1.Children's OSAHS caused by the huge fibrolipoma in pharynx nasalis: a case report.

We reported a case of children's OSAHS caused by the huge fibrolipoma in pharynx nasalis. The patient was a 10-years-old child who went to the hospital with the chief complaint of "Snoring and mouth breathing during sleep for 10 years". Imaging tests found one huge tumor in pharynx nasalis before the operation. The tumor was resected totally. The postoperative pathological diagnosis was fibrolipoma. No recurrence was noted during the follow-up visit one year postoperatively. The clinical features, diagnosis, treatment, pathology and prognosis were reviewed herein.

2.Clinical Study of Chair-side Immediate Ceramic Restoration Technique for Large-defected Teeth

Objective To explore the clinical application of digital chair?side immediate all?ceramic restoration system,design and fabricate inlays and onlays to restore the large?defected molars,and to analyze and evaluate the efficacy of this clinical restoration. Methods Totally 84 inlays and 65 onlays were fabricated with IPS e.max CAD series ceramic blocks by using Cerec AC system to conduct immediate all?ceramic restorations of 149 teeth for 108 patients. The inlays and onlays were applied and cemented to the prepared teeth with dual?cured resin luting agent. The restored molars were re?examined by two dentists after six months. The analysis standard was based on the United States Public Health Service(USPHS criteria) and the results were statistically analyzed(α=0.05). Results There was no statistically significant difference in the case numbers of grade A and grade B for each index comparing the immediate condition to the condition after 6 months. Over 90 percent of ceramic inlays and onlays were quali?fied to be grade A. The restorations were well in the aspects of anatomic morphology,marginal integrity,color match,marginal discoloration,reten?tion,gingival health and secondary caries. Conclusion Computer aided design/computer aided manufacture(CAD/CAM)system combined with machinable glass ceramics can achieve a favorable result in restoration of large?defected teeth. Digital chair?side immediate all?ceramic restoration technique is an ideal solution for large?defected tooth restoration.
3.STUDIES ON LONG-EFFECTIVE VARNISH “FANGYOULING”IN PREVENTION OF SCHISTOSOMA CERCARIA INFECTION

ObjectiveTo observe the effe ct of varnish“Fangyouling” preventing skin invasion from Schist osoma cercariae. MethodsThe“Fangyouling” was made from niclo s amide and permeable improver, and the concentration of the drug was 1%-2%. Exper imenta l mice and rabbits were spread with “Fangyouling” on the abdomen skin without h air at day 1,2,3,4,5,6,7 respectively before infection, compaired with cont rol group. ResultsThe worm reduction rates of mice which were spread with drug 1-4 days and 5-7 days before infection were 100 % and 99 7%-88 1%. The worm reduction rates of rabbits which were spread with dr ug 3-7 days before infection were 86 4%-80 1%. ConclusionThe“Fangyouling” has long efficiency on preventing Schistosoma cercariae from invading skin.
4.Effects of curcumin on the AKT/mTOR signaling pathway in human melanoma cell lines A375 and C8161

