And miRNA are promising to be a new therapeutic target. In this review, we will discuss the advance in the research of miRNA in cancer.
3.Prokaryotic expression of antibiotic peptide Cecropin A gene and identification of expression products
Chinese Journal of Tissue Engineering Research 2011;15(24):4549-4552
BACKGROUND: Cecropins are a kind of micromolecule protein with antibacterial activity. Eukaryotic cell-expressed or artificially synthesized Cecropins is characterized by low efficiency and high cost. OBJECTIVE: To clone and express an antibacterial peptide gene of Musca domestica Cecropin A, and to identify recombinant expression product. METHODS: Mature Musca domestica Cecropin A encoding nucleotide sequence was searched from the GenBank and amplified by RT-PCR. The gene of Musca domestica Cecropin A was cloned into prokaryotic expression vector pET32a and fused with gene of Thioredoxin (Trx) and expressed in E.coli BL2l (DE3). After induction by isopropyl-β-D-thiogalactoside, the sera of the immunized rabbits were collected after rabbits were immunized with the hemolymph of housefly larvae. Recombinant protein was identified by western blot analysis and N-[Tris(hydroxymethyl)methyl]glycine-sodium dodecylsulfate-polyacrylamide gel electrophoresis.RESULTS AND CONCLUSION: After induction by isopropyl-β-D-thiogalactoside, E.coli BL21 expressed mature Cecropin. Rabbit anti- housefly larvae sera, N-[Tris(hydroxymethyl)methyl]glycine-sodium dodecylsulfate-polyacrylamide gel electrophoresis and western blot analysis results confirmed that expression products were mature Cecropin. These suggest that prokaryotic expression system can be utilized to obtain natural mature Cecropin.

5.Perinatal infection and asthma in the infant
International Journal of Pediatrics 2010;37(1):20-22
Epidemiological and clinical studies have provided compelling evidence that suggests a link between the relative lack of infectious diseases and the increase in allergic disorders. Asthma usually starts early in life. The early occurrence of asthma suggests that prinatal influences are of importance.Prenatal or postnatal exposure to endotoxins and other bacterial products may protect against the development of asthma in the infant. We will briefly outline the current state of the relationship of the perinatal infection and asthma in the infant, and focus our review mainly on possible rneehanism of perinatal infection providing protection against asthma in the infant, such as Th1/Th2 immune system, transcription factors, protection by maternal antibodies, endotoxin tolerance, pulmonary inflammation and genetic influences.
6.The experimental study of recombinant adenovirus-mediated human wild type p53 gene transfection into neuroblastoma cell line
Journal of Chinese Physician 2008;10(11):1475-1477
Objective To observe the effect of transduetion of human wild type gene(wt-p53) mediated by recombinant adenovirus on cell growth, cell cycle and apoptosis of neuroblastoma (NB) cell line SH-SY5Y. Methods The NB cell line SH-SY5Y were transfected by recombinant adenovirus with green fluorescent protein(Ad-GFP). The suitable muhiplieity of infection (MOI) was determined after trans-fer efficiency was tested by flow cytometry (FCM). Both recombinant adenovirus(Ad) and recombinant adenovirus mediating wt-p53 gene (Ad-p53) were transfected into SH-SY5Y cells. SH-SY5Y cells without any transfection were used as blank group. The p53 protein expres-sion was validated by Western blot. Cell number was counted and growth curve was drawn. Both cell cycle and apoptesis were detected by FCM. Results The results indicated that the transfer efficiency of recombinant adenovirus to SH-SY5Y was satisfied, and 100 MOI was suitable. After transfection with Ad-p53, p53 protein was high expressed. Cell growth were inhibited, apoptosis and cell cycle arrest were in-duced in p53 gene transfeetion group. Conclusions Our results indicate that clinical application of the in vitro p53 gone modified tumor cell vaccine might be feasible and effective in clinical treatment of NB.

The two groups both received conventional rehabilitation training. In addition, the treatment group was given tongue acupuncture plus balloon dilatation, and the control group was by balloon dilatation alone. The two groups were treated once a day, for a total of 4 weeks. The water drinking test score and Standardized Swallowing Assessment (SSA) were evaluated before and after the treatment.Result The water drinking test and SSA scores were significantly changed after the intervention in both groups (P<0.05). After the treatment, the water drinking test and SSA scores in the treatment group were significantly different from those in the control group (P<0.05).Conclusion Tongue needling plus balloon dilatation is an effective approach in treating deglutition disorders due to achalasia of the cricopharyngeus muscle after cerebral stroke.
8.Effects of drug-contained sera ofNaoluo Xintong versus Zuogui pill on the proliferation and differentiation ofin vitro cultured neural stem cells
Chinese Journal of Tissue Engineering Research 2016;20(32):4845-4851
BACKGROUND:Because of limited source and a relatively weak ability of differentiation and proliferation, how to take positive and effective measures to promote neural stem cel proliferation, differentiation has become the focus of research.
OBJECTIVE:To investigate the effects of drug-contained sera ofNaoluo Xintong versus Zuoguipil on the proliferation and differentiation ofin vitro cultured rat neural stem cels.
METHODS:Embryonic neural stem cels of Sprague-Dawley rats were isolated and culturedin vitro, and then were co-cultured with the serum medium containing 10%Naoluo Xintong and 10%Zuoguipil, respectively. Comparative observations were performed between two groups by inverted microscope and immunofluorescence staining.
RESULTS AND CONCLUSION:Under the inverted microscope, the cels began to grow in cluster and gather into a bal, but the diameter was relatively smal after 24-hour culture; the neurospheres expended further, with relatively regular shape, but no neurosphere differentiation appeared after 48 hours of culture. The average prominent length of the neurospheres in theZuoguipil group was significantly greater than that in theNaoluo Xintong group after 5 days of culture (P < 0.05). The rate of rat neural stem cels differentiating into MAP-2-positive cels in theZuoguipil group was significantly lower than that in the Naoluo Xintong group (P < 0.05), but the rate of differentiated cels positive for glial fibrilary acidic protein in theZuoguipil group was significantly higher than that in theNaoluo Xintong group (P < 0.05). After 48 hours of culture, neurospheres were cultured in the different drug-contained media, and 12 hours later, the neurospheres adhered to the wal, and a smal amount of cel migration occurred. Then, cel migration began to increase with time. Under the immunofluorescence staining: prominent neurons with long protrusions were increased in both two groups, but there were no significant differences in the proportion of neurons and astrocytes between two groups (P > 0.05). These findings suggest that drug-contained sera ofNaoluo Xintong andZuoguipil can both not only promote the proliferation and differentiation ofin vitro cultured rat neural stem cels, but also provide a suitable microenvironment for neural cel proliferation. Additionaly, there are significant differences between the two drugs. Consequently, it is feasible to induce neural cel proliferation and differentiation byNaoluo Xintong andZuoguipil.
