Acta Universitatis Medicinalis Anhui 2017;52(6):892-896
Objective To explore the influence of Lipo PGE1 on endothelin (ET-1) and interleukin (IL-6) in exhaled breath condensate(EBC) of patients with chronic obstructive pulmonary disease (COPD) and pulmonary hypertension(PH) and its clinical significance. Methods A total of 40 cases of patients with COPD and PH were randomly divided into thecontrol group and the treatment group,20 cases in each group. The control group was administered with the conventional treatments such as anti-infection,bronchodilator,antiasthma,expectorant and oxygen therapy;the treatment group was administered with Lipo PGE1 (10 μg/d,iv,for 10 days) besides the conventional treatments.ET-1 and IL-6 in EBC,pulmonary artery systolic pressure(PASP),arterial blood PaO2 and PaCO2,lung function FEV1/FVC,FEV1%pred in both groups were assayedbefore and after the treatment. Results There were no statistical difference between the two groups in the ET-1 and IL-6 in EBC,PASP,arterial blood PaO2 and PaCO2,FEV1/FVC,FEV1% pred before the treatment. After the treatment, the ET-1,IL-6 in EBC,PASP and arterial blood PaCO2 of the treatment group were lower than thoseof the control group (P<0.05);the arterial blood PaO2,FEV1/FVC,FEV1% pred of the treatment group were higher than those of the control group (P<0.05);The levels of ET-1 and IL-6 in EBC wereboth positivelycorrelated with PASP in the two groups. Conclusion Lipo PGE1 can reduce the levels of ET-1 and IL-6 in EBC of patients with COPD and PH.ET-1 and IL-6 may become a curative effect judgment index of COPD and PH,which has a certain clinical significance.
5.Dynamic investigation of cytokines in mice immunizedwith Echinococcus granulosus
Journal of Xi'an Jiaotong University(Medical Sciences) 2017;38(5):639-642,648
Objective To investigate the dynamic changes of IL-2, IFN-γ, TNF-α, IL-4, IL-5 and IL-10 in mice immunized with Eg14-3-3 and then challenged by Echinococcus granulosus protoscoleces. Methods ICR mice were subcutaneously immunized every two weeks for altogether three times with rEg14-3-3, followed by the challenge with Echinococcus granulosus protoscoleces intraperitoneally four weeks after the third immunization. The PBS control group mice were operated in the same way.We collected mouse serum from tail vessel at different time points after immunization and challenge. The serum levels of IL-2, IFN-γ, TNF-α, IL-4, IL-5 and IL-10 were examined with enzyme-linked immunosorbent assay. Results The levels of the six cytokines were higher after immunization and challenge infection in rEg14-3-3 group than in PBS control group. The immunized mice generated a great deal of Th1 cells, namely, IL-2, IFN-γ and TNF-α, after immunization and showed a peak at week 10 (acute infection phase), then the level decreased rapidly at week 30 (chronic infection phase), and maintained a high level for a long time.In contrast, Th2 cells like IL-4, IL-5 and IL-10 kept a low level until week 18, peaked at week 30, and then decreased gradually but maintained a relatively high level for a long time. For PBS control group, IL-2, IFN-γ and TNF-α did not increase obviously before infection, but increased rapidly after challenge infection and peaked at week 18;then the level decreased gradually and maintained a high level for a long time.On the other hand, IL-4, IL-5 and IL-10 kept a low level, but increased gradually after challenge infection.IL-4 peaked at week 18 while IL-5 and IL-10 peaked at week 30, and then they all decreased slowly but maintained a relatively high level for a long time. Conclusion Eg14-3-3 can induce significant antibody response by Th2 cells and cell-mediated immunity response by Th1 cells. Both Th1 and Th2 cells participate in the anti-echinococcusis protective immunity.
6.Application of fetal DNA in maternal plasma in noninvasive prenatal diagnosis.
Journal of Huazhong University of Science and Technology (Medical Sciences) 2004;24(1):59-61
To explore the application of fetal DNA in maternal plasma for noninvasive prenatal diagnosis, the DNA template was extracted by hydroxybenzene-chloroform from 44 maternal (7-41 weeks) plasma. The Fetus-derived Y sequence DYZ-1 gene (149bp) was chosen to be amplified by PCR. The fragment was identified in all the plasma of male bearing pregnant women with the diagnostic accordance rate being 100.00%. Two of the 22 female bearing pregnant women had false positive results. Among the 44 pregnant women, the diagnostic accordance rate was 88.89% at early pregnant stage, 100.00% at medium pregnant stage, and 96.55% at late stage respectively. The final accuracy of 95.45% was obtained in all cases. It was concluded that by means of hydroxybenzene-chloroform extraction the authors of this article promoted the concentration and purity of the DNA template, and diagnosed more accurately. The results showed that free fetal DNA in the maternal plasma could be regarded as the gene resource for noninvasive prenatal diagnosis.

Chinese Journal of Biochemical Pharmaceutics 2014;37(7):31-33,36
Objective To investigate the effect of puerarin on preeclampsia-like rat placental tissue hypoxia inducible factor (hypoxia-inducible factor,HIF)-1αexpression and study the mechanism of protective effects in rats. Methods 60 SD rats were randomly divided into control group, drugs intervention groups(low-dose,middle-dose,high dose)and model group. The rats in drugs intervention groups and model group were adopted endotoxin for induction of preeclampsia model. On the basis of the model group, low, medium and high dose groups were given 20.0,40.0,60.0 mg/kg puerarin by intraperitoneal injection, respectively. The blood pressure of tail artery, urine protein concentration were observed at different points and the expression of HIF-1αwere detected by PT-RCT in each group. Results The systolic blood pressure ,urine protein and HIF-1αprotein in model group were higher than control group(P<0.05 );serum NO concentration was lower than control group (P<0.05 );after the puerarin intervention, systolic blood pressure and urine protein concentration in drugs intervention groups were lower than model group (P<0.05 ),plasma NO concentration was higher than model group (P<0.05 )and HIF-1αprotein was lower than model group (P<0.05 );the placenta weight, weight and body length of newborn rats in drug intervention group were higher than model group (P<0.05 ).Conclusion Puerarin can reduce systolic blood pressure and urine protein levels, promote the level of NO in plasma and reduce HIF-1αlevel in placenta of rats.Puerarin can offer protection to rats with preeclampsia by decreasing the expression of HIF-1αin placental.
8.Diagnostic value of point-of-care lung ultrasonography
Chinese Pediatric Emergency Medicine 2015;22(6):375-378,382
Lung ultrasound(LUS)has gained increasing popularity among intensivists in recent dec-ades.Although limited by the presence of air,LUS has proved to be useful in the evaluation of a number of different acute and chronic conditions, such as pneumonia, pneumothorax, acute lung injury,alveolar-intersti-tial syndrome, pleural effusion, etc, and showed a relatively higher sensitivity, specificity and diagnostic accu-racy compared to routine radiographic imaging. It is also critical in improving the safety of pleural interven-tional procedures. The most common signs for diagnosing and differential diagnosing include the pleural line, lung sliding, the A-line,the quad sign, the sinusoid sign, the fractal, and tissue-like sign, the B-line,lung rock-ets,abolished lung sliding with the barcode sign, the lung point, the dynamic air bronchogram, etc. The BLUE-protocol allows differential diagnosis of acute respiratory failure.LUS is a feasible technique that can be used routinely and integrated into clinical practice in the fields of intensive care, emergency care,respirato-ry and general medicine.
