1.The distribution of platelet glycoprotein I b? variable number tande repeat polymorphism and the relationship with cerebral infarction

Objective To study the distribution of variable number tande repeat(VNTR) polymorphisms of the platelet membrane glycoprotein Ⅰ b? in Han nationality at Harbin and the relationship between these polymorpbisms and cerebral infarction(CI).Methods The identification of alleles and genotypes of VNTR polymorphism of the glycoprotein Ⅰ b? gene was performed by polymerase chain reaction (PCR)in 200 healthy individuls and 200 CI patients(77 lacunar infarction patients and 123 atherosclerotic thrombotic infarction patients),to analyze The relationship between gene polymorphisms and cerebral infarction.Results(1)There were three types of alleles:B、C、D,and five types genotypes:BC,BD,CC, CD,DD in Harbin Han nationality.No person with A allele and BB genotype was found.(2)No statistically significant differences of GP Ⅰ b? gene VNTR polymorphism was found between CI patients or subtype CI patients and controls(P=0.412 and 0.572,respectively).Conclusions(1)This study indicates that the C and D alleles of VNTR polymorphisms of GP Ⅰ b? are the main alleles while the CC and CD genotypes are the main genotypes in Harbin Han people.(2)Our findings indicate that no association exists between the VNTR polymorphism of platelet GP Ⅰ b? gene and CI.
2.Complications analysis on vitrectomy combined with different tampnoades for diabetic vitreous hemorrhage

?AlM:To evaluate and observe the efficacy of silicon oil ( SO ) , perfluoropropane ( C3 F8 ) and balanced salt solution ( BSS ) that can be used as tamponade during vitrectomy to treat proliferative diabetic retinopathy ( PDR) complicated with vitreous hemorrhage ( VH) .
?METHODS: Studied retrospectively on 74 eyes of 60 patients who underwent vitrectomy surgery with diabetic vitreous hemorrhage in our hospital during June 2008 and June 2014. Based on repeated prior examines on fundus details and the vitrectomy tamponades were chosen. All the patients had been followed up at least 3mo. Depending on different tamponades, the paitents were nonrandomized in three groups and contrasted as visual acuity, intraocular pressure ( lOP ) and complications respectively.
?RESULTS: There was statistically significant difference among these three groups in preoperative eyesight ( P<0. 05 ). Moreover, the preoperative eyesight was statistically different between SO and BSS (P<0. 05), and difference for the rest being not remarkable(P>0. 05). The difference being statistically difference in the postoperative vision among these three groups ( P <0. 05). The further analysis showed that the paired-comparisons were statistically significant difference between SO and BSS ( P<0. 05 ), while the rest two groups of comparison were non-respectively(P>0. 05). The preoperative visual function was in contrast to the postoperative (P<0. 05). The lOP before surgery was not statistically significant difference(P>0. 05). However,the difference among three groups being statistically in the postoperative vision(P<0. 05), in addition,the difference existed in each group through pairwise comparison ( P<0. 05) . The occurence rate of complications after surgery in this survey was 47%, the SO group was 50%, the C3 F8 was 56%, the BSS group was 44%.
? CONCLUSlON: Vitrectomy is a safe and effective treatment that can help patients who have diabetic vitreous hemorrhage obtain better visual improvement. Because of the physicochemical properties and different conditions, there still has complications after surgery.
3.Analysis and evaluation of nutritive elements in aerial part of Panax notoginseng.

OBJECTIVETo make full use of the plant resources of Panax notoginseng, nutritional compositions and mineral elements were analyzed in aerial part of P. notoginseng from different areas in Yunnan.
METHODUsing the national standard method, water, ash, crude fat, crude fiber, crude protein and mineral elements were determined in aerial part of P. notoginseng from different growing areas.
RESULTResults showed that there were higher contents of crude fiber and crude protein, and lower content of crude fat in the stems and flowers of P. notoginseng. Meanwhile, a large number of mineral elements were determined in two locations of P. notoginseng, and the contents of Zn, Fe, Mn, Ca and Mg were obvious higher among these mineral elements.
CONCLUSIONThis study showed that the stems and flowers of P. notoginseng were nutritious and suggested that the aerial part may be utilized as new resources foods.

