1.Regulatory Mechanism of Fucoidan for Disorder of Lipid Metabolism in Rats

Objective To explore the mechanism of Fudoidan for regulating the disorder of lipid metabolism in rats. Methods Rat hyperlipidemic model was established. The effect of Fudoidan on rats levels of blood lipids, lipid metabolic enzyme, and the lipid contents in the stool, as well as bile metabolismand LDL-R mRNA expression in the liver were observed. Results Fudoidan can notably restrain the concentration of serum total cholesterol (TC), triglyceride (TG), low-density lipoprotein cholesterol (LDL-C) of hyperlipidemic rats and increase the concentration of high-density lipoprotein cholesterol (HDL-C) and activities of 1ipoprotein lipase (LPL), hepatic lipoprotein (HL) and lecithin cholesterol acyltransferase (LCAT).Fucoidan can remarkably increase the contents of cholesterol, TG, bile acid , but has no effect on the secretion of bile, the concentration of cholesterol and bile acid in bile. The results of RT-PCR experiment showed that Fudoidan can notably improve the expression level of low-density lipoprotein receptor (LDL-R) mRNA in the liver of hyperlipidemic rats. Conclusion Fucoidan can regulate the disorder of lipid metabolism probably by restraining the absorption of lipids (TC, cholesterol, bile acid) , activating the activities of lipid metabolic enzyme (LPL, HL and LCAT), and promoting the expression of LDL-R mRNA in the liver.
2.Diagnostic value and clinical significance of MSCT in postoperative intestinal obstruction of rectal cancer patients

Objective To evaluate the value of multi-slice spiral computed tomography (MSCT) in diagnosing postoperative intestinal obstruction of rectal cancer.Methods The CT imaging results and clinical manifestation of 50 patients with postoperative intestinal obstruction of rectal cancer confirmed by pathology were reviewed retrospectively.These cases including Miles (24 cases),Dixon (18 cases),Hartmann(8 cases) procedure were collected from Tianjin People's Hospital from 2011 to 2013.Conservative management was done in 10 cases and the remaining 40 patient underwent surgical intervention.Statistical difference between the three kinds of surgical causes of intestinal obstruction was analysed by x2 test.Results Most common cause after Miles and Hartmann postoperative intestinal obstruction was of adhesion,while anastomotic stenosis was the most common reason of intestinal obstruction after Dixon procedure.The difference of incidence of intestinal obstruction caused by hernia and anastomotic stenosis between the three kinds of operation method was statistically significant (x2 =5.382、20.486,P < 0.05).Conclusions MSCT could identify the etiology and site of postoperative intestinal obstruction of rectal cancer,providing favorable imaging evidence for clinical treatment.
3.Value of 64-slice spiral CT functional imaging techniques for diagnosis of acute kidney injury in rabbits

Objective To quantitatively assess renal hemodynamic changes in hypertensive acute kidney injury in rabbits induced by L-NAME using 64-slice spiral CT functional imaging techniques,and to explore the application of these techniques in evaluation of early kidney functional changes.Methods Fourteen female New Zealand white rabbits were randomly divided into normal control group (n=6)and L-NAME group (n=8).The control group was injected NaCl solution and the L-NAME group was injected the same amount of L-NAME solution to make hypertensive acute kidney injury model.64-slice spiral CT and SPECT were scanned af-ter injection.Blood samples were collected before and after injecting NaCl and L-NAME solution to detect serum creatinine (Cr).Cr level and CT perfusion parameters of the two groups were analyzed and compared with the pathology results.GFRCT detected by con-trast-enhanced CT and GFRSPECT detected by SPECT were analyzed by the rank correlation test.Results Renal blood volume,blood flow,permeability surface,time to peak,and peak value had statistically significant differences between the control and L-NAME group (P <0.05).GFRCT and GFRSPECT had obvious correlation.GFRCT of L-NAME group was obviously lower than that of the con-trol group.The kidneys of L-NAME group showed obviously injured under both light microscope and microscope.Conclusion 64-slice spiral CT functional imaging techniques can dynamically observe and quantitatively assess early hypertensive kidney dysfunc-tion,especially unilateral renal blood flow abnormalities.It is an effective examination in quantitatively assessing kidney function.
5.Efficacy of postmastectomy radiotherapy for HER2-positive T 1-2N 1M 0 breast cancer

