2.Effect of emulsified isoflurane preconditioning on focal cerebral ischemia-reperfusion injury in rats
Chinese Journal of Anesthesiology 2010;30(6):736-738
Objective To investigate the effect of emulsified isoflurane preconditioning on focal cerebral ischemia-reperfusion (I/R) injury in rats and the mechanism. Methods Fifty-six healthy male adult SD rats weighing 250-280 g were randomly divided into 4 groups (n = 14): group Ⅰ sham operation (group S); group Ⅱfocal cerebral I/R; group Ⅲ emulsified isoflurane preconditioning and group Ⅳ fat emulsion preconditioning. Focal cerebral ischemia was induced by middle cerebral artery occlusion (MCAO) using a nylon thread with rounded tip inserted into internal carotid artery and advanced cranially until resistance was met. MCAO was maintained for 2 h followed by 24 h reperfusion. In group Ⅲ and Ⅳ 8% emulsified isoflurane 7.5 ml/kg and 3% fat emulsion 7.5 ml/kg were injected intraperitoneally at 24 h before MCAO respectively. Neurologic outcome was evaluated at 24 h of reperfusion and scored (0 = no deficit, 4 = unable to crawl, loss of consciousness). The animals were then killed and brains removed. The infarct size was assessed. The apoptotic cells were detected by TUNEL and calculated. The expression of Bax, Bcl-2, cytochrome C and caspase-3 protein was detected by immunohistochemical staining. Results The neurologic deficit scores were significantly lower in emulsified isoflurane preconditioning group than in I/R group. Preconditioning with emulsified isoflurane significantly decreased the infarct size and the number of apoptotic cells and increased the expression of Bcl-2 protein and inhibited the expression of Bax, cytochrome C and caspace-3 protein. Conclusion Emulsified iscflurane preconditioning can protect the brain from focal cerebral I/R by increasing Bcl-2 protein expression and decreasing Bax protein expression, and reducing cytochrome C release from mitochondria, caspase-3 activation and neuronal apoptosis.

Chinese Journal of Geriatrics 2010;29(8):656-657
Objective To observe the efficacy of Lipo PGE combined with Shuxuetong injection in treatment of diabetic peripheral neuropathy (DPN). Methods The 68 DPN patients were randomly divided into treatment group with Lipo PGE plus Shuxuetong injection and comparison group with Shuxuetong alone. The treatment period was 14 days. Results The pain, numbness, paresthesia and other symptoms in the limbs of patients were significantly reduced in treatment group than in comparison group (P<0. 01). At the same time, the conduction velocity of median nerve and sural nerve was significantly increased in treatment group after treatment (P<0.05), there were no significant differences in comparison group before and after the treatment. Conclusions The treatment of DPN with Lipo PGE combined with Shuxuetong injection is safe and effective.
4.Effect of dexmedetomidine on antioxidant ability in rats brain with focal cerebral ischemia/reperfusion
The Journal of Clinical Anesthesiology 2016;32(5):488-490
Objective To explore the effect of dexmedetomidine preconditioning on antioxidant ability in Rats Brain with focal cerebral ischemia/reperfusion.Methods Forty-two healthy male SD rats, weighted 250-280 g,were randomly divided into three groups (n = 14):sham-operation group (group Sham):in which carotid artery was exposed but MCAO was not performed;ischemia/reper-fusion group(group IR):NS were injected intraperitoneally at 30 minutes before the MCAO;dexme-detomidine group (group Dex):dexmedetomidine 100 μg/kg were injected intraperitioneally injected at 30 minutes before the MCAO.Focal cerebral ischemia-reperfusion (IR)model in rats was made by transient occlusion of the middle cerebral artery occlusion (MCAO)using a nylon thread with rounded tip inserted into internal carotid artery and advanced cranially until resistance was met.MCAO was maintained for 2 hours followed by 24 hours reperfusion.Neurologic deficit scores(NDS),the infarc-tion volume as well as the activities of endogenous antioxidants (such as superoxide dismutase(SOD), glutathione peroxidase(GSH-PX),glutathione reductase (GR),catalase (CAT))in ischemic brain were evaluated 24 h after reperfusion. Results Compared to group Sham, the neurologic deficit scores and the infarction volume in group Dex and IR were significantly higher, but the concentration of SOD,GSH-PX,GR,CAT were significantly lower(P <0.05).Compared to group IR,however, the neurologic deficit scores and the infarction volume in group Dex were significantly lower, but the concentra-tion of SOD,GSH-PX,GR,CAT were significantly higher (P <0.05).Conclusion Dexmedetomidine can protect the brain from focal cerebral ischemia-reperfusion injury, and its mechanism maybe relate to preserving the activities of endogenous antioxidants.
5.Signal transduction pathways of autophagy after cerebral ischemia
International Journal of Cerebrovascular Diseases 2016;24(5):463-468
Autophagy can be activated after cerebral ischemia and involved in the occurrence and development of ischemic brain injury. Therefore, the study of autophagy related signal transduction pathways may provide a new therapeutic target for ischemic brain injury. This article reviews the signal transduction pathways of autophagy after cerebral ischemia.
6.Neurobehavior and motor development changes in the 3-day-old rats with ischemic brain injury
Chinese Journal of Neonatology 2017;32(1):59-63
Object To investigate the pathological changes, physiological condition, neurobehavior and motor development of the 3-day-old rats with ischemic brain injury. Methods Ninety six 3-day-old male Sprague-Dawley rats were randomly assigned to experimental and control groups.Occlusion of both carotid arteries was performed in the experimental group, while the rats in the control group only received skin incisions without carotid ligation.Physical examinations and neurobehavioral development of the rats were recorded daily from the first day after operation until 3 weeks after birth, including weight, eye opening, incisor eruption, ear unfolding, righting reflex, negative geotaxis, limb placing and grasping reflex. Specimens of the brain tissue were obtained in 24 hours after operation and 3 weeks after birth for the hematoxylin-eosin staining and immunohistochemistry staining to investigate the pathological changes. All the reaults were compared between the 2 groups. Results Compared with the control group, rats in the experimental group were found growth retardation, suppression of primitive reflexes and impaired motor abilities (P < 0.05 ).The brain tissue obtained from the rats after operation showed white matter rarefaction, liquefaction and microglia hyperplasia with Hematoxylin-eosin staining and myelin formation disorder with immunohistochemistry staining. Conclusions Ischemic brain injury of the 3-day-old rats could result the mental retardation, neurobehavioral and motor development disorder because of the white matter injury.

10.Antibiotics Resistance Status Changing among 3 Kinds of Bacteria in Enterobacteriaceae
Chinese Journal of Nosocomiology 1994;0(01):-
OBJECTIVE To investigate the drug resistance status of Klebsiella pneumoniae,Escherichia coli and(Enterobacter cloacae) for reasonable use of antibiotics. METHODS After genus identification, drug resistance profile was analyzed by K-B method. RESULTS The predominant isolates from Enterobacteriaceae were(K.penumoniae) and E.coli.The resistant rate of the three commonly encountered bacteria in Enterobacteriaceae to antibiotics ascended yearly, with increasing isolates harboring ESBLs from 16.5%(2000) to 32.4%(2004). CONCLUSIONS Rational use of antibiotics should be obey the results of laboratory antibiotics susceptibility test to control the increasing trend and forbid outbreak of resistance.
