Objective To investigate the ability of immune response of cotransfection of rat breast cancer cells with MIP-1αand B7-1 .Methods Tumor growth was observed every day after the SHZ-88 breast cancer cells were inoculated in the rats .The tumor-bearing rats were randomly divided into control group ,MIP-1αgroup ,B7-1 group and combination group ,which were injected with corresponding transgenic cells .The change of tumor volume in 1~4 weeks and the survival time were observed and compared .The peripheral blood was collected .CD4+ ,CD8+ percentage and CD4+ /CD8+ ratio and the levels of IL-2 and IFN-γof the peripheral blood were detected and compared .Results The tumorigenic rate were 100% ,20% ,30% and 0% after the SD rats were inoculated with SHZ-88 ,SHZ-88/MIP-1α,SHZ-88/B7-1 and SHZ-88/MIP-1α+B7-1 cells for 10 days .After the transgenic therapy ,the tumor volume of the combination group in 1~4 weeks were significantly smaller than the other groups .The survival time of the combina-tion group was significantly longer than the other groups .The CD4+ ,CD8+ percentage and CD4+ /CD8+ ratio and the levels of IL-2 and IFN-γof peripheral blood in the combination group were significantly higher than the other groups .The differences above were all statistically significant (P<0 .05) .Conclusion Cotransfection of MIP-1αand B7-1 could induce immune response in breast cancer rats and play anti-cancer effects .

Objective To investigate the chemical constituents in the whole plant of Clerodendrum canescens. Methods The compounds were isolated by column chromatography over silica gel or Sephadex LH-20 gel and their structures were elucidated by chemical and spectroscopic methods. Results Seven triterpenoids were isolated and identified as: lupeol (Ⅰ), ?-amyrin 3-undecanotate (Ⅱ), lupeol acetate (Ⅲ), lupeol 3-palmitate (Ⅳ), melastomic acid (Ⅴ), ?-amyrin acetate (Ⅵ), betulinic acid (Ⅶ), respectively. Conclusion Among those compounds, compound Ⅱ is a new one, while compounds Ⅳ-Ⅵ are isolated from the plants of Clerodendrum Linn. for the first time.

Chronic kidney disease (CKD) is common worldwide and in Singapore. The primary care physicians play an important role in managing patients with CKD, especially in the early stages of CKD. The primary objectives of CKD management are (1) slowing down the progression of CKD, (2) managing the complications of CKD, and (3) establishing the longterm kidney care plan. The interventions to slow down the progression of CKD are to identify the cause of CKD, use the renin-aldosterone system blocker and the sodium-glucose cotransporter-2 inhibitor in suitable CKD populations, optimise blood pressure and glycaemic control, correct acidosis, avoid acute kidney injury and nephrotoxin, and modify dietary and lifestyle habits. For complications of CKD, the focus is on reducing cardiovascular risk, and managing anaemia, mineral bone disease, electrolytes imbalances and fluid overload. Lastly, there is a need to establish CKD patient’s treatment goals and initiate advanced care planning in a patient with progressive CKD to facilitate future care.

Objective To construct an RNA interference vector to down-regulate X-linked inhibitor of apoptosis(XIAP)gene and study the RNA interference effect on the cell cycle and growth of ovarian cancer.Methods Oligonucleotides of 64 base pairs for hairpin RNA targeting XIAP were designed, chemically synthesized,annealed,and cloned into the pSUPER vector.After identification by restriction digestion,the correct vectors were transiently transfected into SKOV3 cells,a human ovarian cancer cell line.The XIAP mRNA was detected by RT-PCR.The proteins were detected by western blot and indirect immunofluorescence staining.Flow cytometry(FCM)analysis and methyl thiazolyl tetrazolium(MTT)assay method were applied to measure cell cycle,cell growth and sensitiveness to cisplatin.Results SKOV3 cells had a high level expression of XIAP.The vector of RNA interference,which can interfere with XIAP gene was successfully constructed.After transient transfection,the expression of XIAP protein was significantly decreased in SKOV3 cells and the value of relative density was 3584?124,2138?65,1973?80 and 110 ?12,respectively(P=0.0334).At the same time,the expression of XIAP mRNA was decreased accordingly and the value of relative density was 6674?274,4532?107,2322?57 and 1864?78, respectively(P=0.0127).The FCM results showed that,the vector could increase the number of cells in G_1 phase compared with parent cells and compared with the cells transfected with pSUPER(P

