1.THE EFFECTS OF ACUTE ARTIFICIAL INTRAOCULAR HYPERTENSION ON THE ACTIVITY OF SUCCINATE DBHYDROGENASE OF THE RETINAL GANGLION CELLS IN RATS

Twelve Sprague-Dawley albino rats (24 eyes) were used in this study. Nor- mal saline was injected into the anterior chamber of the experimental eye until the b-wave of the electoretinogram was extinguished, then maintained the critical high intraocular pressure (HIOP) for 90 minutes. Just a puncture, without HIOP, was given to the anterior chamber of the other eye of the same rat as a control Both retinas of the experimental and control eyes were whole-mounted and treated with succinate dehydrogenase (SDH) histochemical technique. The reacted products of SDH showed in either retinal ganglion cells (RGCs) or optic nerve fibers of the experimental eyes were lighter than those of the control eyes under microscope. Furthermore, the grey scale values of the reacted products of SDH on the matched ratinas were measured at corresponding microscopic fields and RGCs with an image analysis system. Rapid Golgi preparations of golden hamster dorsal lateral geniculate nucleus (LGd)show three classes of neurons with different morphological characteristics. Class Ⅰ and class Ⅲ cells are distributed throughout the LGd.Class Ⅱ cells are found more common medially at mid-genieulate levels.Class Ⅰ cells are multipolar neurons with large cell body.The smooth primary dendrites branch extensively. Spine-like and small excrescences are found along distal dendrites.Class Ⅱ cells have medium size pear-like cell body.The distal dendrites are usually derived from one primary dentrite.Dense cluster of dentritic protrusions is usually found at branch points of the dendrites.Class Ⅲ cells have the smallest cell body and are paucidendritie but the dendrites have complex protrusions including pearl-like dendritic appendages.Class Ⅰ and class Ⅱ cells possess an axon but most of class Ⅲ cells are not found to have an axon. The connections between the LGd and visual cortex was investigated using the retrograde horseradish peroxidase(HRP)histochemical method.After injection of HRP into the visual cortex,Class Ⅰ and class Ⅱ neurons were labelled but class Ⅲ celts were not suggesting that class Ⅰ and class Ⅱ cells are projecting neurons.Class Ⅲ cells do not seem to project to the visual cortex and are probably local circuit neurons.
3.The effect of concentrate growth factor extract on MC3T3-E1 cells

Objective:To evaluate the effect of concentrate growth factor extract(CGFe)on the proliferation and differentiation of MC3T3-E1 osteoblasts.Methods:MC3T3-1 cells were cultured with and without CGFe respectively.At 1 ,3,5 d of culture the pro-liferation of the cells was detected by MTT assay,the activity of alkaline phosphatase (ALP)of the cells was examined by ALP kit;at 1 4,21 d of culture the calcium nodus formation was observed by Alizarin red staining;RUNX2 and OSX expression was quantified by real-time PCR.Results:With the extension of culture time,the proliferation,ALP activity,the calcium nodus and the level of RUNX2 and OSX mRNA of the experimental group were increased more than those of the control group.Conclusion:CGFe may pro-mote the proliferation and differentiation of osteoblasts.
4.Effects of concentrated growth factor extract on the proliferation and differentiation of MC3T3-E1 cells attached to titanium surfaces.

OBJECTIVETo evaluate the effect of concentrated growth factor extract (CGFe) on the proliferation and differentiation of MC3T3-E1 osteoblasts attached to sandblasted and acid etched titanium surfaces.
METHODSTrials were divided into experimental and control groups. The experimental group used a-MEM that contained CGFe (10% FBS), whereas the control group only used a-MEM (10% FBS). MTT assay was employed to detect the number of osteoblasts on the first, third, and fifth days. Alkaline phosphatase (ALP) activity and scanning electron microscope (SEM) were used to detect the osteoblast differentiations on the third and fifth days and to observe the osteoblast extensions on titanium surfaces for 12 h, respectively. Meanwhile, the levels of the osteogenetic biomarkers Runt-related transcription factor-2 (Runx2) and Osterix (Osx) on the third and seventh days were quantified via real-time polymerase chain reaction (PCR).
RESULTSMTT assay indicated that on the first, third, and fifth days, the absorbance in the experimental group significantly increased than that in the control group (P < 0.05). ALP activity: on the third and fifth days, the absorbance of the experimental group was significantly higher than that of the control group (P < 0.05). SEM: at 12 h, the extension of the experimental group cells was larger than that of the control group. Real-time PCR: given the standardization in the group, the gene expression level of the control group on the third day was 1, and the Runx2 and Osx gene expressions in the experimental group were larger than those of the con- trol group.
CONCLUSIONCGFe can efficiently stimulate the proliferation, differentiation and extension of MC3T3-E1 cells.

5.HISTOCHEMICAL INVESTIGATION OF THE CYTOCHROME OXIDASE OF RAT RETINAL GANGLION CELLS BY ACUTE EXPERIMENTAL ELEVATION OF INTRAOCULAR PRESSURE

The changes of cytochrome oxidase activity of retinal ganglion cell (RGC) caused by the artificial high intraocular pressure (HIOP) was presented in this paper. Normal saline was injected into the anterior chamber of the right eye of 36 rats. that resulted in an intraocular pressure up to 60 mmHg for 3 hours. The experimental rats were divided into 0-, 3- and 7-day groups according to their survival time. Both retinae of each rat were whole mounted on the same slide, the left one being used as control. Thirty-six pairs of retina were treated with the modified cytochrome oxidase (CCO) histochemical technique under similar conditions. The high CCO activity of RGC were counted. The extinction of the cytoplasm of RGC, which indicates the degree of CCO activity, was measured with microphotometer. As compared with the normal eyes, the high CCO activity RGC of experimental eyes were reduced significantly. It was found that the high CCO activity of RGC in the temporal side of retina has been reduced much more than that of the nasal side. However, the high CCO activity of RGC in 3- and 7-day groups were more than that of 0-day group, the extinction of CCO activity of RGC in 7-day group was lower than that of the 3-day group. These facts showed that the CCO activity might restore in various degrees followed by a longer survival time. This experiment emphasized the fact that the HIOP led to metabolic changes of RGC, which may be of value to the study of glaucoma.
6.The effects of p38MAPK inhibitor SB203580 on MC3T3-E1 cell proliferation and differetiation under high glucose concentration

