Objective To investigate the pathophysiological changes in irradiation-induced heart injury in rats in order to provide preventive and therapeutic strategies for irradiation-induced heart disease(RIHD).Methods Experiments were performed using female Wistar rats with body weight of approximately 220g.12 female Wistar rats were randomly assigned to two groups(sham-irradiated group and irradiated group).Heart of rats was locally irradiated with 60Co ?-ray at a dosage of 20 Gy.During the post-radiation period,the intake of food and activities of the animals were observed daily and the body weight was recorded monthly.On the 120th day after irradiation,myocardial perfusion was tested using real time myocardial contrast echocardiography(RTMCE).On the 180th day after irradiation,the hearts were isolated from the rats of the both groups,and they were perfused with a Langendorff apparatus.Hemodynamic measurements,including peak systolic LV pressure(LVSP),maximal rate of LV pressure rise(LV dp/dtmax)and fall(LV dp/dtmin)were studied.Then the hearts were examined pathologically with HE staining.Results Animals irradiated with single dose of 20Gy locally were apparently in a good condition during the observation period of 180 days.No differences in food intake,physical activities and body weight were observed between irradiated and control animals.However,myocardial blood velocity(beta)of irradiated rats was significantly lower than that of sham irradiated group(P

Objective To construct expressing recombinant of recF and recO from Deinococcus radiodurans R1and express the target protein in E.coli BL21(DE)3.Methods recF and recO gene were amplified by PCR from genomic DNA of Deinococcus radiodurans R1 and inserted into expression plasmid vector pET30b(+) to construct pET30b(+)-recF and pET30b(+)-recO.The recombinant plasmids were transformed into E.coli BL21(DE)3 and the recombinant proteins were expressed by the isopropyl-?-D-thiogalactopyranoside(IPTG) and were analyzed with SDS-PAGE.The changes in the survival rate of bacteria in each group before-and after UV-irradiation were calculated.Results The recombinant plasmid pET30b(+)-recO and pET30b(+)-recF was obtained and the recombinant protein could be highly expressed in E.coli BL21(DE)3.Conclusion This study has provided a foundation for further studies and applications of the recombinant RecF and RecO,and initial detection shows that recO and recF gene can increase the resistant ability of E.coli.

Objective To evaluate short-term clinical effects of modified total pelvic floor reconstruction surgery in treatment of severe pelvic organ prolapse.Methods Thirty-nine cases with severe pelvic organ prolapse including vault prolapse diagnosed by pelvic organ prolapse quantification (POP-Q) system received modified total pelvic floor reconstruction surgery.Clinical parameters associated peri-operative period and 12 months after surgery and complications were analyzed Results Median operation time was 70 minutes (30-240 minutes),median blood loss was 100 ml (10-200 ml).Seventy-seven percent (30/39) patients were able to micturate spontaneously in the next morning after surgery with postvoid residual volume less than 100 ml (0-650 ml).No severe intra-operative complications were recorded and the rate of postoperative morbidity was 20% (8/39 ).Median post-operative hospital stay was 4 days (1-11 days).The patients were followed up at median 24 months(13-29 months).According to POP-Q system evaluation,the successful rate of operation reached 100% .Two cases (5%,2/39) were recorded as symptomatic recurrence which manifested as posterior wall prolapse within 24 months after operation.During follow-up,8% (3/39) patients were found to have erosion within 7 months after surgery,and urgent urinary incontinence was observed in 5% (2/39) cases,while constipation occurred in 8% (3/39) cases.The most remarkable complication was dyspareunia (36%,5/14); while 50% (7/14) experienced better sexual life after surgery.Conclusions Modified total pelvic reconstruction is a safe,efficient and micro-invasive surgery in treatment of severe pelvic organ prolapse.However,its influence on post-operative sexual life should be concerned.

Objective To investigate the relation between serum asymmetric dimethylarginine (ADMA) level and endothelial function before and after living donor kidney transplantation in uremic patients.Methods A total of 38 renal transplant patients (21males and 17 females) and 36 healthy controls (20 males and 16 females) were enrolled. Plasma ADMA, symmetric dimethylarginine (SDMA), malondialdehyde (MDA), glutathione peroxidase (SeGSHPx), C-reactive protein (CRP) were mea-sured before transplantation and on Days 1, 3, 7, 14, and 28 posttransplantation. The brachial artery flow mediated dilatation (FMD) was studied before transplantation and on the 28th day after the transplantation. Results Serum levels of ADMA,SDMA,MDA and CRP were significantly increased, and the activities of nitric oxide (NO) and SeGSHPx were decreased in uremic patients compared with age matched healthy subjects (all P＜0.01). Serum levels of ADMA,SDMA,MDA and CRP decreased while the activities of NO and SeGSHPx increased significantly at the first day after the transplantation (all P＜0.01). The decrement of plasma SDMA normalized on the 28th day (P＞0.05). The FMD was lower in the patients than the control group (P＜0.01) and improved significantly on the 28th day of posttransplantation (P＜0.05). Serum levels of ADMA were positively correlated with MDA (r=0.412, P＜0.01;r=0.342,P＜0.01) and negatively correlated with the values of SeGSHPx (r=-0.345, P＜0.01;r=-0.315, P＜0.01) and FMD (r=-0.452,P＜0.01;r=-0.416,P＜0.01) both before and after kidney transplantation. Conclusion The level of serum ADMA is associated with endothelial function improvement in uremic patients both before and after kidney transplantation.

