1.Testing the Human Serum for HBsAg by Ultrasensitive Chemiluminescent Photographic Detection-Dot-ELISA Technique

The most new ultrasensitive chemiluminescent photographic detectiondot-ELISA (CPD-Dot-ELISA) technique was developed by combining the chemiluminescence technique with Dot-ELISA. The sensitivity of testing for pure HBsAg by CPD-Dot-ELISA was 30 and 60 times higher than by general Dot-ELISA and plate ELISA, respectively. 243 clinical serum specimens had been tested for HBsAg by both plate ELISA and CPD-Dot-ELISA, indicating that of 243 specimens tested for HBsAg by the former, 149 were positive, while of 243 specimens tested for HBsAg by the latter, 194 were positive. Of 149 positive specimmens tested by the former, only Ⅰ wasn't detected by the latter. 14 specimens randomly sampled from the additional 45 positive serum specimens detected by the latter, and 2 serum specimens which proved to be positive by both methods, had then been subject to neutralized test for HBIG, indicating that all 16 mentioned above specimens were positive. The results showed that the CPD-Dot-ELISA technique not only had its high sensitivity, good specificity and repro-ducibility, but also it was simple in manipulation, economically practical and worthy to be widely spread.

Early onset narcolepsy is a chronic nervous system disease with unknown origin,which has gained nationwide attention in recent years.Children with this disease often experience atypical clinical symptoms.The review summarizes existing studies on early onset narcolepsy with regard to its atypical clinical symptoms,pathogenesis,diagnosis,and treatment.
3.Quantitative assessment of segmental ventricular wall motion abnormalities in patients with coronary artery heart disease

Objective To assess segmental ventricular wall motion abnormalities in patients with coronary artery heart disease by velocity vector imaging (VVI).Methods Eight-six patients of chest pain having underwent coronary angiography were selected,and were divided into myocardial ischemia group (31 cases),myocardial infarction group(25 cases) and normal group (30 cases) according to test results.All the patients were checked with ultrasonic cardiogram in specified time.Systolic rotation angle and speed,diastolic rotation speed in the bottom and apex heart of each segment (anterior,anterior septum,posterior,lateral,inferior wall,interval) was analyzed by VVI.Results Rotary motion of the left ventricle was not consistent in different segment in myocardial infarction group,and the extent and form was different compared with that in normal group.Compared with that in normal group,bottom and apex heart angle of rotation of each segment in myocardial infarction group showed significant difference (P < 0.01 or < 0.05).The rotation angle of each segment in apex and bottom heart between myocardial ischemia group and normal group had no statistical significance (P >0.05).In bottom heart:the systolic rotation speed on anterior wall,anterior septum,posterior wall and lateral wall in myocardial infarction group was statistically lower than that in normal group (P < 0.01 or < 0.05),and the diastolic rotation speed on anterior wall,anterior septum,posterior wall,lateral wall,inferior wall was statistically lower than that in normal group (P < 0.01 or < 0.05).The systolic rotation angle between two groups was not statistically significant (P > 0.05).The diastoic rotation speed on anterior interval septum in myocardial ischemia group was statistically lower than that in normal group (P < 0.05),the systolic rotation angle and speed,diastolic rotation speed in other segment was not statistically significant compared with that in normal group (P > 0.05).In apex heart,the systolic rotation speed in anterior wall,anterior septum,posterior wall in myocardial infarction group was statistically lower than that in normal group (P < 0.01 or < 0.05),and the diastolic rotation speed in anterior wall,posterior wall,lateral wall,inferior wall was statistically lower than that in normal group (P < 0.01 or < 0.05).The diastolic rotation speed on anterior and inferior wall in myocardial ischemia group was statistically lower than that in normal group (P < 0.05),and the systolic rotation angle and speed,diastolic rotation speed in other segment was not statistically significant (P > 0.05) compared with that in normal group.Conclusion VVI technique in ultrasonic cardiogram can check out difference between myocardial ischemia and myocardial infarction incipiently.
4.Research in operation room nurses' job stress and mental health status

Objective To understand the operation room nurses' job stress and mental health status and discuss the countermeasures to ease pressure. Methods 82 patients were selected from five hospitals from April to August 2010. We used symptom checklist, self-rating anxiety scale, self-rating depression scale and nurse' job stress scale to carry out the questionnaire survey. Results The pressure of operation room nurses were related with their mental health status. Conclusions Some problems existed in mental health and work stress in operation room nurses, which should be highly valued by operation room nursing managers.
5.Research Progress in Membrane Fusion of the Respiratory Syncytial Virus.

The respiratory syncytial virus (RSV) is one of the most common causes of acute infection of the lower respiratory tract among children. For viruses in the Paramyxoviridae subfamily, membrane fusion requires a specific interaction between two glycoproteins: the fusion protein and attachment protein. Membrane fusion of the RSV appears to be unique among paramyxoviruses in that fusion is accomplished by the fusion protein alone without help from the attachment protein. Here, we review recent achievements and advances in the study of membrane fusion triggered by the RSV published in high-impact-factor journals. We also review and make a comparative analysis of the popular hypotheses regarding membrane fusion of the RSV. Finally, we discuss the "hot topics" in current research and controversial data published in recent years in the hope of providing references for Chinese researchers.

