1.Qualitative research on pressure source of caregivers in need of hematopoietic stem cell transplantation

Objective To investigate the pressure sources of caregivers in need of hematopoietic stem cell transplantation (HSCT) and to provide evidence for targeted intervention. Methods According to Hill′s ABC-X family pressure model, 12 main family caregivers were received semi-structured in depth interviews with phenomenological methods. Results Pressure sources of caregivers for HSCT patients were summed up to 7 aspects: search of donor, isolation with patient during transplantation, lack of knowledge in patient care, family resource difficulties, family economic overburden, social role transformation and uncertainty of disease prognosis. Conclusions The pressure sources of caregivers for HSCT patients is a persistent process, which is over layed as the patient′s condition worsens. The analysis on the pressure sources of primary family caregivers is becoming a new topic in HSCT nursing area.
2.EFFECT OF Se ON Na~+ ,K~+-ATPase AND 5'-NUCLEOTIDASE ACTIVITIES OF CARDIAC SARCOLEMMA IN RATS

Sprague—dawey rats were fed for one to three months with Se—deficient grains from Keshan disease area, Se—supplemented diet and stock diet,respectively. Changes of Na~+ ,K~+ ATPase and 5'—nucleotidase activities in cardiac sarcolemma ,Se Contents in liver and plasma and GSH—px activities in Liver and red blood cells of rats were observed. The results showed that Na~+ ,K~+—ATPase. and 5'—nucleotidase activities in cardiac sarcolemma in Se-deficient group rats were significantly decreased,hepatic and plasma Se contents as well as hepatic and erythrocyte GSH—px activities were also significantly decreased at of feeding 30,60 and 90 d as compared to Se—supplemented or stock diet group. There was no significant difference in these parameters between Se—supplemented or stock diet groups There was no significant difference in these paramefers between Se—supplemented and stock diet groups。
3.Clinical Observation of Acertil in the Treatment60Cases of Stroke Sequel

OBJECTIVE:To observe the therapeutic effect of Acertil on stroke sequel.METHODS:The liver function,kidney function,ion,blood sugar and blood pressure of enrolled patients were examined at given time periods.RESULTS:Dis?ability degree0~1∶Acertil group14cases(49.99%),control group8cases(30.67%);Disability degree3~4∶Acertil group1case(3.5%),control group5cases(19.22%).CONCLUSION:Acertil can abate the disability degree of stroke sequel patients.
4.Cordycepin negatively modulates LPS-induced cytokine production by induction of Heme Oxygenase-1

Objective To investigate the mocelualr mechanism of Cordycepin negative modulates LPSinduced cytokine production in murine macrophages.Methods The RAW264.7 murine macrophages were cultured in vitro and were pre-treated by different concentration of Cordycepin,and then stimulated by LPS for 8 h.Production of TNF-o,IL-6 and IL-12,and the content of p65 in the nuclear were detected by ELISA.Expression of heme oxygenase-1 (HO-1) and phosphorylation of IκB and p38 were measured by Western blot.Nuclear translocation of Nrf2 was detected by Immunofluorescence.Results 1 ~ 30 μg/mL of Cordycepin treatment significantly abrogated LPS-induced TNF-α,IL-6 and IL-12 production,p65 nuclear translocation and IκB phosphorylation.In addition,different concentration of Cordycepin could also induce RAW264.7 cells expression of HO-1,phosphorylation of p38 and nuclear translocation of Nrf2.Application of p38 inhibitor and small interfering RNA-mediated knock-down of Nrf-2 significantly inhibited surfactin-induced HO-1 expression.Treatment with a selective inhibitor of HO-1 reversed the Cordycepin mediated inhibition of pro-inflammatory cytokines.Conclusions Cordycepin induces antiinflammatory effects by inhibition of NF-κB and activation of Nrf-2 and p38 mediated HO-1 induction.
5.Three-dimensional reconstruction technique in breast volume measurement and its artificial influencing factors