Objective:To investigate the influence of p38MAPK inhibitor SB203580 on osteoblast MC3T3-E1 cells under high glu-cose concentration.Methods:MC3T3-E1 cells were exposed to glucose at 5.5 mmol/L,25.5 mmol/L and 25.5 mmol/L combined with SB203580 respectively.Cell proliferation was analysed by MTT assay,calcified nodules were quantitated by Alizarin Red S stai-ning.Osteogenesis gene(OCN,Runx2)mRNA level was examined by realtime PCR.Results:High glucose decreased cell prolifera-tion(P<0.05),ALP activity(P<0.05),calcified nodule formation and osteogenesis gene expression of MC3T3-E1 cells.SB203580 reversed the down-regulation of osteogenic markers under high glucose,increased the ALP expression but decreased cell proliferation. Conclusion:SB203580 is antagonistic to P38MAPK in the regulation of preliferation and differetiation of MC3T3-E1 cells.
7.Experimental study of the effects of olfactory lamina propria transplantation and ganglioside GMI treatment on spinal curd injury

Objective To observe the effects of olfactory lamina propria (OLP) transplantation and ganglioside GM1 treatment on spinal cord injury (SCI) in rats.Methods Totally 50 healthy pure breed female Sprague-Dawley (SD) rats after spinal cord hemiseetion were randomly divided into 5 groups and were given different treatments: (OLP + GM1) treatment group (group A), GM1 treatment group (group B), OLP treatment group (group C), spinal cord injury but without treatment group (group D) and healthy control group (group E). The recovery of neurological function was evaluated by somatosensory evoked potential (SEP) and pathological examination after surgery. Results In group A, in some rats an escaping response in right hind leg occurred, but in other groups, the motor function was not significantly improved. Histological examination showed that transplanted olfactory lamina propria survived in the transplantation area and expanded on certain routes. NF positive nerve fibers passed through the transplantation area. Compared with group B, C, D, the N1-wave latency was(4.71±0. 72)ms 4 weeks after operation(P<0. 01), and the NF density was(7. 31±0. 26) ×104/mm28 weeks after operation in group A(P<0. 05). Conclusions Olfactory lamina propria (OLP) transplantation and ganglioside GM1 treatment have a synergistic effect on SCI.

Heparin-induced thrombocytopenia is a profoundly dangerous,potentially lethal,immunologically mediated adverse drug reaction to unfractionated heparin or low-molecular weight heparin.Clinical vigilance of this disease process is important to ensure its recognition,diagnosis,and treatment.Misdiagnosis of the syndrome,as well as misunderstanding of the disease process,continues to contribute to its morbidity and mortality.In this comprehensive review,the authors highlight heparin-induced thrombocytopenia's clinical presentation,diagnostic principles,and treatment.

A patient was treated by multiple dental implants,the implanted 6 was adjucent to impacted 8 .Immediately after implanta-tion,4 month and 3 year after implantation the distace between 8 and 6 implant central line was 4.4,3.2 and 2.5 mm,the angle between 8 long axis and 6 implant central line was 42.3°,45.5°and 50.3°.Then 8 was extracted.
10.A study of parameters of a volumetric modulated arc therapy plan for cervical and upper esophageal cancer

Objective To figure out the optimal parameters of a volumetric modulated arc therapy ( VMAT) plan for cervical and upper esophageal cancer by quality evaluation of VMAT plans with different parameters, and to provide a reference for the design of clinical VMAT treatment plan. Methods Ten patients with cervical esophageal cancer and ten patients with upper esophageal cancer were enrolled as subjects. The Nucletron Oncentra 4. 3 treatment planning system was used to generate plans for Elekta Synergy VMAT accelerator. Six VMAT plans were made with variation in the gantry angle ( 2°, 3°, and 4°), the maximum delivery time (80 s, 110 s, and 150 s), and the collimator angle (0° and 45°). The doses to the planning target volume and organs at risk were analyzed by paired t test. Results For cervical and upper esophageal cancer, the quality of VMAT plans with a collimator angle of 45° was better than those with a collimator angle of 0°(P=0. 003?0. 007). For cervical esophageal cancer, there was no significant difference in quality between VMAT plans with a maximum delivery time of 110 s or 150 s and those with a maximum delivery time of 80 s ( P>0. 05 );for upper esophageal cancer, there was also no significant difference in quality between VMAT plans with three different maximum delivery times ( P>0. 05 ) . For cervical esophageal cancer, the VMAT plans with a gantry angle of 3° had a better quality than those with a gantry angle of 2° or 4°(P=0. 010?0. 048). For upper esophageal cancer, the VMAT plans with a gantry angle of 3° had a better quality than those with a gantry angle of 4° ( P=0. 010?0. 048) . Compared with those with a gantry angle of 2° , the VMAT plans with a gantry angle of 3° had a slightly better dose distribution in the target volume ( P=0. 046 ) , but a slightly higher dose to lung tissue ( V25 and V30 , P=0. 007 and 0. 026) . Conclusions The optimal initial parameters of a VMAT plan for cervical and upper esophageal cancer are a collimator angle of 45°, a maximum delivery time of 80 s, and a gantry angle of 3°.
