1.Molecular biological properties of lncRNA and their regulatory functions in the development of central nervous system

Long noncoding RNA (lncRNA) refers to RNA transcripts with more than 200 nucleotides in length, which plays regulatory roles directly in the form of RNA. Their functions are affected by their secondary structures, post transcriptional modification and so on. LncRNA can be classified to various subtypes with complicated mechanisms. In human-induced-pluripotent stem cells (iPSCs) and embryonic stem cells (ESCs), dynamic expression of some lncRNAs is detected during the neuronal differentiation, and a series of lncRNAs associated closely with the development process of the central nervous system (CNS) are found. This review summarizes the molecular biological properties of lncRNA and their regulatory functions in the development of the CNS, which is helpful to reveal the mechanisms of the pathogenesis and progression of certain neurological diseases and results in innovation of their diagnosis and treatment.
2.Treatment of ectopic pregnancy by laparoscopic surgery:A report of 73 cases.

Objective To assess the value of laparoscopic treatment of ectopic pregnancy. Methods Laparoscopic operations were performed,including injection MTX into the tube of ectopic pregnancy,linear salpingostomy,salpingectomy and partial oophorectomy. Results Among 73 cases of ectopic pregnancy,70 cases(95.9%)were confirmed as tubal pregnancy,of which there were 38 cases of tubal abortion(54.3%)and 19 cases(27.1%) of rupture.No blood was found in the abdoment in the cases of unrupture and unhappened tubal abortion,whereas there was 100ml~3,000ml blood in others.There was only spotting in the cases of unrupture and unhappened tubal abortion,and there was 10ml~80ml of bleeding in other cases.The operating time varied from 34 min to 108 min. Conclusions The laproscopic surgery can be chosen as the first method in the treatmentof ectopic pregnancy.
3.Study on miR-148a regulation function of cardiac differentiation on MSCs by targeting DNMT1

Objective:To verify DNA methyltransferase 1(DNMT1) is the direct target of miR-148a and explore the internal mechanism by which miR-148a regulates the cardiac differentiation of mesenchymal stem cells(MSCs)by directly targeting DNMT1.Methods:miRNA microarray was used to screen out the abnormally expressed miRNAs in MSCs before and after 5′-azacytidine(5′-aza) treatment.Target scan was uesd to predict the target of miR-148a.miR-148a mimics and DNMT1-wt,scramble and DNMT1-wt,miR-148a and DNMT1-mut,scramble and DNMT1-mut were co-transfected in MSCs cells and luciferase activity were detected by single photon.MSCs cells were transfected with miR-148a lentivirus plasmid.Respectively extract DNA,RNA and protein 1,7,14 and 28 days after transfection.The CpG methylation level on DNA regulatory sequences of Gata-4 upstream gene was detected by methylation detection,the mRNA and protein expressions of myosin heavy chain(MHC),cardiac troponin T(cTnT),CD90,CD29,Nkx2.5 and Gata-4 were detected by qRT-PCR and Western blot.Results:miRNA Microarray screened out the abnormally expressed miRNAs in MSCs before and after 5′-aza-induced,including miR-146a-5p,miR-148a,miR-539,etc.Among which miR-148a was remarkably upregulated.Targetscan,Luciferase Reporter Gene and qRT-PCR verified that DNMT1 was the direct target of miR-148a.By infected MSCs cells with miR-148a lentivirus plasmid,we confirmed that the methylation level of Gata-4 gene upstream was changed,and with prolong of transfection,the methylation level of Gata-4 was decreased,the expression of MHC and cTnT were increased,while CD90 and CD2 were continually decreased,the mRNA expression of Nkx2.5 and Gata-4 were increased MSCs cells started myocardial differentiation.Conclusion:miR-148a can regulate the cardiac differentiation of MSCs by directly targeting DNMT1.

6.Effect of microgroove surface morphology on the adhesion and proliferation of human gingival fibroblasts

Objective:To investigate the effects of microgroove surface morphology on the adhesion and proliferation of the human gin-gival fibroblasts(HGFs).Methods:Microgroove surfaces of titanium were fabricated by photolithography with parallel grooves of 15,30 or 60 μm in width and 5 μm or 10 μm in depth.The groups of the samples were denoted as T15 /5,T15 /10,T30 /5,T30 /10,T60 /5 and T60 /10.Smooth titanium surface(T0)was used as the control.The surface topography was observed by enviroment SEM(ES-EM).HGFs were cultured on the topographically modified surfaces.Morphology was observed by SEM.Cell proliferation was examined by CCK-8 kit.Results:HGFs on the microgroove surfaces had “contact guidance”parallel to the microgrooves,whereas the cells on T0 were oriented randomly.Cell proliferation was promoted and kept for longer period on T60 /10 surface.Conclusion:Surfaces of mi-crogrooves with increasing groove width and depth may achieve “contact guidance”for HGFs and promote cell proliferation.
7.The effects of microgroove surface form of titanium on the adhesion and cell cycle progression of human gingival fibroblasts

