1.In vitro biological characteristics and immune regulation defects of bone marrow-derived mesenchymal stem cells from patients with aplastic anemia

BACKGROUND: Previous research has indicated that bone marrow-derived mesenchymal stem cells had immunological regulation and hematopoiesis role. There were significant differences in immunological regulation and hemopoietic function between patients with aplastic anemia and normal cases. OBJECTIVE: To summarize the in vitro biological characteristics and its immune regulation defects of bone marrow-derived mesenchymal stem cells from patients with aplastic anemia.METHODS: A computer-based online search was conducted in PUMMED (1987-2009) and VIP database (1989-2009) with the key words of "mesenchymal stem cell, bone marrow" in both Chinese and English. There were 55 articles in total. Articles which were related to biological characteristics and its immune regulation defects of bone marrow-derived mesenchymal stem cells were included; however, duplicated articles were excluded. Finally, 32 articles were included, containing 3 reviews in English, 23 original articles in English, and 6 original articles in Chinese. RESULTS AND CONCLUSION: Bone marrow-derived mesenchymal stem cells had the capacities of high proliferation, self-renewal and multilineage differentiation; in addition they had the roles of supporting hematopoiesis, promoting implantation and hematopoietic reconstitution in vivo. Bone marrow-derived mesenchymal stem cells still had the effects of negative immune regulation and reducing the incidence of graft-versus-host disease (GVHD). Aplastic anemia correlated with hematopoietic stem cells in the pathogenesis of intrinsic defects in the proliferation or differentiation of hematopoietic microenvironment and immune system abnormalities and other related disorders. Bone marrow-derived mesenchymal stem cells of patients with aplastic anemia played an important role in the pathogenesis of this disease.

Epigenetics has become a hot topic in biomedical research in last a few years.Epigenetics refers to as epigenome,and it is interpreted as epigenetic changes (DNA methylation,histone modifications and non-coding RNAs such as microRNAs) in the regulation of genes expression on chromatin-based pathways rather than a single gene.Epigenetic factors such as DNA methylation,histone modification and miRNA serve as an interface for environmental stimuli and interacts each other to regulate gene expression and control phenotypes of cell.All of those epigenetic factors are essential in the keeping homeostasis of human body and contribute to many biologic conditions.In addition,the application of epigenetic drugs has been come into clinical trials especially in the treatment of cancers,therefore the understanding the mechanism of epigenetics in the development of human diseases and the therapeutic potential of epigenetic regulators in the treatment of human disease will bring us into the epigenetics era in biomedical research.

5.Effect and mechanism of jianpi qinghua recipe on renal functions of adriamycin-induced nephropathic rats from the angle of inhibiting renal fibrosis.

OBJECTIVETo explore the effect of Jianpi Qinghua Recipe (JQR) on renal functions of adriamycin-induced focal segmental glomerular sclerosis (FSGS) rats from the angle of activating fibroblasts to myofibroblast (MyoF).
METHODSTotally 56 rats were randomly divided into the normal control group (n=8), the sham-operation group (n =8), and the model group (n=40). The FSGS rat model was induced by nephrectomy of left kidney plus intravenous injection of adriamycin. Successfully modeled rats were further divided into 5 groups, i.e., the model group, the JQR group, the JPR (Jianpi Recipe) group, the QHR (Qinghua Recipe) group, and the NDQ (Niaoduqing) group, 8 in each group. Corresponding drugs were administered to rats in all groups, 2 mL each time, for 56 days. The effect of JQR on serum creatinine (SCr), urea nitrogen, 24-h urinary protein excretion, a-smooth muscle actin (alpha-SMA) mRNA, collagen type III (Col III) mRNA, fibronectin (FN) mRNA, and collagen type IV (Col IV) mRNA were observed.
RESULTSJQR could significantly lower SCr, urea nitrogen, and 24-h urinary protein excretion levels (P < 0.01), and significantly decrease mRNA levels of alpha-SMA, Col III, FN, and Col IV (P < 0.01). It was advantageous over the NDQ group. Compared with JPR, the relative expression levels of Col III mRNA and FN mRNA of JQR and QHR were significantly lower (P < 0.01).
CONCLUSIONSJQR could improve the renal function and renal fibrosis in the adriamycin-induced nephropathic model rats. Its efficacy was superior to that of NDQ. Its mechanisms might be linked with inhibiting activation of fibroblasts.

6.Effects of Arsenic Trioxide on the Tyrosinase Activity and Melanogenesis in Human Malignant Melanoma Cell Line A375

Objective To study the effects of arsenic trioxide on the tyrosinase activity and melanogenesis in human malignant melanoma cell line A375. Methods MTT was used to measure the proliferation of human malignant melanoma cell A375. The tyrosinase activity and the melanin content were measured. Results When the concentration of arsenic trioxide was lower than 0.01 ?mol/L, a positive effect was shown along with the increase of the concentration, such as cell proliferation, tyrosinase activity and up-regulation of melanogenesis. When the concentration of arsenic trioxide was above 0.01 ?mol/L, an inhibitory effect was observed. If the concentration of arsenic trioxide was above 1?mol/L, cytotoxic effect occurred. Conclusion Arsenic trioxide has diphasic effect on human malignant melanoma cell line A375.

Objective To evaluate the efficacy and safety of Prostant TM and the patients’ compliance with the treatment of chronic prostatitis of different types. Methods A multi-central,randomized,double-blind,placebo-controlled clinical trial was conducted between June 2002 and December 2002.A total of 125 patients who had been diagnosed as chronic prostatitis and classified according to NIH classification system for prostatitis were divided into two groups: the trial group treated with Prostant TM anally one pill per night for 30 days and the control group given placebo in the same way.The efficacy was evaluated by the NIH chronic prostatitis symptom index (NIH-CPSI) and the WBC count in EPS after the treatment. Results Based on leukocyte and culture results,124 evaluable patients were stratified,with 48 cases of categories Ⅱ(38.7%),45 cases of Ⅲa(36.3%) and 31 cases of Ⅲb(25.0%)with chronic prostatitis.The overall NIH-CPSI scores were averagely reduced by 10.37 points in trial group and 6.65 in control group,and the symptom scores were averagely reduced by 7.34 in trial group and 4.72 in controll group,compared with pre-treatment.Significant differences of reduction were found between the two groups(P
8.Image characteristics of renal cysts with high density

Objective To discuss the imaging characteristics of renal cyst of high density.Methods Nine cases (5 women and 4 men; mean age,48 years; age range,31-79 years) with high density renal mass(>30 HU) were reviewed.For all patients,the symptom was intermittent lumbar pain in 2 patients,gross hematuria with lumbar pain in 1 patient,gross hematuria without pain in 1 patient.The other 5 patients were detected during routine physical examination without symptom.Preoperative examination included CT,MRI and DSA.Results All the patients had solitary and unilateral high density renal cysts,which size ranged from 2 to 5 cm in diameter.The cysts displayed predominantly hypoechoic or irregular internal echoes on sonography.Excretory urography showed an unenhanced round mass without further distinguishing characteristics.Their cystic features were con-firmed on nonenhanced and contrast-enhanced CT scans.MRI showed a round mass with long T1 and long T2 signal.Angiography revealed lack of vessels around the mass.All patients were operated by partial nephreetomy.There was no recurrence in all patients during the follow-up for 1-5 years.Conclusion Renal cyst of high density has many special features of imageology.
