2.T cells harboring anti-CD20 scFv-CD8-TCR? fusion gene in treatment of human B-cell lymphomas: an experimental study
Academic Journal of Second Military Medical University 2000;0(07):-
Objective:To observe the anti-tumor effect of T lymphocytes harboring anti-CD20 scFv-CD8-TCR? fusion gene on human B lymphomas in vitro and in vivo,so as to explore the feasibility of CD20-mediated autogenous T lymphocytes in killing B-cell lymphomas. Methods: A fusion gene containing anti-CD20 scFv,CD8 molecule and CD3? chain was constructed and was cloned into pcDNA3.After confirmed by restriction endonuclease analysis, the fusion gene was used to transfect the human peripheral T lymphocytes through electroporation and expression of anti-CD20 scFv-CD8-TCR? fusion protein was induced. The CD20 antigen-recognition ability of transfected T cells was determined by flow cytometry. The killing effect of transfected T cells on B cell lymphomas-Raji cells was tested in a cytotoxicity assay. The antitumor efficacy of this gene-modified T cell against the Raji tumor cell line was also evaluated in BALB/c nude mice. Results: We successfully constructed the anti-CD20 scFv-CD8-TCR? fusion gene and expressed its protein on T cells.Flow cytometry and cytotoxicity assay demonstrated that the gene engineered T cells specifically recognized CD20 antigen and specifically inhibited B-cell lymphomas Raji cells.Furthermore, the T cells significantly inhibited the transplanted Raji cells in irradiated BALB/c nude mice. Conclusion: T lymphocytes transfected with anti-CD20 scFv-CD8-TCR? fusion gene have antigen-specific anti-tumor effect in vitro and in vivo, which lays a foundation for utilizing human T lymphocytes to treat human B-cell lymphomas.
3.Risk Factors and Antibiotic Resistance of Lower Respiratory Tract(LRT) Fungal Infection in 76 Hospitalized Patients
Chinese Journal of Nosocomiology 2006;0(05):-
OBJECTIVE To explore the main risk factors of fungal infection in lower respiratory tract(LRT) of hospital patients, strains distribution and sensitivity to common antifungal agents so as to provide basis for earlier diagnosis and effective treatment.METHODS Retrospective analysis was taken of the 76 cases with fungal infection in LRT in patients of Taizhou Hospital from Jul 2004 to Jun 2006 based on the diagnostic criteria for nosocomial infections by Ministry of Health of PRC and also following the definition given by European Organization for Research on Treatment of Cancer(EORTC) and us Mycosis study Groap(MSG) in 2002.RESULTS Of the 76 cases,84.2% were over 60 years old.All of them had underlying diseases.Sixty cases had been repeatedly treated with 2 or 5 antibiotics before fungal infection;21 cases received glucocorticoid;6 cases had chemol and radiotherapy and 21 cases underwent invasive treatment procedures. In the three years,77 strains of fungi were isolated from all kinds of samples.Susceptibility testing indicated that the resistance to ketoconazole and miconazole was higher compared with that to fluconacole and itraconazole.CONCLUSIONS Fungal infection in LRT of hospital patients is closely related with age, long stay in hospital, wide administration of broad-spectrum antibiotics,immuno-suppressants and cytotoxic drugs. The most common fungi are Aspergillus and Mucor. There is an increasing number of strains of fungi resistant to drugs, especially to azole antifungal agents.Effective counter measures should be taken without delay.
5.Imaging Findings of Vertebral Eosinophilic Granuloma(An Analysis of 10 Cases)
Journal of Practical Radiology 2001;0(08):-
Objective To improve the understanding of the radioactive charcteristics of vertebral eosinophilic granuloma. Methods The imaging features (X-ray?CT and MRI) of 10 cases with vertebral eosinophilic granuloma were retrospectively analyzed.Diagnoses of all the patients had been confirmed with pathlolgical examination after operation. Results All the patients had vertebral body damages,7 with osteolytic damages and 3 with cystic damages.Intervertebral crevice became narrowing or disappearing .Some vertebral appendix and paravertebral soft tissue were involved. Conclusion It is out of proportion among the vertebral body damages, the involved vertedral arch, the changes of intervertebral crevice and the mass of paravertebral soft tissue. The remaining bone fragment without sequestrum in the damage ares is considered as the main characteristic expression of vertebral eosinophilic granuloma.
6.Appearance of lipid storage in muscle fiber in neuromuscular disorders
Chinese Journal of Rehabilitation Theory and Practice 2004;10(3):185-187
ObjectiveTo compare the appearance of abnormal lipid accumulation in skeletal muscle fibers in various neuromuscular disorders by analyzing the results of Oil-Red-O staining of muscle biopsy samples. Methods523 muscle biopsies performed in 1998—2002,consisted of lipid storage myopathy, mitochondrial diseases, inflammatory myopathy, muscular dystrophy, steroid myopathy, alcoholic myopathy and neurogenic muscle atrophy, were studed. ResultsType Ⅰ fibers were selectively involved in lipid storage of muscle fibers. In all diseases, men were more frequently affected than momen in which men to women was 3∶2.Marked uniformity lipid storage in muscle fibers was found most frequently in patients with lipid storage myopathy and mitochondrien-lipid-glycogene storage diseases. Moderate uniformity lipid storage in the muscle fibers was found in patients with steroid myopathy, alcoholic myopathy and malnutrition. Moderate or marked non-uniformity lipid storage in muscle fibers was found in patients with mitochondrial cytopathy, inflammatory myopathy, muscular dystrophy. Moderate non-uniformity lipid storage in muscle fibers was found in patients with neurogenic muscle atrophy. Mild lipid storage in muscle fibers was found in numerous systemic disease and neuromuscular disorders. ConclusionNumerous neuromuscular diseases can associated with abnormal lipid metabolism in muscle fibers, in which the men are more involved than women. The appearance of lipid accumulation in the muscle fibers markedly changes in different neuromuscular diseases. The diagnostic value of lipid storage in muscle fibers should be considered under analysis clinical symptoms and other myopathological findings.
7.Difference of Respiratory Function Between Spleen-qi Deficiency Pattern and Lung-qi Deficiency Pattern in Chronic Obstructive Pulmonary Disease
Journal of Guangzhou University of Traditional Chinese Medicine 2001;0(03):-
To explore the difference of respiratory muscle strength and respiratory drive between lung qi deficiency pattern and spleen qi deficiency pattern in chronic obstructive pulmonary disease (COPD).Forty two cases of COPD were differentiated as lung qi deficiency pattern (n=23, Group A) and spleen qi deficiency pattern(n=19, Group B) and twenty healthy volunteers served as control (Group C). Pulmonary ventilatory function such as forced vital capacity(FVC), forced expiratory volume in one second (FEV 1), FVC/ FEV 1 and maximum voluntary ventilation (MVV) were examined; and the maximum inspiratory pressure (MIP), maximum expiratory pressure (MEP) and oral blocking pressure were also detected. As compared with Group C, pulmonary ventilation was decreased in both Group A and Group B and more obviously in Group B (P0 05); MIP and MEP in Group B differed from those in Group C (P
8.Vertebral artery stenosis and its treatment
International Journal of Cerebrovascular Diseases 2012;20(9):706-711
Vertebral artery stenosis (VAS) is often caused by atherosclerosis. The clinical diagnosis is more difficult because the symptoms are not typical, and the non-invasive examination is not easy to be detected. The annual stroke incidence is higher in patients with VAS,particularly the symptomatic severe vertebral artery stenosis.About 20% of posterior circulation strokes are caused by VAS.This article reviews the epidemiology, natural history, diagnosis and clinical treatment of VAS.
