1.Differentiation of mesenchymal stem cells to chondrocytes and its applications in cartilage repair

Articular cartilage as a load-bearing connective tissues may often be caused defects by trauma,sports, degenerative or age-related diseases. As articular cartilage has a limited inherent repair capacity, a large variety of treatment approaches including autologous chondrocyte transplantation, microfracture and mosaicplasty methods have been developed. However, there are limitations in these strategies especially when using autologous chondrcytes. Mesenchymal stem cells are considered the most promising seed cells in the study of tissue engineered cartilage. This review focuses on the induction of mesenchymal stem cells into chondrocytes in vitro and their application in cartilage repair.

3.Risk factors for symptomatic intracranial hemorrhage after intravenous thrombolysis in patients with acute ischemic stroke

Intravenous thrombolysis has been proved as an effective therapy for acute ischemic stroke.However,symptomatic intracranial hemorrhage (sICH) occurred after thrombolysis will affect the recovery of patients,even endangers life when it is serious.Therefore,determining the risk factors for sICH after thrombolysis is helpful to select appropriate patients for thrombolytic therapy.The risk factors for slCH after thrombolysis include advanced age,male gender,obesity,higher National Institutes of Health Stroke Scale scores,hypertension,diabetes or higher blood glucose,atrial fibrillation,delayed thrombolysis time,antiplatelet therapy,anticoagulation therapy,and leukoaraiosis.However,some of the risk factors still have conflicting reports.Further large sample clinical studies are needed to confirm them.
4.Role of autophgy in atherosclerosis and its modulation

Autophagy is a process for the degradation of long half-life proteins and organelles via the lysosome system. A large body of evidence indicates that the autophagic phenomena exist in the progressive atherosclerotic plaques. Vascular smooth muscle cells, macrophages, and endothelial cells treated with some stimulants that result in atherosclerosis formation in in vitro experiments, these cells show certain autophagic features, such as myelin-like structures, accumulation of ubiquitinated inclusions in the cytoplasm, and extensive vacuolization. However, although the interests in research on autophagy are increasing, the exact role of autophagy in atherosclerosis remains unclear. Therefore, understanding the underlying cellular and molecular mechanisms of autophagy will provide a new idea for studying the mechanisms and treatment of atherosclerotic disease.
5.Vertebral hemangiomas treated with the CT-guided percutaneous vertebroplasty

Objective To discuss the value of vertebral hemangiomas treated with the CT-guided pereutaneous vertebroplasty (PVP).Methods Forty-two patients with 49 vertebral lesions were guided by CT and injected with bone cement percutaneous into the vertebral.The effectiveness was valued by preoperative and postoperative visual analogue scale (VAS),locomotor activity scores,pain medication scores and comprehensive evaluation of long-term efficacy of WHO criteria,and undertook statistical analysis.Results The success rate in all patients was 100.0%(42/42),no serious complications.All the patients were followed up for 6 - 18 ( 12.50 ± 1.15 ) months,all lesions of the vertebral body imaging showed no tumor recurrence and vertebral collapse.The VAS scores decreased from preoperative (7.64 ± 1.29 ) scores to postoperative 1-3 days (4.82 ± 1.21 ) scores and to postoperative 3 -6 months (0.22 ±0.19) scores (P<0.05).The locomotor activity scores decreased from preoperative (3.4 ± 0.9) scores to postoperative 1-3 days (2.1 ±0.6) scores and to postoperative 3-6 months (1.0 ±0.3) scores (P<0.05).The pain medication scores decreased from preoperative ( 1.9 ± 0.6) scores to postoperative 1 - 3 days months ( 1.1 ± 0.2 ) scores and topostoperative 3-6 months (0.2 ± 0.1 ) scores (P < 0.05 ).Comprehensive evaluation of long-term efficacy of WHO criteria:efficiency rate was 97.6%(41/42).Conclusions CT-guided percutaneous vertebroplasty for vertebral hemangioma can effectively relieve the pain caused by vertebral body to increase stability,improve motor function,which is a method with safe,minimally invasion,effect,and is superior than C-arm fluoroscopy.
6.Expression of nm23 mRNA,p16 in human lung cancer and their significance on tissue microarray

