1.The role of TLR4 in pathophysiology of depressive disorder

Depression is one of the most common mental disorders.Since 1960s,researchers have proposed a number of etiological hypotheses from the aspects of genetics,biochemistry,neuroendocrine and immune,but so far it has not been fully under-stood.Toll like receptor 4(TLR4),as a bridge between innate immunity and adaptive immunity,could induce inflammatory by activating related signal transduction pathways and promoting the expression of inflammatory factors in c-ells.More and more studies showed that the occurrence of depression was related to inflammation.Reviewing relationship of TLR4 and depressive disorder and exploring the effects of some interventions for depression on TLR4.Research on TLR4 shed light on pathological mechanism of depressive order.TLR4 could play an important role in inhibiting the production of 5-serotonin,the disorders of HPA axis,interfering with the hippocampus and cortical nerve regeneration,the autophagy of hippocampus.
3.Application of "Three-step training and Two-part Examination" in Utilization and Training of Defibrillator

Objective To research the applicable ways on training and examination of defibrillator utilization in grassroots units. Methods The use of the "three-step training and two-part examination" mode, that is, three-step training(theoretical instruction, grouped practice and animal experiment), a total of 83 medical officers from inferior troops of PLA were trained for the use of defibrillators, and through passing two-part test(written test and skill test), the trainees were subsequently examined so that the efficacy of the training could be evaluated. Results The trainees can be acquired satisfying achievement in the two-part test after having passed the three-step training. The respectively average scores were 86.7 ?15.9 (written test) and 90.1?14.8 (skill test). Conclusion The "three-step training and two-part test" mode is a successful defib- rillators' training mode for inferior troops.
4.Effects and evaluation of misoprostol in hysteroscopy

AIM : To observe the effects of misoprostol used for intenerating cervical and ease pain in hysteroscopy. METHODS : 63 of 107 cases were randomly chosen as the group of misoprostol, and 200 ?g misoprostol was taken orally 1h before hysteroscopy. Another 44 cases were taken as the group of antitheses, and no medication was taken before hysteroscopy. RESULTS : The diameter of cervical, hemorrhage, the rate of PAAS and bellyache in the group of misoprostol were lower than that in the group of antitheses. CONCLUSION : 200 ?g misoprotol orally 1h before hysteroscopy is an advantageous and ideal method with alleviating pain from wound, reducing manipulation, decreasing hemorrhage, reducing PAAS happening in intenerating cervical.
5.Evaluation of clinical efficacy of combination of active immunotherapy and allylestrenol in treatment of patients with unexplained recurrent spontaneous abortion

AIM: To evaluate the clinical efficacy of combination of active immunotherapy and allylestrenol in treatment of patients with unexplained recurrent spontaneous abortion (URSA) and to investigate a best therapeutic method in treatment of patients with URSA. METHODS: 435 patients with primary URSA were randomly assigned to three groups: 185 in combination medication group which was treated with active immunotherapy and allylestrenol, 152 in active immunotherapy group which was only treated with active immunotherapy, 98 in allylestrenol group which was only treated with allylestrenol. 96 secondary URSA in secondary URSA group were treated with active immunotherapy and allylestrenol. RESULTS: The successful pregnant rates of combination medication group, active immunotherapy group, allylestrenol group and secondary group were 92.05 %, 71.43 %, 31.51 % and 86.76 %, respectively. The successful pregnant rate in combination group were higher than that in the active immunotherapy group and the allylestrenol group (P 0.05 ). CONCLUSION: Combination of active immunotherapy and allylestrenol is better than other therapy method in treatment of patients with URSA, which is worth further widespread application in clinical practice.
6.Dynamic changes of Kir2.3 mRNA in the hippocampus of rats with chronic temporal lobe epilepsy

Objective To observe dynamic changes of Kir2. 3 mRNA in the hippocampus of rats with chronic temporal lobe epilepsy, and to discuss the relationship between Kir2. 3 expression and the pathogenesis of chronic temporal lobe epilepsy. Methods We used pilocarpine to induce status epilepticus (SE) in rats,which became chronic temporal lobe epileptic rats in 2 weeks. The expression of Kir2.3 mRNA was measured by reverse transcription polymerase chain reaction (RT-PCR) at the time points of 0, 6, 72 hours and 2 weeks after SE. Results The ratios of Kir2. 3 mRNA to β-actin of normal control and 0, 6, 72 hours, 2 weeks after SE were 0. 080 ± 0. 030, 0. 103 ± 0. 045, 0. 164 ± 0. 026, 0. 132 ± 0. 024, and 0. 011 ± 0. 008, respectively. The ratio was significantly higher 6 and 72 hours after SE and significantly lower 2 weeks after SE than that of the normal control. Conclusions Two weeks after SE, when the rats had spontaneous recurrent seizures, the expression rate of Kir2.3 reached a turning point, which possibly became the basis of epileptogenesis.
7.Analysis of the policy support for running community healthcare centers by various entities

Objective To study the financial inputs into community health School of Medicine and Health Management, TONGJI Medical College of HUAZHONG[service (CHS) institutions by entities of different ownerships, by comparison of their investment, housing and human resources for policy recommendations.Methods Choose 6 cities in the East, Central and West of China through stratified random sampling. Collect data by literature review, in-depth interview, focus group discussion and questionnaire survey. Analyze quantitative data through EPI data 3.02 and SPSS 13.0.Results The institutions owned by the government and hospitals were advantageous in financial investment and housing; institutions under social organizations or individuals were inferior with development supports (P=0.00). Conclusion The community health service principle of government dominance and social involvement should be advocated, with equal opportunities for community service providers of various ownership. Emphasis should be placed on betterment of policies and measures, specific budget subsidy, as well as performance appraisal and service supervision. These measures aim at encouraging non-government providers to play greater role in community health service.

9. Study on the mechanism of gambogenic acid-induced apoptosis of human colon cancer HCT116 cells

Objective: To investigate the pro-apoptotic effect of gambogenic acid (GNA) on human colonic carcinoma HCT116 cells, and to explore the possible molecular mechanism. Methods: MTT assay was performed to detect proliferation inhibition effect of GNA on HCT116 cells. After staining with DAPI, the pro-apoptotic effect of GNA on HCT116 cells was observed under a fluorescence microscope. The changes in cell cycle distribution of HCT116 cells induced by GNA were examined by FCM. The expressions of cyclin D1, cyclin E, P21, P27 and poly (ADP-ribose) polymerase (PARP) proteins in HCT116 cells were detected by Western blotting. Results: GNA exerted an significant inhibitory effect on HCT116 cell proliferation in a time-and dose-dependent manner, and it could induce the typical nuclear apoptotic morphology. Under a fluorescence microscope, DAPI staining results showed that GNA could obviously induce the apoptosis of HCT116 cells. FCM showed that GNA could significantly increase the percentage of cells at G0/G1 phase, whereas obviously decrease the percentage of cells at S phase, which indicated that the cell cycle was arrested at G 0/G1 phase. Western blotting analysis showed that GNA could efficiently down-regulate the expression levels of cyclin D1 and cyclin E proteins, whereas up-regulate the expression levels of P21 and P27 proteins, and also promote the cleavage of PARP. Conclusion: GNA can induce the cell cycle arrest at G0/G1 phase by down-regulating the expression levels of cyclin D1 and cyclin E, and up-regulating the expression levels of P21 and P27. Therefore, GNA can efficiently promote the cell proliferation inhibition and the apoptosis of HCT116 cells. Copyright© 2011 by TUMOR.