Objective:To identify the function of extracellular soluble vimentin that promotes proliferation, activation and chemotaxis of inflammatory cells. Methods: The proliferation of rat splenocytes stimulated with vimentin was evaluated in vitro. The lymphocyte counts and vimentin-antibody levels of peripheral blood in mice immunized with vimentin were detected in vivo. Peritoneal macrophages were collected and cultured with different concentrations of vimentin to detect the effect on phagocytosing chicken red blood cells. The chemotaxis of NIH3T3 fibroblast towards vimentin was observed in transwell chamber. Results:In vitro vimentin dose dependently promoted the proliferation of splenocytes. The proliferation indexes of primed and naive splenocytes cultured with 16μg/ml vimentin were reached up to ( 196. 0 ± 9. 7 )% and ( 208. 9 ± 4. 6 )% respectively without significant difference. In vivo vimentin significantly enhanced the lymphocytes number(109 L-1)of peripheral blood(5. 74±0. 51 vs. 1. 69±0. 13)and the levels of vimentin specific antibody( OD value 2. 31 ± 0. 06 vs. 0. 19 ± 0. 08 ) that shown no significant difference from immunization with vimentin plus CFA. In vitro vimentin dose dependently stimulated phagocytic ability of macrophages and performed the chemotactic effects on NIH3T3 fibroblasts. Conclusion:Extracellular soluble vimentin promotes the proliferation,activation and chemotaxis of concerned inflammatory cells and possesses the functions as an alarmin.

Objective: To study the nutritional status and problems of Chinese elite athletes for instructing training scientifically. Method: 599 elite athletes were investigated by means of dietary survey, physical measure and biochemical detection. Results: (1) The daily average energy intake reached the adequate intake (AI) recommended for athletes, but the energy ratio both of protein and fat higher, carbohydrate lower, being 18.9%, 38.6% and 42.5% respectively. Except vitamin A and B1, B2 the intakes of other vitamins and minerals were sufficient. (2) The average BMI and body fat percentage were 23.0?3.0 and (12.1?3.2)% in men and 21.9?3.0 and (20.5?3.9)% in women respectively. (3) The average hemoglobin levels were (145.7?13.3)g/L in men and (130.6?11.8)g/L in women. The rates of anemia and iron deficiency anemia were 12.6% and 5.2% respectively, female higher than male. (4)The hyperlipidemia rate was 22.3%, including 12.6% high TG, 9.7% high TC and 1.2% high LDL, female higher than male. (5) The rates of VB1 and VB2 insuficiency were 46.2% and 32.7% respectively, both including 9.6% deficiency. Conclusion: The nutritional status of Chinese elite athletes was good, but still with anemia, vitamin insufficiency and hyperlipidemia.

Objective To investigate the respiratory mechanics and treatment outcomes of different types of mechanical ventilation for patients with neonatal acute respiratory distress syndrome(ARDS):the high frequency oscillation ventilation + pulmonary surfactant (HFOV+PS),conventional mechanical ventilation + pulmonary surfactant(CMV+PS),conventional mechanical ventilation(CMV). Methods Seventy-five cases with neonatal ARDS,25 cases in the HFOV+PS group,30 cases in the CMV+PS group,20 cases in the CMV group. Patients in the former two groups received 70 mg/kg PS at a time. PaO2,PaCO2,PaO2/FiO2,oxygenation index(OI)and respiratory index(RI)were detected at 0 h,12 h,24 h,48 h,72 h post-mechanical ventilation. Results At 12, 24,and 48 hours post-mechanical ventilation,patients in the HFOV+PS group had a significantly higher level of PaO2 and a significantly lower level of PaCO2 than those of patients in the CMV+PS group and the CMV group(P<0.05). At 12,24,48,and 72 hours post-mechanical ventilation,patients in the HFOV+PS group had a significantly higher level of PaO2/FiO2 and significantly lower level of OI and RI than those of patients in the CMV+PS group and the CMV group (P < 0.05,respectively). Patients in the HFOV+PS group also had significantly shorter durations of mechanical ventilation and oxygen usage than those of patients in the CMV+PS group and the CMV group (P < 0.05). No significant differences were observed in Gas leak,the incidence of intracerebral haemorrhage and cure rate among the three groups. Conclusions Application of HFOV with PS therapy for patients with neonatal ARDS can timely improve the oxygenation,shorten the time of mechanical ventilation and the usage of oxygen,without increasing complications.

Objective: To explore the effects of Polygonatum odoratum on body based on metabonomics. Methods: The ultra performance liquid chromatography coupled with mass spectrometry (UPLC-MS) and principle component analysis(PCA) and orthogonal partial least squares discriminant analysis(OPLS-DA) were used to evaluate the changes of endogenous substances in rats after intragastric administration of Polygonatum odoratum. Results: Significant differences between the control group and administration group were observed in PCA and OPLS-DA model. Five potential biomarkers between control group and administration group were identified. The relative content of Alpha-Tocotrienol, PC(14:1(9Z)/14:1(9Z)), Stearic acid, Theasapogenol A, Docosahexaenoic acid increased. Conclusion: The biomarkers had many beneficial activities, so the Mongolian medicine Polygonatum odoratum had the function of health care. Objecttive To observe the repaired effect of distinct source olfactory ensheathing cells (OECs) on spinal cord injury (SCI) rats. Methods These OECs were dissociated from olfactory bulb and olfactory mucosa of SD rats and transplanted to the injuried region of spinal cord injury rats. The function of nerve, motor evoked potential of hind legs and the histopathlogical diversities of injuried spinal cord were observed. Results The OECs grafts into the SCI area could survive longer time. The BBB scale, incubation stage of EP and histopathologic manifestations showed that the group with transplanted OECs regained more improvement in hindlimb than the control group. Conclusion The OECs of two sources have the same ability to regain and improve the axonal function which can promote axons regeneration of SCI.