Objective To set up a method of three-dimensional breast volume measurement, to observe the effect of the artificial influencing factors on the final data, which are used for 3-D reconstruction of breast volume. Methods The CT data were used to reconstruct a digital prototype of the breast implant and the chest. The 3-D models were established firstly, and then the model was reasonablly cut to get single breast volume, breast implant volume, differences of the both sides of chest and breast volume. These methods were put up by the repeated test when the reference frame and the ranges were fixed. And the analysis of the artificial influencing factors was performed on 10 women whose breasts were unsym-metrical. The effects of the artificial influencing factors were analyzed by moving the coordinate axis, revolving the chest around the coordinate axis and changing the selected area. Results The repeatable test showed that the error was less than 4. 08 ml. The average volume differences between both sides of the breast before and after the given influencing factors were set, were 10. 339 ml and 7. 73 ml, as moved ± 4mm along the axis X; as revolving ±2° along the axis Z, that were 8. 654 ml and 7. 971 ml; as increasing or decreasing of the selected diameter of ± 4 mm, that were 6. 764 ml and 4. 267 ml, respectively. Conclusion Using the 3-D reconstruction technique for breast volume measurement is more simple, accurate and repeatable.
6.Application of 3.0T Dynamic Contrast-enhanced MRI in the Grading of Gliomas

Objective To explore the clinical significance of 3.0T dynamic?contrast enhanced MRI scan in the grading of intracranial glioma. Methods The magnetic resonance examination were performed in 40 cases of patients of brain tumors confirmed by surgery pathology(29 pa?tients with glioma),including conventional MRI and dynamic contrast?enhanced MRI. Using Kinetic Modeling?version 3.0 software on the GE 3.0T magnetic resonance workstation calculation of intracranial tumor parenchyma area corresponding quantitative parameters Ktrans and Ve values. The quantitative parameters between any two classification were compared ,and the difference was statistically analyzed. The characteristics of the different level of intracranial glioma's dynamic enhanced scan parameters were preliminary analyzed. The receiver?operating characteristic curve (ROC)analysis of Ktrans value and Ve value was performed,and the diagnosed threshold,sensitivity and specificity were acquired. Results While applying dynamic?contrast enhanced MRI scan acquired Ktrans and Ve values ,both values of high grade gliomas include gradeⅢandⅣwere significantly higher than that of low grade gliomas include gradeⅠandⅡ(P<0.05);there was no statistically difference of the parameters of Ktrans and Ve values between gradesⅠwithⅡand gradeⅢwithⅣ(P>0.05). To identify low grade gliomas with high grade glioma ,Ktrans and Ve diagnosis threshold was 0.204/min and 0.099 respectively. To distinguish between gradeⅡandⅢglioma,Ktrans and Ve diagnosis threshold was 0.247/min and 0.176 respectively. Conclusion Combining quantitative parameters Ktrans and Ve value that come from 3.0T dynamic con?trast enhanced MRI scan with regular enhancement MRI can distinguish low grade gliomas with high grade gliomas glioma ,as well as distinguish gradeⅡwithⅢglioma;However,it is still difficult to identify low grade gliomaⅠwithⅡ,as well as high grade gliomaⅢwithⅣ. The Ktrans and Ve value plays an important role in discriminate different grade intracranial tumors in a preoperative noninvasive way.
7.Effect of ferulic acid on the proliferation of, as well as melanin synthesis, tyrosinase activity and expressions of c-kit and ERK proteins in keratinocytes

