1.STUDY ON THE DETECTION OF TOXOPLASMACIRCULATING ANTIGENS USING IMMUNO-PCR

Aim to explore an assay for the early diagnosis of Toxoplasma infection. Methods Free streptavidin was used to attach a biotinylated DNA to the biotinylated second antibody, through amplication of the DNA label, antigen which bound with antibodies was detected, thus Toxoplasma circulating antigens Immuno-PCR assay was estabished. detecting the CAg in serial dilutions of serum and the dynamic variation of serum CAg of Toxoplasma in experimental infected mice using Immuno-PCR and ELISA parallely to compare the sensitivity Result The detection limit of the immuno-PCR was at about 1/1000 dilution, it was at 1/5 dilution for ELISA. CAg of Toxoplasma could be detected as early as the third day after infection using Immuno - pcr, Wheareas the ELISA on the same samples, revealed no detectable CAg elevations until fifth day after infection. Conclusion In comparison with CAgdetecting by ELISA parallelly performed, this method is more sensitive than ELISA, The detection limit of the immuno-PCR was an 200 fold improvement compared with a conventional ELISA. the positive result could be detected earlier than that of ELISA,It provide scientific basis for early diagnosis of Toxoplasmosis.
2.In Vivo Kinetics and Biodistribution of a Hantaan Virus DNA Vaccine after Intramuscular Injection in Mice

To study the kinetics in vivo of a Hantaan virus DNA vaccine, we constructed a fusion DNA vaccine,pEGFP/S, by cloning the S segment of Hantavirus into the vector, pEGFP-C1, which encodes Green fluorescent protein EGFP. In this report, we provide evidence that pEGFP/S was distributed and persistently expressed for more than 60 days in several organs after inoculation. Our findings suggest that the persistent immune responses induced by a Hantaan virus DNA vaccine are likely due to the plasmid pEGFP/S deposited in vivo, which acts as a booster immunization.
3.Identification on pheretima by X-Ray diffraction Fourier pattern method

Objective: To develop a new identification and analysis method of crude drug Pheretima . Method: Powders of pheretima were identified by means of X ray differacion Fourier pattern. Results: Experiments and analysis were carried out on five samples of pheretima. The standard X ray diffraction Fourier pattern and characteristic diffraction peaks of Pheretima were obtained.Conclusion: This method can be used for identification on crude drug Pheretima .
4.The expression of S-100 protein, CD1a, CD83 and Ki-67 in oral Langerhans cell histiocytosis.

OBJECTIVETo study clinicopathological features, diagnosis, differential diagnosis of oral Langerhans cell histiocytosis (LCH), retrospective clinicopathologic study was carried on and a variety of immune phenotype were detected.
METHODSThe clinicopathological features of 29 cases of oral LCH were analyzed. The immunohistochemical staining of S-100 protein, CD1a, CD83 and Ki-67 were used in above cases by immunohistochemical streptavidin-biotin peroxidase (SP) and Elivison two-step method. Statistical analysis was adopted for the results.
RESULTSOf the 29 cases of LCH, the expression of S-100 protein and CD1a were positive in 24 cases and negative in 5 cases, so 5 cases were excluded from the diagnosis of LCH. Among 24 cases of LCH, 15 patients were male and 9 were female. The median age was 7.50 years. 14 lesions were in the mandible, 5 were in the maxilla and 5 involved the mandible and maxilla. 9 cases were in stage I, 13 in stage II and 2 in stage III, according to Bartnick classification. Immunohistochemistry showed all 24 cases staining for S-100 protein and CD1a were positive. Comparing with maxillofacial lesions involved soft tissue, Ki-67 positive rate was lower and CD83 positive rate was higher in maxillofacial single bone lesion.
CONCLUSIONThe immunohistochemical staining of S-100 protein and CD1a are important for the diagnosis of LCH. Maxillofacial bone single LCH might have lower proliferative activity and a higher state of maturity. Maxillofacial LCH involved soft tissue might have a higher proliferative activity and a lower state of maturity.

5.Mucinous adenocarcinoma of salivary gland: a case report.

6.Research and development of remote training system for radiotherapy CT simulation based on virtual reality technology

Objective:To develop a remote training system for CT simulation positioning of radiotherapy using virtual reality technology, and to explore a new method of medical training.Methods:The 3DMax and Maya were employed to establish the 3D model. The unity3D engine was adopted to develop 3D virtual operation and interaction system. Java spring MVC architecture was utilized as the system background service. MySQL was used as the background database system. The users were assigned into two roles: teacher and student, and the modes were divided into teaching and assessment modes.Results:The function of the system covered the whole process of CT simulation positioning, mainly including modules of patient information management, CT simulation positioning machine cognition, body position fixation technology, CT positioning scanning, and emergency handling, etc. Since it was put into use in 2018, the system has been running stably, with 14 920 pages views and an 86.66% pass rate. Compared with the traditional training, the training efficiency has been significantly improved and has received unanimous recognition.Conclusions:The remote training system can effectively improve the clinical practice ability and humanistic care ability of the trainees, which has good autonomy, sharing, and innovation. At present, the system has been put online and has strong popularization with prospects for broad application.
7.The remote training system for quality assurance of medical electronic linear accelerators based on extended reality technology

Objective:To improve the quality assurance (QA) skills of radiotherapy personnel and medical students and reduce the radiation risk of training by developing a remote training system for QA of medical electronic linear accelerators.Methods:This training system was built based on radiotherapy technology and quality control contents of medical electronic linear accelerators, and a virtual reality interactive software was developed using extended reality (XR) technology Unity 3D. A remote control module of multi-terminal platform was also developed. A multi-perspective evaluation system was adopted and a questionnaire was designed to analyze the application value of this system.Results:The training system reproduced the live environment and physical objects of medical electronic linear accelerator treatment room. It built a multi-terminal virtual simulation training system with radiotherapy technology as well as QA knowledge system. This system could provide 5G remote control of medical electronic linear accelerator for off-site quality control demonstration and guidance. By March 1, 2022, a total number of 133 people had been trained using this system, 76 valid questionnaires had been taken, of which 90.79% (69/76) of the respondents trusted the experimental results shown by the system and 88.16% (67/76) of the respondents considered the training system necessary.Conclusions:The training effect of this system is widely recognized. It fundamentally reduces the training radiation hazard and provides reference for the reform and progress of QA training mode of medical electronic linear accelerators.