Objective:To investigate the efficacy of postmastectomy radiotherapy (PMRT) for human epidermal growth factor receptor 2 (HER2)-positive T 1-2N 1M 0 breast cancer in the context of HER2-targeted therapy. Methods:This study collected the clinical data of 105 female patients with HER2-positive T 1-2N 1M 0 breast cancer who underwent modified radical mastectomy in the First Affiliated Hospital of Bengbu Medical College from January 2013 to December 2019. Then, the clinical outcomes of these patients were observed, and the prognostic factors and the efficacy of PMRT were analyzed. Results:The median follow-up time was 50 months (ranging from 14 to 107 months), and the 5-year overall survival (OS), local-regional recurrence-free survival(LRFS), and disease-free survival (DFS) were 81.6%, 91.9%, and 76.2%, respectively. The multivariate analysis indicated that independent prognostic factors for OS and DFS include the age, pathologic grade, and tumor size; the independent risk factors for LRFS include positive lymph node ratio (LNR) and hormone receptor (HR) status; and the independent prognostic factor for DFS was PMRT (HR: 2.85, 95% CI: 1.10-8.80, P < 0.05). The subgroup analysis suggested that PMRT significantly improved the OS of various high-risk subgroups ( χ2=4.01-9.18, P < 0.05). However, the further stratified analysis indicated that PMRT only increased the OS of the patients who did not receive HER2-targeted therapy in various high-risk subgroups ( χ2=4.50-6.70, P < 0.05), while there was no statistical difference before and after PMRT for the individuals who received targeted treatment ( P > 0.05). Conclusions:PMRT is an independent prognostic factor for the DFS of patients with HER2-positive T 1-2N 1M 0 breast cancer who underwent modified radical mastectomy. PMRT can improve the OS of high-risk patients with ages < 45 years old, pathologic grade Ⅲ, tumor diameter ≥ 3 cm, LNR > 10%, and HR (-) who received no HER2-targeted therapy. However, the efficacy may be compromised to some extent in the context of the application of HER2-targeted therapy.
6.Effects of the deep inspiration breath-hold technique on cardiac dosimetry in internal mammary node irradiation with intensity-modulated radiation therapy for postoperative left breast cancer

Objective:To explore the effects of the deep inspiration breath-hold (DIBH) technique on cardiac dosimetry in internal mammary node irradiation with intensity-modulated radiation therapy (IMN-IMRT) for postoperative left breast cancer.Methods:Totally 23 left breast cancer patients in the First Affiliated Hospital of Bengbu Medical College from Octorber 2021 to July 2022 receiving postoperative IMN-IMRT were enrolled in this study. The changes in dosimetric parameters for their heart and left anterior descending coronary artery (LAD) in the DIBH mode were observed, and the potential factors affecting DIBH effects were analyzed.Results:Compared with the free breath (FB) mode, the DIBH mode manifested a heart volume decrease by 18% ( t = 10.47, P < 0.001), a left lung volume increase by 42% ( t = -14.55, P < 0.001), and significantly reduced dosimetric parameters ( Dmean, Dmax, V5- V30) for the heart and LAD, exhibiting statistically significant differences ( t=-13.38 to -3.30, P<0.05). As indicated by the Pearson correlation analysis, the relative ratio of cardiac dose reduction was positively correlated with that of left lung expansion ( r = 0.82, P < 0.001) and negatively correlated with the patient′age ( r = -0.56, P = 0.005). Conclusions:DIBH can effectively reduce the heart and LAD radiation doses in IMN-IMRT for postoperative left breast cancer and that the patient's age, and the DIBH effects might be affected by the vital capacity.
7.Effect of silencing MFGE8 on proliferation,apoptosis,invasion, and migration of osteosarcoma cells U2OS

Objective :
To investigate the expression of milk fat globule epidermal growth factor 8 protein(MFGE8) in osteosarcoma cell lines and its effects on the proliferation,apoptosis,invasion and migration ability of osteosarco- ma cells(U2OS) .
Methods :
The expression of MFGE8 protein in normal osteoblasts and osteosarcoma cell lines was examined by Western blot method.The relationship between MFGE8 mRNA expression and prognosis of osteo- sarcoma patients was analyzed by GEO database.The effects of down-regulation of MFGE8 on proliferation,apopto- sis,migration and invasion of U2OS cells were examined.
Results :
Western blot method showed that the expres- sion of MFGE8 protein was higher in osteosarcoma cells than that in normal osteoblasts and its was negatively corre- lated with the prognosis of osteosarcoma patients ( all P <0. 05) ; compared with sh-Ctrl ( U2OS cells transfected with the null group) group,the U2OS cells in sh-MFGE8 (U2OS cells transfected with sh-MFGE8) group had de- creased healing rate and invasion number while increased proliferation inhibition rate and apoptosis rate ,and all differences were statistically significant (P<0. 05) .
Conclusion
MFGE8 is highly expressed in U2OS ofosteosar- coma cells,and down-regulation of MFGE8 can inhibit the proliferation,invasion and migration of U2OS and in- duce their apoptosis.
