3. Quantitative study on medicinal properties of traditional Chinese medicine based on BP neural network
Chinese Traditional and Herbal Drugs 2020;51(16):4277-4283
Objective: It is difficult to accurately grasp the essential characteristics of medicinal properties of traditional Chinese medicine due to the abstraction and vagueness. This paper proposes a Quantitative Model of Traditional Chinese Medicine's Properties based on BP Neural Network (QM-BP Model) to train and realize quantitative representations of Chinese herbal medicine (CHM). Methods: Data for analysis were obtained and organized by conceptual analysis. Sample pairs of the associations were obtained based on the relationships of CHM and their efficacy. Then a QM-BP model with three-tier structure in form of CHM-drug vector-efficacy was constructed, initialized and trained according to prior organized CHM data. Finally, rules of correlation of CHM and their efficacy was obtained by training dataset with drug vectors representing quantitative attributes of CHM. Results: Based on the training of QM-BP model, 474 TCM and 528 effects included in the textbook of TCM were trained and combined based on the training of QM-BP model. It was found that the BP drug vectors representing drug properties after training reflected the attribute characteristics of CHM better than the initial quantitative values. In addition, as BP drug vector and word vector have similar properties, the BP drug vectors for CHM with similar efficacy was relatively close in Euclidean distance while the CHM with different efficacies were relatively far in Euclidean distance. Conclusion: In this paper, a BP neural network was adopted to construct a medicine vector training model. Based on the correlation between the medicinal properties and efficacy of TCM, the quantified values of the medicinal properties were modified to represent medicinal properties more accurately. In future work, the QM-BP model can be applied to the analysis of herb pairs and prescriptions to analyze the rules of combination related to medicinal properties and the compatibility within prescriptions.

Microbiology 2008;0(08):-
In the experiment, the production of plagues by Bdellovibrio bacteria in solid medium cultivation, the reproduction of Bdellovibrio bacteria in liquid medium cultivation and the attachment of Bdellovibrio bacteria to carrier were observed, which aimed to study the effects of enrofloxacin on the growth and at-tachment ability of Bdellovibrio bacteria BDF-H16. Results indicated that in solid medium cultivation, the production of plagues by Bdellovibrio bacteria BDF-H16 was inhibited by different concentrations (2 ?g/mL, 5 ?g/mL, 10 ?g/mL, 20 ?g/mL, 50 ?g/mL) of enrofloxacin and the inhibitory effects of enrofloxacin became stronger with the increase of the concentration of enrofloxacin. Similarly, in liquid medium cultivation, the reproduction of Bdellovibrio bacteria BDF-H16 was also obviously inhibited by different concentrations ofenrofloxacin and higher concentrations of enrofloxacin such as 10 ?g/mL, 20 ?g/mL, 50 ?g/mL had stronger inhibitory effects on the reproduction of BDF-H16. However, the growth tendency of Bdellovibrio bacteria BDF-H16 was not inhibited in 10 ?g/mL enrofloxacin. Additionally, when zeolite was added, enrofloxacin had also inhibitory effects on the numbers of Bdellovibrio bacteria BDF-H16 attached to zeolite. With the increase of the concentrations of enrofloxacin, the numbers of Bdellovibrio bacteria BDF-H16 attached to zeolite became smaller and smaller. However, the attachment rate of Bdellovibrio bacteria BDF-H16 to zeo-lite became higher under 2 ?g/mL-20 ?g/mL enrofloxacin. The results above showed that enrofloxacin had inhibitory effects on the plague production and reproduction of Bdellovibrio bacteria BDF-H16, but the at-tachment ability of Bdellovibrio bacteria BDF-H16 was strengthened in liquid medium cultivation with 2 ?g/mL-20 ?g/mL enrofloxacin and zeolite, and adding zeolite helped to reduce the adverse effects of en-rofloxacin on Bdellovibrio bacteria BDF-H16.

AIM: To explore the mutual effect of co-culture of wild-type astrocytes (ASC) and motor neurons VSC4.1 (VSC) on the respective ability to produce reactive oxygen species (ROS). METHODS: The inhibition rates of cell growth in ASC and VSC in co-culture or independent culture was detected after exposed to excitatory stimulus by MTT method. Real-time observation of ROS production by ASC and VSC labeled with Hoechst 33342 was detected by confocal microscopy under the conditions of co-culture or independent culture. RESULTS: Higher concentration of glutamate induced a higher inhibition rate in mixed cell growth than that in ASC alone, while lower concentration of glutamate induced a higher inhibition rate in mixed cell growth than that in VSC only. Real-time observation by confocal microscopy showed that ROS production by VSC under the condition of co-culture, which showed a notable increase at 15 min, was significant less than that in independent culture, which peaked at 5 min and was gradually decreased. ROS production by ASC in co-culture began to increase significantly at 10 min. CONCLUSION: Compared to independent culture, ASC reduces the resting ROS production by co-cultured with VSC, while ASC prolongs the duration of ROS production by VSC after exposed to excitatory stimulus.

Based on Staphylococcus aureus 16S rRNA gene sequences, sequence comparisons have been applied to design a kind of stem-loop structured oligonucleotide probes whose loop sequence mismatched other bacterial strains 16S rRNA gene sequences in more than two positions with high specificity and sensitivity.According to the principle of molecular beacon technology and enzyme linked immunosorbent assay,a method has been evaluated using immobilized stem-loop structured oligonucleotides probe as the conformation switch which is applied to enzymatic detection of nucleic acid targets. As its specificity has been strengthened, the system is able to successfully eliminate false-positive result that is mismatch in an oligonucleotide.Employing a microtiter assay format,4ng of S.aureus 16S rRNA could be detected at least. This approach is more sensitive than other conventional method at least one order of magnitude.