Objective To explore molecular mechanisms underlying the in vitro counteracting effect of curcumin on malignant melanoma.Methods Cultured A375 and C8161 human melanoma cells were cultivated in vitro,and randomly divided into several test groups and a control group to be treated with different concentrations of curcumin and dimethyl sulfoxide respectively for different durations.Then,methyl thiazolyl tetrazolium (MTT) assay,Transwell assay,flow cytometry and Western blot were performed to evaluate the effect of curcumin on the proliferation,invasion and cell cycle of,as well as expressions of AKT/mTOR signaling pathway-related proteins in A375 and C8161 cells respectively.Statistical analysis was carried out by using t test.Results MTT assay showed that the treatment with curcumin of 5-35 mg/L for 24-96 hours significantly inhibited the proliferation of both A375 and C8161 cells compared with that with dimethyl sulfoxide (all P < 0.001),and the inhibitory effect was in a dose-dependent manner within the range of 5-15 mg/L for A375 cells and within the range of 5-10 mg/L for C8161 cells,and in a time-dependent manner from 0 to 48 hours for both cells.After treatment for 24 hours,the 50% inhibitory concentration (IC50) of curcumin against A375 cells and C 8161 cells was 10 mg/L and 5 mg/L respectively.Transwell assay demonstrated that the invasion of A375 and C8161 cells was significantly suppressed by 72-hour treatment with curcumin at 10 mg/L and 5 mg/L respectively (both P < 0.001).Flow cytometry showed that the cell cycle of A375 and C8161 cells was arrested at G2/M phase after 24-hour treatment with curcumin at 10 mg/L and 5 mg/L respectively,with significant differences in the proportion of A375 cells and C8161 cells in G2/M phase between the test group and control group (A375 cells:35.00% ± 3.54% vs.120.80% ± 7.46%,P< 0.001;C8161 cells:19.33% ± 4.04% vs.85.00% ± 9.53%,P < 0.001).Western blot revealed that the expressions of AKT/mTOR signaling pathway-related proteins were decreased in A375 and C8161 cells after 24-hour treatment with 10 mg/L and 5 mg/L curcumin respectively.Conclusion Curcumin can inhibit the proliferation and invasion of A375 and C8161 cells,likely by blocking cell cycle and inhibiting activation of the AKT/mTOR signaling pathway.
5.EXPERIMENTAL STUDIES ON EFFICACY OF LEVAMISOLE IN PREVENTION FR OMSCHISTOSOMA JAPONICUM CERCARIA INFECTION

Objective To study the effect o f levamisole against the invasion of Schistosoma japonicum cercariae. Methods Mice infected wit h Schistosoma cercariae were administered orally with levamisole hydrochloride or alkali levamisole two days before the infectio n at a dose of 26.25 mg/kg for 7 days. The liniments of levamisole hydrochlorid e and alkali levamisole were embrocated on the mouse skin two days, one day and 0 day before the infection respectively, and the concentrations of the drug were 1%, 2%, 3%, 5% and 7% respectively. The experimental animals were dissected 4 we eks after the treatment and adult worms were collected. Results The worm reduction rates of mice administered orally with leva misole hydrochloride or alkali levamisole were both 0. The worm reduction rates were both 100% when the mice were embrocated with 5% levamisole hydrochloride on the infection day or with 7% levamisole hydrochloride one day before the infect ion. The worm reduction rates were all 100% when the mice were embrocated with 2 %, 3% or 5% alkali levamisole one day before the infection. Conc lusions Levamisole liniments can prevent from S . japonicum cercaria infection, and alkali levamisole is better th an levamisole hydrochloride. When levamisole is given orally, no effect was show n.
6.Cox regression analysis of recurrence and metastasis factors of colon cancer radical resection in elderly patients

Objective To analyze the clinicopathologic data and operative parameters of 209 elderly co-lon cancer patients treated by radical resection between January 2002 and December 2011 ,and to investigate the factors related to recurrence and metastasis after colon cancer radical resection in elderly patients .Methods We used univariate and multivariate analysis of Cox regression ,including 14 variables:age,gender,disease duration, hospitalization duration,surgeon experience,operation duration,laparoscopicsurgery,tumor location,tumor size, gross morphology ,differentiate degree ,depth of bowel wall invasion ,lymph node involvement and obstruction .The survival curve was obtained by Kaplan -Meier method.Results Univariate analysis showed that tumor size (RR=2.658,P<0.0001),gross morphology(Infiltrating type,RR=3.407,P=0.0054),degree of differentiation (RR=0.32,P<0.0001) were associated with tumor relapse and metastasis .Multivariate analysis showed that gender(RR=0.585,P=0.0359),tumor size(RR=2.364,P<0.0001),degree of differentiation (Infiltrating type,RR=0.246,P=0.0437),gross morphology(RR=0.31,P<0.0001)were the significant factors.Conclu-sion Gender,tumor size,degree of differentiation,gross morphology were the independent factors of recurrence and metastasis of colon cancer after radical resection in elderly patients .Targeted follow -up for high -risk groups will improve patients′life quality and prolong their survival time .
7.Characteristics and immune mechanisms of CD71+ CD235a+ nucleated erythroid cells in elderly