Objective:To investigate the effect of microgroove surfaces of titanium on the adhesion and cell cycle progression of human gingival fibroblasts(HGFs).Methods:Microgroove titanium surfaces were fabricated on silicon plate by photolithography with parallel grooves:1 5,30 and 60 μm in width and 5 μm and 1 0 μm in depth,the groups were denoted as T1 5 /5,T1 5 /1 0,T30 /5,T30 /1 0, T60 /5 and T60 /1 0,respectively.Smooth titanium surfaces (T0)were used as the controls.Surface topography were observed by ES-EM.HGFs were cultured on the microgroove surfaces.Morphology of “contact guidance”was observed by immunofluorescence tech-nique.Cell cycle distribution was analyzed by Flow cytometry.Results:HGFs on the microgroove surfaces had “contact guidance”par-allel to the microgrooves,whereas the cells on T0 were oriented randomly.T60 /1 0 group had the highest percentage of S phase cells, followed by T30 group and T1 5 group,but still higher than that in the control group.In groups with higher groove width (T60 group and T30 group),the increase of groove depth benefited the increase of S phase percentage,while in T1 5 group,the increase of groove width decreased the S phase percentage.Conclusion:Surfaces of microgrooves with different dimensions achieved “contact guidance”for the cultured HGFs.The surfaces with increasing groove width and depth benefit the cell cycle progression.
8.Implemention of cognitive behavioral therapy in the young female with facial burn

Objective To explore the effects of cognitive behavioral intervention in psychological nursing of young female patients with facial burn.Methods 64 cases of young women patients with facial bum were selected as the research object,and randomly divided into the observation group and the control group,with 32 cases in each group.The observation group was given the cognitive behavioral intervention and the control group was done by conventional nursing methods.The anxiety and depression scores between the two groups before and after the intervention were compared.Results After injury,the anxiety and depression scores of both the observation group and the control group were higher than those of the domestic norm; the anxiety and depression scores between two groups showed no statistic significance before intervention,and after intervention,the anxiety and depression scores between two groups had significant difference,scores of the observation group were lower than those of the control group.Conclusions Cognitive behavioral intervention can reduce anxiety and depression of young female patients with facial burn.It is a kind of effective psychological nursing method,and worthy of clinical promotion.
9.Experimental study on effects of blood-stage treatment on Th17/Treg in patients with psoriasis vulgaris

Objective To investigate the action mechanism of blood-stage treatment that affects psoriasis vulgaris using observation of the Th17/Treg expression.Methods A total of 32 patients ( blood heat group,n =17 ; blood stasis group,n =15) and 15 healthy people ( control group,n =15 ) were observed.The frequencies of Th17 and Treg in peripheral blood were detected by flow cytometric analysis in patients before and after treated by heat-clearing and blood-cooling decoction,qi-enriching and blood-activating decoction.Results The ratio of Th17/Treg in peripheral blood [ blood heat group (4.21 ± 0.52 )% ;blood stasis group( 3.32 ± 0.43 )% ] was significantly increased in patients compared with controls [ ( 1.79 ±0.18)% ] ( P <0.01 ).After herbal treatment,the ratio of Th17/Treg expression in the blood heat group [ ( 2.41 ± 0.22 ) % ] and in blood stasis group [ ( 2.02 ± 0.12 ) % ] was significantly decreased ( P < 0.05 ),respectively.Conclusions Blood-stage treatment works well on Th17/Treg expression in patients with psoriasis vulgaris.
10.The long time aerobic training effect on the mitochondrial DNA content and the activity of mitochondrial respiratory chain complexes in aging rats

Objective: To study long time aerobic training effect on mitochondrial DNA (mtDNA) content and activities of mitochondrial respiratory chain complexes in the myocardium and brain tissues of aged rats. Method: Forty-five rats were randomly divided into three groups Group A(n=15)younger rats did not received training; Group B (n=15) old rats did not received training; Group C (n=15) old rats received gradually training for 90 days. The mtDNA content were determined by methods of Yah erc, the activity of mitochondrial respiratory chain complexes in myocardium and brain tissues were determined by methods of Wu etc. Result: The mitochondrial DNA content in myocardium and brain tissues significantly increased (P<0.01) and the activity of mitochondrial respiratory chain complexes in myocardium and brain tissues significantly decreased (P<0.05-0.01) in the aging control group as compared with those in the young group. The mitochondrial DNA content in myocardium significantly decreased (P< 0.05), while there was no significant difference in brain. The activity of mitochondrial respiratory chain complexes in myocardium and brain tissues significantly incroased(P<0.05-0.01) in the aging training group as compared with those in the aging control group. Conclusion: the long time aerobic training could decline the mitochondrial DNA content and increase the activity of mitochondrial respiratory chain complexes in myocardium and brain tissues in aging rats.