Objective To evaluate the in vitro effect of ferulic acid on the proliferation of,as well as melanin synthesis,tyrosinase activity and expressions of c-kit and ERK proteins in cultured normal human epidermal melanocytes.Methods Cultured normal human epidermal melanocytes were treated with various concentrations of ferulic acid for different durations,and those remaining untreated served as the control.Then,3-(4,5-dimethylthiazol2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium,inner salt (MTS) assay was performed to estimate cell proliferative activity at 24,48 and 72 hours,sodium hydroxide solubilization method to quantify melanin content in melanocytes at 72 hours,dopa oxidation assay to evaluate tyrosinase activity at 72 hours,Western blot to measure the expressions of c-kit and ERK1/2 proteins at 72 hours.Results Cellular proliferative activity was significantly inhibited in melanocytes treated with ferulic acid of 0.01,0.1 and 1 mg/ml for 24,48 and 72 hours compared with untreated melanocytes (all P < 0.05),and the 72-hour treatment with ferulic acid of 1 mg/ml showed the strongest inhibitory effect.Ferulic acid at 0.01,0.1 and 1 mg/ml all markedly suppressed melanin synthesis and tyrosinase activity,decreased the expressions of c-kit and ERK1/2 proteins in melanocytes,with significant differences in these parameters between ferulic acid-treated and untreated melanocytes (all P < 0.05).Conclusions Ferulic acid could downregulate the proliferation of,as well as melanin synthesis,tyrosinase activity,and expressions of c-kit and ERK proteins in cultured human epidermal melanocytes.
8.Introduction of dynamic balancing randomization method and its application.

Following the development of clinical trials, the methodology of clinical trials has developed a great deal, including randomization method and blinding method, etc. The method of randomization includes simple randomization, stratification randomization, etc. Dynamic randomization is also considered as a method of randomization. Two dynamic randomization methods are introduced in this paper, including a method considering the balance of numbers in each group and a method considering prognostic factors. This paper also demonstrates the procedure of dynamic randomization of the above two methods by simulations and actual examples.
9.Applying contact-mode argon plasma coagulation for retroflex colonoscopic treatment of ultra-lower rectal polyps

Objective To explore the clinical values of applying contact-mode argon plasma coagulation (APC) for retroflex colonoscopic treatment of ultra-lower rectal polyps. Methods 46 wide outsole and applanate polyps smaller than 1.0 cm located at ultra-lower rectum in 17 cases of patients were treated by contact-mode APC under U-type retroflex colonoscopy after failure in regular colonoscopic treatment. Then observe the contacting rate of APC probe with polyps, success rate of curing polyps, rate of probe being adhered and blocked by the solidification structures, incidence of submucosal emphysemas, and incidence of colonoscope ambustion. Results Under U-type retroflex colonoscopy, the probe could contact with polyps in 17 patients. The polyps in every patient were cured by APC in the first time of colonoscopic treatment, whereas all 46 polyps were cured by 97 times of APC spurt. Mild adhesions occurred between the probe and solidification structures at 5 times (5.15 %) among 97 times of APC spurt, without injuries to the coagulation surface from which when the probe separated. The solidification structures blocked the probe only twice (2.06 %). When the solidification structures were cleared, efficiency of the probe restored. No sub-mucosal emphysemas and colonoscope ambustion happened. Conclusion Applying contact-mode APC for retroflex colonoscopic treatment of ultra-lower rectal polyps is safe and effective, it can prevent the damage of colonoscope from the argon knife.
10.A priliminary study on targeting effect of scFv25 fusing to TNFα against hepatocellular carcinoma

Aim To obtain genetically engineered cytokine with potentialities of clinical application. Methods Anti-human hepatocellular carcinoma (HCC) single chain gene fragment(scFv25) was linked with human TNFα gene to form a fusion gene of anti--HCC cytokine, then it was subcloned into prokaryotic GST fusion expression vector pGEX 4T-1 and expressed in the host E.coli. Indirect immunofluorescent staining was performed on HCC cell smearing slides in order to evaluate the activity of purified aim protein, then initial tumor regression trial was carried out in nude mice bearing HCC to evaluate the targeting therapeutic value of scFv25-TNFα . Results scFv25-TNFα had similar specificity as parental antibody HAb25 for SMMC-7721 antigen. The tumor regression trial to the 2 mm HCC xenografts in nude mice showed that scFv25-TNFα had certain targeting cytotoxicity capacity and the efficiency was 3/3 with complete remission. The cytotoxicity of scFv25-TNFα was better than that of TNFα , whose efficiency was only 2/3 and no complete remission was seen. Conclusion scFv25-TNFα is the anti-HCC cytokine, which have certain potentialities of clinical application.
