6.Protective effects of zileuton on the acute lung injury induced by focal cerebral ischemia-reperfusion in rats
Chinese Pharmacological Bulletin 2003;0(11):-
Aim To investigate the protective effects and mechanisms of a 5-lipoxygenase (5-LO) inhibitor zileuton on the acute lung injury (ALI) induced by focal cerebral ischemia-reperfusion injury (CIRI) in rats. Methods The right middle cerebral artery in rats were occluded by inserting a thread through internal carotid artery for 2 h,and then reperfused for 24 h.Zileuton(10,50 mg?kg-1) was orally administered 2h before ischemia and 0,5,10 h after reperfusion.Morphological changes of the lung tissue were observed by hematoxylin-eosin (HE)staining technique. Lung wet/dry weight ratios were detected to observe the water content of the lung. The mRNA of cysteinyl leukotrienes recepor1 (CysLTR1) was detected by RT-PCR. The content of Leukotriene B4 (LTB4) in serum was measured by enzyme linked immunosorbent assay (ELISA) technique. The expression of TNF-? protein was measured by immunohistochemical technique. The activities of MPO, T-AOC, and Na+,K+-ATPase from the lung tissue were measured by biochemical method. Results With the use of zileuton 10, 50 mg?kg-1,the pathological changes of the lung were alleviated. and the wet/dry weight in the lung tissue was reduced(P
7.HPLC Determination of Active Constituents in Xiaoerniuhuang Powder
Chinese Traditional Patent Medicine 1992;0(02):-
The reversed-phase high-performance chromatography was developed for determing the content of berberine, palmatine, emodin and chlorogenic acid in Xiaoerniuhuang Powder on ODS column. The powder samples extracted with methanol were determined separatly at their particular absorption points using external standard method. This method had advantages of perfect separatin, high sensitivity, good reproducibility and simple operation.
8.Receptor and Intracellular Signaling Mechanisms Involved in TNF?-induced Expression of Heme Oxygenase-1 in Endothelial Cells
Journal of Medical Research 2006;0(11):-
Objective To investigate the receptor involved in the TNF?induced HO-1 expression and the related signal transduction mechanisms in the brain microvessel endothelial cells. Methods TNF? receptor 1(TNFR1)knockout murine brain microvessel endothelial cells(BMVECs)in vitro culture were stimulated respectively by TNF?(5ng/ml).After 24 hours, the level of HO-1 gene mRNA were detected by semi-quantitative RT-PCR,the effect of TNF?on JNK 、ERK or transcription factor AP-1 were analyzed by Western blot, while ERK inhibitor(PD98059)or JNK inhibitor(SP600125)were used. Results Exposure of wild-type BMVECs and TNFR1(p55)knockout BMVECs [BVEC/TNF-RI(-/-)] to TNF?(5ng/ml,24 h)caused a marked increase in HO-1 mRNA expression.Furthermore,TNF? stimulation also lead to the activation of JNK,ERK and the transcription factor AP-1 in the BVEC/TNF-RI(-/-),However,incubation with the JNK inhibitor SP600125 but not the ERK inhibitor PD98095 attenuated the TNF?-induction of HO-1 in the BVEC/TNF-RI(-/-).Conclusions TNFR2 and JNK activation but not TNFR1 might be essential for mediating the TNF?-induction of the HO-1 in the endothelial cells.
9.Limit test for aristolochic acid in Herba Asari by HPLC
Chinese Traditional Patent Medicine 1992;0(06):-
AIM:To study the detection limits of aristolochic acid in Herba Asari preparation by HPLC. METHODS: Whether Asarum hetertropoides Fr.Schmidt ver.Mandshuricum(Maxim.) Kitag contained aristolochic acid A or not by HPLC and MS,and the difference in the reference substance and the sample solution was compared. RESULTS: Aristolochic acid A could not be found in A.hetertropoides Fr.Schmidt ver.Mandshuricum(Maxim.)Kitag by HPLC,but be found by MS. CONCLUSION:A.hetertropoides Fr.Schmidt ver.Mandshuricum (Maxim.)Kitag contained trace aristolochic acid, but it is difficult to measure aristolochic acid in Herba Asari by HPLC.The baseline noise of the sample solution can interfere with the detection limit of the trace compound compound
10.Clinical features of drug-induced liver injury: a review of Chinese literatures 2007-2011
Chinese Journal of General Practitioners 2013;(5):361-365
Objective To analyze the clinical characteristics of drug-induced liver injury (DILI) from Chinese literatures published in Chinese Periodical Full-text Database (CNKI) from 2007 to 2011.Methods Using drug-induced liver injury as key words, the literatures in CNKI were searched, and the data of age, gender, medication, clinical manifestation and prognosis were analyzed. Results In search of CNKI,281 literatures were collected; finally 111 relevant papers were obtained.Total 10 693 cases of DILI were included with a male/female ratio of 1.14:1 and a mean age of 45.6 y ranging from 7 months to 93 y.Among 1093 cases with the age grouped, the highest incidence of DILI was in age 41-60 y(38.52%)and 20.68% patients were over 60 y.Among 6592 cases with clinical manifestation recorded,43.51% patients were asymptomatic and abnormal liver function was detected in health check-up; and others presented nonspecific gastrointestinal symptoms, including fatigue, nausea, vomiting and jaundice. In 9340 cases with medication recorded, the 6 most common drugs inducing DILI were antituberculotics (32.74%),Chinese herbs(22.12%),antibiotics(9.18%),antineoplastics(6.34%),NSAIDs (4.80%)and antithyroid drugs (4.37%).The most common type of toxicity was hepatocellular injury (63.66%,4438/6971).In general the prognosis was good with a fatality rate of 1.67%.Conclusions Antituberculotics and Chinese herbs are the main causes of DILI.Some DILI patients have occult onset and others have nonspecific gastrointestinal symptoms. Most patients have good prognosis if they are treated timely.