Objective: To establish a method for the determination of 17 heavy metals and harmful elements in Xiyanping Injection (XI). Methods: The samples were digested and determined by inductively coupled plasma mass spectrometry (ICP-MS). The RF power was 1 550 W; The peristaltic pump speed was 0.3 r/s; The plasma gas velocity was 15 L/min; The auxiliary gas velocity was 0.2 L/min; The carrier gas velocity was 1 L/min; The sampling depth was 10 mm and the signal was measured for three times. Results: The detection limits of Li, Al, V, Cr, Fe, Co, Ni, Cu, As, Ag, Cd, Sn, Sb, Ba, Hg, Pb, Bi were 9.584, 49.858, 0.504, 3.016, 51.209, 0.142, 1.116, 0.675, 0.924, 1.421, 0.403, 2.770, 0.711, 3.584, 0.590, 0.411, 0.169 ng/mL, and the quantitative limits were 28.933, 151.085, 1.528, 9.139, 155.179, 0.429, 3.381, 2.046, 2.799, 4.312, 1.220, 8.394, 2.155, 10.861, 1.965, 1.244, and 0.513 ng/mL within a certain concentration range, respectively. The response value had a good linear relationship with the concentration (r > 0.999 1); The RSDs (n = 12) of the injection precision test were in the range of 0.8%-3.8%, the RSDs (n = 6) of the repeatability were in the range of 0.7%-2.0%, and the average recoveries of 17 elements were in the range of 95.7%-104.8%. Detection results of 20 batches of mass production samples: Ag was not detected, Li and V were detected but lower than the quantitative limit, the maximum mass concentration of Al, Ni, Cu, As, Ba, Hg and Pb were 0.685, 0.013, 0.007, 0.006, 0.208, 0.070 and 0.027 μg/ml respectively, Cr, Fe, Co, Cd, Sn, Sb, and Bi were not detected or lower than the quantitative limit. Conclusion: The specificity, linearity, sensitivity, precision, accuracy and durability of the method were all good. It can be used to determine 17 elements of Li, Al, V, Cr, Fe, Co, Ni, Cu, As, Ag, Cd, Sn, Sb, Ba, Hg, Pb and Bi in XI. The test results of 17 elements in 20 batches of mass production samples were less than the limit requirements, meeting the requirements.

OBJECTIVE: To develop a method for the determination of ethanol extracts content of Pogostemon cablin (Blanco) Benth. using near-infrared spectroscopy. METHODS: The ethanol extracts contents of 139 samples were determined by the method of ethanol cold-soak extraction. Near-infrared spectrograms were obtained, then multiplicative signal correction, first derivative method and Savitzky-Golay filter method were used to process the spectrograms, at last the calibration model of ethanol extracts content was established by partial least squares regression analysis method. RESULTS: The correlation coefficients, root-mean-square error of calibration, root-mean-square error of prediction and root-mean-square error of cross-validation of the calibration model of ethanol extracts content were 0.98958, 0.590, 0.623 and 0.97518, respectively. CONCLUSION: The method is stable, accurate, and can be used to predict ethanol extracts content of Pogostemon cablin (Blanco) Benth. rapidly.

Autophagy, a cellular housekeeping process, is indispensable to controlling the homeostasis of cytoplasm by removing unused proteins and damaged cell organelles. This process involves different types of human diseases, including cancers, neurodegenerative diseases and infectious diseases. Neurodegeneration is a critical pathological process of many eye diseases, such as glaucoma and age-related macular degeneration. The retina and all intraocular cells are constantly exposed to environmental stress and injuries, including oxidative stress and starvation, which lead to autophagy. Autophagy promotes cell survival through the recycling of metabolic precursors, or promotes cell death if autophagy is over-active. Additionally, autophagy and apoptosis have been shown to be harmonious or contrasting, depending on different experimental contexts. All of this contributes to the pathogenesis of many diseases. This paper reviews the mechanisms and regulation involved in autophagy, current understandings of neuronal autophagy in glaucoma and retina and strategies for therapeutic modulation.
