medical nano - materials have been extensively studied by various means all over the world. Is it possible that nano - bone implants overcome the existing limitations of the traditional bone implants for fusing operations in the future? State Food and Drug Administration (SFDA, CN) authorized the nano-bone implants- - nHA/ PA66 (YZB/0063 -2003) in 2005 but there were still lots of counterviews in this field. This review considers the advanced researches via comparing and analyzing the correlatire literatures recently.
9.A Clinical Observation of the Effects of In-flow Anesthetic Gas Absorber on Recovery Time

Objective:To observe whether the in-flow anesthetic gas absorber can reduce the recovery time after stopping inhalation of enflurane or isoflurane. Methods: In fixed tidal volume, minute volume and fresh gas flow, the recovery time of isoflurane or enflurane anesthesia with or without in-flow anesthetic gas absorber were compared after closing the vaporizer.Results: With in-flow anesthetic gas absorber, the time for anesthetic gas concentration in circle to reduce the MAC to 0.3was 3.3±0.5 min for isoflurane, which was significntly shorter than that without absorber (20±0.3min) ( P<0.01 ) and the time for enflurane was 3.5±0.5min and 25±0.1min respectively. Conclusion:Anesthetic gas absorber can reduce the recovery time of either enflurane or isoflurane inhalational anesthesia significantly.
10.Advances in detection of circulating tumor cells in non-small cell lung cancer

Lung cancer is the most common cancer worldwide, accounting for approximately 18%of all cancer-related deaths. Circulating tumor cells (CTCs) have the capacity to spread into the blood stream. This capacity of CTCs is one of the most critical fac-tors for unfavorable clinical outcome of patients with NSCLC. Detection of CTCs using a simple blood test may assist in early detec-tion of lung cancer. Researchers have attempted to isolate CTCs from peripheral blood using various techniques. Although reports on CTC detection are abundant, methodological aspects concerning sensitivity, specificity, and reproducibility have prevented a clear ap-praisal of the clinical impact. This paper gives a brief description of each CTC detection method.
