1.The virulence and the gene regulatory mechanism of SpA
Journal of Medical Postgraduates 2003;0(04):-
SpA,one of the important virulence factors of Staphylococcus aureus, can impede phagocytosis, induce apoptosis of B cells, initiate staphylococcal pneumonia and endocarditis,et al.The expression of Spa gene is regulated by a very subtle and complicated systematic web. With the mechanism of the virulence and the gene regulation of Spa were known more and more clearly,New methods of the infection by Staphylococcus aureus were would be found.
2.Ethical Issues in Geneticaly Modified Organisms
Chinese Medical Ethics 1996;0(01):-
Genetically modified organisms, as a high-tech product, break through the limitation of different organisms and create new organisms through gene implanting from other kinds of organisms. They not only bring great benefits to human beings but also bring some ethical issues including the effect on environment, the effect on human health, and the respect to hatural law. Moreover, this article argues some ethical issues during the process of commercialization and sale including how to maintain the consumers' benefits, the protection on researchers through potent right, the precautionary principle and label issues.
3.Surface marker changes during induced differentiation of bone marrow mesenchymal stem cells into neuronal-like cells
Chinese Journal of Tissue Engineering Research 2010;14(14):2643-2646
BACKGROUND:Adult central nervous system lacks the ability to regenerate,so it is of great significance to find a new source of neural stem cells.OBJECTIVE:To investigate the differences between basic fibroblast growth factor(bFGF)and epidermal growth factor(EGF)in inducing bone marrow mesenchymal stem cells(MSCs)to differentiate into neurons in vitro.METHODS:MSCs were isolated from normal human bone marrow using density gradient centrifugation and cell attachment method.MSCs were plated in 96-well culture plates at a density of 0.25×108/L and cultured with 200 μL DMEM/F12 for 0,1,2,3,4,5,6,and 7 days, with 20 μL MTT(5 mg/mL)in 5 wells at each time point. The supernatant was removed and 100 μL dimethyl sulphoxide was added to each well for 4 additional hours of incubation. In addition, some cells were exposed to bFGF and EGF.Growth curve was determined with MTT method.Telomerase activity were examined by TRAP(PCR)-ELISA.Additionally,the functional differences of the two cytokines were checked by RT-PCR.RESULTS AND CONCLUSION:RT-PCR revealed that nestin, glial fibrillary acidic protein(GFAP)and neurofilament subunit M (NF-M)mRNA were expressed in un-induced MSCs of passage 4.Nestin expression reduced at 7 days. The expression of micro-tubule-associated protein-2(MAP2)mRNA was not detected until the induction, and increased thereafter. The expression of MAP2 mRNA was greater in bFGF+EGF and bFGF alone groups compared with EGF alone group, and the expression of GFAP in EGF alone group was greater than other groups. Results showed that MSCs can be cultivated, proliferated and differentiated into neural stem cells in vitro. The differentiated neural stem cells have the activity of proliferation, but not have the ability of infinite proliferation as tumor cells.
4.Molecular markers in the bone marrow micrometastasis of esophageal cancer
Journal of International Oncology 2013;40(11):839-843
In recent years, the bone marrow micrometastasis of esophageal cancer has become the research focus. Many studies show that epithelial cell molecules, angiogenesis markers, chemokine receptor-4 (CXCR-4),HER2,activated leukocyte cell adhesion molecule (ALCAM) and stanniocalcin-1 (STC-1) are closely related to the bone marrow micrometastasis of esophageal cancer. These molecular markers play important roles in esophageal cancer diagnosis, prognosis and treatment.
5.Study on lymphocytes activation in peripheral blood of patients with multiple sclerosis
Chinese Journal of Laboratory Medicine 2003;0(12):-
Objective To explore changes and its clinical significa nce of lymphocyte activation in patients with multiple sclerosis. Methods CD3、CD4、CD8、CD19、CD16+56、CD25、CD69、HLA-DR in peripheral blood (PB) were tested by flow cytometry. Results The percentage of CD3、CD4、CD8、CD16+56 on PB lymphocytes in MS group were lower than those in controls, CD4/CD8、CD19 on PB lymphocytes in MS group were higher than in controls. The percentage of CD69、CD25、HLA-DR、CD3/HLA-DR on PB lymphocytes of patients with relapse MS group were higher in controls and remission MS groups. The percentage of CD3/HLA-DR on PB lymphocytes of patients with remission MS group was higher than controls. Conclusion The content on PB lymphocytes in MS group were unusual. The lymphocyte activation was much higher and may be as a marker of MS.

International Journal of Laboratory Medicine 2017;38(8):1016-1018,1022
Objective To explore microRNA (miRNA) expression patients with β-thalassemia major. Methods MiRNA differential expression were detected in β-thalassemia major by miRNA microarray analysis and quantitative real-time PCR.Results The results of miRNA array showed that 26 differential expression miRNAs were up regulated and 30 differential expression miRNAs were down regulated.Hsa-miR-618 which expression was up regulated and hsa-miR-103a-2-5p which expression was down regulated were selected for quantitative real-time PCR detection. It was showed that the expression tendency of hsa-miR-618 and hsa-miR-103a-2-5p were consistent. So the method of miRNAs array was reliable. The target genes of 17 miRNAs which were up regulated and 24 miRNAs which were down regulated were predicted by using database software. Conclusion It is notable that the differential expression of miRNAs in patients with β-thalassemia major. It will be afforded new direction and thinking for the mechanism research and disease treatment through the further research of the miRNAs regulation pathway in β-thalassemia major.

Chinese Journal of Digestive Endoscopy 2016;33(4):215-218
Objective To investigate the safety and effectiveness of endoscopical therapy for small grastric stromal tumor( d<2 cm) . Methods Clinical records of 80 patients dignosed as having small grastric stromal tumor by pathology and immunohistochemistry were retrospectively analyzed. Endoscopic or laparo?scopic or open surgery was performed, so patients were divided into endoscopic treatment group ( n=37) and surgical treatment group ( n=43) accordingly. Perioperative period data, effectiveness and follow?up data were analysed. Results Thirty?five cases( 94?6%) of endoscopic treatment group were successfully treated by endoscopic surgery. Intraoperative perforation occurred in 8 cases, among which 2 cases transferred for conventional operations.Pneumoperitoneum occurred in 2 cases (better after symptomatic treatment), while no serious infection or perioperative death occurred. There were no significant differences in the complete re?section rate[ 91?89% ( 34/37) VS 100?0%( 43/43) , P=0?095) , the incidence of postoperative complica?tions[5?71%(2/35)VS 2?33%(1/43),P=0?855)or the improvement of postoperative symptoms[93?10%( 27/29) VS 85?71% ( 30/35) , P=0?589] ,while statistically significant differences were found in operation time[(37?41±13?45)min VS(84?56±38?37)min, P=0?000], intraoperative blood loss[(5?65±5?88)ml VS(31?48 ±39?57)ml, P=0?000], postoperative feeding time[(2?47±0?61)d VS(3?26 ±1?27)d, P=0?001], postoperative hospital stay[(5?76 ±2?28)d VS(7?64±2?99)d,P=0?022],and medical expenses [(18 554?4±9 736?45)yuan VS(31 138?11±1 206?24)yuan, P=0?000]. Conclusion Endoscopic re?section is safe and effective for small grastric stromal tumor with minimal invasion, simple procedure, faster recovery and less economic burden for the patients.