4.Effect of different local anesthetics on analgesia after harvesting grafts from the scalp in burn patients

Objective To investigate the different effect among ropivacaine,bupivacaine,lidocaine on analgesia after harvesting grafts from the scalp in burn patients. Methods 84 patients who need harvesting grafts from the scalp after burn were divided in 4 groups random-ly(n=21). Patients in group C hypodermically injected with saline 200 mL were control,while patients in group R injected with 0. 05% ropi-vacaine 200 mL,group B with 0. 188% bupivacaine,and group L with 0. 1% lidocaine. Motor activity assessment scale( MAAS) and visual analogue scale(VAS) were made before anesthesia(T0) and 20 min,5 h,10 h after awake of patients. VAS were made focus on head and body in part. Vital signs were also monitored and recorded for assessment of security. Results All patients in 4 groups had passed the period of operation safely. Patients in group R have better VAS than other groups. Conclusion Low concentration ropivacaine hypodermically injec-tion of head is helpful to relieve the pain after harvesting grafts from the scalp.
6.Long Non-coding RNA CCAT1 Sponges miR-454 to Promote Chemoresistance of Ovarian Cancer Cells to Cisplatin by Regulation of Surviving

Purpose:
Colon cancer-associated transcript 1 (CCAT1) was identified as an oncogenic long non-coding RNA (lncRNA) in a variety of cancers. However, there was a lack of understanding of the mechanism by which CCAT1 conferred cisplatin (also known as DDP) resistance in ovarian cancer cells.
Materials and Methods:
Cell viability of A2780, SKOV3, A2780/DDP, and SKOV3/DDP cells upon cisplatin treatment was monitored by MTT assay. Quantitative reverse transcription polymerase chain reaction (qRT-PCR) detected the expression levels of CCAT1 and miR-454. The effect of sh-CCAT1 on cisplatin response was investigated in xenografts study. Bioinformatic analysis, luciferase reporter assay and qRT-PCR were conducted to validate the direct interaction among CCAT1, miR-454, and survivin. Apoptosis was determined by flow cytometry after dual staining of Annexin-V-FITC/propidium iodide, and the expression of apoptosis-related proteins Bcl-2, Bax and survivin were detected by qRT-PCR and Western blotting. Xenograft study was conducted to monitor in vivo tumor formation.
Results:
CCAT1 was highly expressed in cisplatin-resistant ovarian cancer cell line A2780/DDP and SKOV3/DDP. Knockdown of CCAT1 restored sensitivity to cisplatin in vitro and in vivo. Our data revealed that silencing of CCAT1 promoted cisplatin-induced apoptosis via modulating the expression of pro- or anti-apoptotic proteins Bax, Bcl-2, and survivin. CCAT1 directly interacted with miR-454, and miR-454 overexpression potentiated cisplatin-induced apoptosis. Survivin was identified as a functional target of miR-454, restoration of survivin attenuated the effect of miR-454 on cisplatin response. In addition, miR-454 inhibitor or overexpression of survivin was found to abolish sh-CCAT1–induced apoptosis upon cisplatin treatment.
Conclusion
CCAT1/miR-454/survivin axis conferred cisplatin resistance in ovarian cancer cells.
7.Research on the growth references of height, weight and body mass index for children under 7 years old

Objective To understand the varying features and the trend of length/height-for-age,weight-for-age and body masa index(BMI)-for-age of children under six years old in Ningbo city,Zhejiang province.Methods By random clustered sampling,64 038 healthy children under six years old in Ningbo were chosen and their length/height and weight measured.Means,standard deviation and percentiles of length/height-for-age,weight-for-age and BMI were calculated by SAS systems.Results of this study were compared with growth standards from WHO 2006 and nine urban areas of China,in 2005.Results Data from our study showed that the annual growth rates were basically the same in boys and girls.All these indicators increased along with age but the younger the age,the faster the growth rate appeared.There were notable differences seen between genders and urban-rural distribution.BMIs were also varied with age and sex.BMI reached peak between 4 to 6 months,then dropped slowly when age increased.Conclusion The means of length/height-for-age,weight-for-age in Ningbo city had exceeded the WHO standards,and the growth by year under six year-olds in Ningbo was close to the data from nine urban areas of China in 2005.
8.Using three different screening methods to study overweight and simple obesity among children under age 7 in Ningbo city