6.Effect of hepatitis C virus infection on the expressions of multiple sialyltransferases and galactosyltransferases in Huh7.5.1 cell

Objective To investigate the altered glycotransferases and glycan profile in HCV-infected cells, the expression levels of 35 kinds of sialyltransferases and galactosyltransferases were evaluated in hepatitis C virus (HCV) infected Huh7.5.1 cell cultured model.Methods Western blot was used to measure the expressions of HCV-nonstructural protein NS3 and and quantitative real-time PCR (qRT-PCR) was performed to determine HCV-RNA levels and mRNA levels of 35 kinds of sialyltransferases and galactosyltransferases.Lectin microarray was used to compare the glycan profiles of sialyltransferases-and galactosyltransferases-associated glycan-profile in Huh7.5.1 cells and HCV infected Huh7.5.1 cells.Results Among the 35 kinds of sialyltransferases and galactosyltransferases, the mRNA levels of four sialyltransferases (including ST3Gal Ⅲ, ST6GalNAC Ⅲ, ST8Sia Ⅲ and ST8SiaⅣ) and five galactosyltransferases (including B3GALT1, B3GALT 2, B3GALT3, B3GALT4 and B4GALNT2) were found to be 11-45 fold higher in HCV-infected cells compared with naive Huh7.5.1 cells.The up-regulated level of B3GALT 1 was the most evident (45-fold change), followed by ST8Sia Ⅳ and ST8Sia Ⅲ, with 39-and 37-fold respectively.Consistently, lectin microarray showed that glycans recognized by EBL (binding terminal sialic acid, Neu5Acα6Gal, 1.97-fold change), ECA letin (binding terminal galactose, Galβ1,4GalNAc,4.3-fold change), and ACA(binding terminal galactose, Galβ1-3GalNAc, 3-fold change) were elevated in glycoprotein products of sialyltransferase and galactosyltransferase respectively.Conclusion HCV infection causes the increased expression levels of mutiple sialyltransferases and galactosyltransferases in Huh7.5.1 cell line and hence the upregulation of glycan profiles of the glycoproteins.These results provide potential therapeutic targets and diagnostic markers for HCV infection and insights on the molecular mechanism of HCV infection.

MicroRNAs (miRNAs) are a class of small endogenous RNAs, which handle post-transcriptional gene regulation activity through targeting messenger RNAs.As the deep understanding of them, miRNAs had been found to participate in numerous basic biological processes, including the normal growth process of tissues and organs ,as well as the pathologic course of diseases.Different miRNAs may be involved in different tissues and organs, the same miRNA may have different expression in various physiological process or stages of pathological conditions.Now, the recent studies about the miRNAs in the lung development and pulmonary diseases were reviewed.
8.The influence of ovarian artery on the outcomes of uterine fibroid embolization

Although many studies have confirmed that uterine arterial embolization has good short-term and long- term effects for the treatment of uterine fibroids, certain failure rate does exist. Many researches have indicated that the surgical failure is mainly related to the collateral arterial supplies, among which the ovarian artery supply is the commonest cause. This review aims to make a further understanding of the influence of ovarian artery on the outcomes of uterine fibroid embolization.
9.Effects of bone marrow mesenchymal stem cells on the biological behavior of prostate cancer cells

BACKGROUND:Bone marrow mesenchymal stem cel s have a close relationship with tumor occurrence and development, which are not only involved in tumor recruitment but impact the biological behavior of tumor cel s. OBJECTIVE:To investigate the effect of bone marrow mesenchymal stem cel s on the biological behavior prostate cancer cel line. METHODS:Human bone marrow mesenchymal stem cel s from healthy adults were col ected and cultured in vitro. Cel morphology and cel markers were observed. Using the Transwel technique, human dermal fibroblasts HDF-a were co-cultured with human prostate carcinoma DU145 cel s in positive control group, wild-type prostate carcinoma DU145 cel s cultured alone in negative control group, and bone marrow mesenchymal stem cel s co-cultured with human prostate carcinoma DU145 cel s in experimental group. Flow cytometry assay was used to detect the cel cycle of prostate carcinoma DU145 cel s, soft agar colony formation assay performed to detect cel proliferative ability, and crystal violet staining used to observe DU145 cel migration at 3 days after culture. RESULTS AND CONCLUSION:Compared with the negative control group, cel proportion at G 2/M+S phase, number of transmembrane cel s and colony counting were significantly decreased in the experimental group. These findings indicate that bone marrow mesenchymal stem cel s may inhibit the proliferation and invasion of prostate carcinoma DU145 cel s by shortening the G 2/M+S phase, thereby providing a new insight into the treatment of human prostate cancer.

Cytochrome P450 is an important kind of enzymes for the metabolism of drug and other endo-or xeno-biotics. Classification of metabolic enzymes, molecular biological characteristic of CYP3A4, CYP2D6, CYP2C9, CYP2C19, CYP2E1, CYP1A2 and CYP2A6 were described in this review. Influence of traditional Chinese medicine(TCM) on metabolic enzyme and its application in clinical drug therapeutics and drug development were also reviewed in this article.