Purpose:To explore the relationship between the expression of nm23mRNA,p16 and the oncogenesis,development and metastasis of lung cancer.Methods:Expressional level of nm23mRNA,p16 was detected in 72 human lung cancer samples and 23 normal tissues adjacent to cancer respectively,and their correlation with lymph node metastasis and the degree of cell differentiation was studied by immunohistochemistry (IHC) and in situ hybridization (ISH) methods on tissue microarray.Results:The positive expression for nm23mRNA in cancer samples,in human tissues adjacent to cancer and in primary cancer with lymph node metastasis was 55.6%(40/72),82.6%(19/23),25.0%(6/24) respectively.The positive expression for p16 was 45.8%(33/72),73.9%(17/23),25.0%(6/24) respectively.The expression level of nm23mRNA and p16 were closely related to the degree of cell differentiation( P 0.05).Conclusions:These results suggest that nm23 gene and p16 gene are closely related to the degree of cell differentiation and lymph node metastasis of lung cancer.The lower expression level of nm23mRNA and p16 predicts the metastasis of lung cancer, and the two genes may be an index in monitoring the status and evaluating the prognosis of the lung cancer.

9.Effect of miR-21 on migration and invasion ability in human laryngeal squamous carcinoma cell Hep 2

Objective To explore the effect of microRNA‐21(miR‐21) on the migration and invasion ability in human laryn‐geal squamous carcinoma cell Hep2 .Methods The MTT method was used to detect the viability of Hep2 cells at 48 h after miR‐21 inhibitor and miR‐21 NC transferring into Hep2 cells by LipofectamineTM 2000 .The cell migration ability was detected by using the scratch test .The cell invasion ability was detected by using the Transwell method .The activation of phosphatase and tensin homo‐logue deleted on chromosome 10 (PTEN)/phosphatidylinositol 3 kinase (PI3K) /protein kinase B(Akt) signal pathway and the expression of matrix metalloproteinase 2 (MMP2) ,MMP9 ,reversion inducing cysteine rich protein with kazal motif (RECK) was detected by using the Western blotting .Results Compared with miR‐21 NC ,miR‐21 inhibitor could significantly reduce the Hep2 cellviability[(0.688±0.043)vs.(0.375±0.012)],inhibitedthemigrationability[(6.57±0.02)μm vs.(20.49±2.18)μm]and invasion ability[(100 .7 ± 10 .2) vs .(46 .8 ± 4 .3)] ,and the differences were statistically significant (P<0 .01) ,meanwhile miR‐21 inhibitor could down‐regulate the expression of PI3K ,MMP2 and MMP9(P<0 .01) ,and reduced the phosphorylation level of Akt (P<0 .01) ,up‐regulated the expression of PTEN and RECK (P<0 .01) .Conclusion miR‐21 inhibitor can significantly suppress the migration and invasion ability of Hep2 ,which may be related with the PTEN/PI3K/Akt signal pathway .
10.Clinical Observation of Tripterygium Glycosides Combined with Mizolastine in the Treatment of Dermatitis and Eczema

OBJECTIVE:To observe the efficacy and safety of tripterygium glycosides combined with mizolastine in the treatment of dermatitis and eczema. METHODS:138 patients with dermatitis and eczema were randomly divided into control group (69 cases) and observation group (69 cases). Control group received Mizolastine sustained release tablet 10 mg,orally,once a day. Observation group additionally received Tripterygium glycosides tablet 20-30 mg,orally taking after a meal,3 times a day. All patients treated for 3 weeks,patients’life way remained unchanged during treatment. Clinical efficacy,total score of symptoms, IL-2,IL-6,CRP levels and the incidence of adverse reactions in 2 groups were observed. RESULTS:The total effective rate in observation group was significantly higher than control group,with significant difference (P<0.01). After treatment,the total score of symptoms,IL-2,IL-6,CRP levels in 2 groups were significantly lower than before,and observation group was lower than control group,with significant differences(P<0.05 or P<0.01). The incidence of adverse reactions in observation group was significantly lower than control group,with significant difference (P<0.05). CONCLUSIONS:Tripterygium glycosides combined with mizolastine shows better efficacy than mizolastine alone in the treatment of dermatitis and eczema,with better safety.