Objective To investigate the occurrence of simple obesity among children under age 7 in Ningbo and to analyze the data regarding body mass index (BMI) from overweight screening under different standards. Methods Randomized cluster sampling was adopted and 64 038 children under 7 years old in Ningho were chosen and their length/height and weight were measured but pathological and secondary obesity cases were excluded. According to the Standardized Height and Weight set by WHO, more than 10% referring to overweight and more than 20% referring to obesity. Moreover, results of this study were compared with growth standards with WHO' s 2006 and IOTF standards. Results According to the Standardized Height and Weight set by WHO, the prevalence rates of overweight and obesity of children under age 7 were 4.25% and 2.88%. The ratio for boys were 4.45% and 3.01%, while for girls as 3.86% and 2.56%, with significant difference (P<0.01). Data showed that the rates for overweight and obesity increased along with age, reaching 11.84% and 9.68% for boys and 10.14% and 9.46% for girls at the age of 6. The ratios of overweight/obesity among different age groups and sex were 1.15-1.94:1. The critical value for P85, P95 were lower than the standards set by WHO and IOTE The rates for overweight and obesity for latter were 9.72%, 2.83% and 6.11%, 0.55% respectively. Conclusion Early childhood is the key period for obesity prevention, with boys in particular. Prevention and control for childhood obesity should be included in the programs for children' s health. Value of classification standards for the BMI screening of overweight and obesity for children under age 7 should be established in no time.
9.Transgenic mouse models of the truncated platelet integrin β3 cytoplasmic tail established by stem cell transplantation.

This study was purpose to establish the transgenic mouse models of the truncated platelet integrin β3 by retrovirus-infected hematopoietic stem cells (HSCs) transplantation and to provide the basis for further study of the role of integrin β3 cytoplasmic domain in platelet bi-directional signaling pathways. Wild-type β3, β3-Δ759 (R(760) GT(762) truncated β3) and β3-Δ754 (T(755) NITYRGT(762) truncated β3) cDNAs were subcloned into MSCV MigR1 retroviral vector bearing a GFP gene and packaged into infective retrovirus with BOSC23 cell strain. The bone marrow HSCs of the β3 deficient mice were infected by the retroviruses, and transplanted into lethally-irradiated wild type C57BL/6 mice. GFP positive rate and surface β3 expression of the recipients' platelets at 6 to 8 weeks after transplantation were detected by flow cytometry to evaluate the transgenic efficiency. The results showed that four kinds of transgenic mouse models including vector, wild-type β3, β3-Δ759 and β3-Δ754 were established successfully. GFP positive rates of transgenic mouse platelets ranged from 18% to 66% and the β3 expression of transgenic mouse reached heterozygote (β3(+/-) level of mouse). It is concluded that establishment of transgenic mouse models mediated by retrovirus-infected HSCs transplantation is a feasible, fast, and high throughput transgenic approach and laid a solid foundation for further research on the role of integrin β3 cytoplasmic domain for bi-directional signaling of platelets in vivo, and for the gene therapy of platelet disorders.

10.The cytology mechanism of anti-parainfluenza virus infection of total flavone of Scutellaria barbata.

In order to research into the cytology mechanism of anti-virus action of total flavone of Scutellaria barbata (TFSB), the effects of TFSB on host cells membrane potential, Na(+)-K(+)-ATPase activity and membrane fluidity after parainfluenza virus type1 (PIV-1) infection were studied. The changes of membrane potential which was fluorescent labeled with DiBAC4(3) and its changes were measured by flow cytometer. Phosphorus determination method and spectrophotometry were used to measure the Na(+)-K(+)-ATPase activity of Hep-2 cells membrane after PIV-1 infection. Hep-2 cells membrane phospholipids were fluorescent labeled with NBD-C6-HPC and membrane fluidity was measured by confocal scanning laser microscope. The result demonstrated that post PIV-1 infection membrane potential decreased significantly and the membrane was in a state of hyperpolarization, Na(+)-K(+)-ATPase activity increased significantly and membrane fluidity decreased significantly. There was no apparent interfere effect of TFSB on the changes of membrane potential and Na(+)-K(+)-ATPase activity after PIV-1 infection, while membrane fluidity improved significantly. It was indicated that the cytology mechanism of PIV-1 infection might be related to membrane hyperpolarization, Na(+)-K(+)-ATPase activity increase and membrane fluidity decrease. TFSB can improve membrane fluidity and prevent the infection by protecting the cell membrane. But it is possible that the anti-PIV-1 mechanisms of TFSB had nothing to do with membrane potential and Na(+)-K(+)-ATPase activity.