Objective To explore the immune property and possible mechanism of the CD71+ CD235a+ nucleated erythroid cells from peripheral blood in elderly,as compared with those of healthy young.Methods Peripheral blood obtained by venipuncture from healthy young(n=59,mean age=28 years) and elderly(n=78,mean age=68 years)donors were measured by flow cytometry to evaluate the frequency of the CD71+ CD235a+ cells in peripheral blood mononuclear cells (PBMC).In vitro assays,CD71+ CD235a+ cells were sorted by flow cytometry and T cells were sorted using CD4+T cell isolation kit,then the T cell proliferation assays were conducted by following groups:T cell + CFSE group;T cell +CFSE co-cultured with CD71+ CD235a+ cells from the young donors;T cell + CFSE co-cultured with CD71+ CD235a+ cells from the older donors.Real-time PCR were used to identify the expression of the immune cytokine secreted by CD71+ CD235a+ cells.Results (1)the results showed a significant increase in the percentage of CD71+ CD235a+ cells in the elderly compared with the young [(9.93± 2.95)% vs (1.96 ± 1.16)%,t =3.37,P < 0.001];(2)the CD71+ CD235a+ cells from the older donors could suppress the proliferation of the T cells (t =2.91,P< 0.05)while from the young group we could not observe this phenomenon(t =0.387,P>0.05).(3)The results of Real-time PCR revealed that,compared with CD71+ CD235a+ cells from the young,CD71+ CD235a+ cells from the elderly expressed higher levels of Arg-2 (t =9.04,P<0.01),IL-1β,IL-6 and TGF-β(t =4.51,5.46,6.92,all P<0.05).Conclusions There are higher frequencies of CD71+ CD235a+ cells in aged samples than the young.And the CD71+CD235a cells from the elderly could secrete more Arg-2,IL-1β,IL-6 and TGF-β to suppress the T cell proliferation.

Objective To construct five shRNA-expression plasmids and to investigate the expression of Smad2 in TGF-?/ Smads signal transduction treated with shRNA-expression plasmid.Methods Five shRNA-Smad2 DNA sequences from mRNA sequence of mouse Smad2 gene were designed and synthesized.DNA oligonucleotides encoding an appropriate shRNA were inserted to shRNA expression vector respectively.Five shRNA-Smad2 expression plasmids were obtained and then transfected into NIH/3T3 cells.The suppressed expression of Smad2 was assessed by RT-PCR and Western-blotting.Results The shRNA-expression plasmid numbered 2.4 could markedly reduce the expression of Smad2.The suppression effect of the RNAi-pool composed of four different plasmids was more obvious than that of any single.Conclusion The shRNA-expression plasmids were successfully constructed,which could specifically and effectively suppress the expression of Smad2.The method of using a mixture of RNAi plasmids to improve the RNAi efficiency was established.

Objective To study characteristics and immune mechanisms of CD11b+ GR-1-myeloid-derived suppressor cells (CD11b+ GR 1+ MDSCs) in elderly mice,as compared with those of healthy young mice.Methods Totally 20 healthy C57BL/6 young mice (aged 1-2 months) and 20 elderly mice (aged over 18 months) were randomly chosen and splenetic CD11b+ GR-1+ MDSCs were sorted with the MDSCs Isolation Kit.In vitro assays,the effects of young and elderly CD1 1b+ GR 1+ MDSCs on the proliferation of T cells were determined by Brdu Elisa.Transwell co-culture and real-timePCR were used to identify the mechanisms of different immune suppressive functions of CD11b+GR 1+ MDSCs sorted from young mice and elderly mice.Results Compared with young MDSCs,elderly MDSCs could evidently suppress the proliferation of T cells (t=8.67,P<0.001),and this function could be reversed by trans-well co-culture (t=6.93,P<0.001).The results of realtime PCR revealed that,compared with young MDSCs,elderly MDSCs expressed higher levels of arginase-1 (ARG-1),inducible nitric oxide synthase (iNOS),reactive oxygen species (ROS),interleukin 10 (IL-10),IL13 and transforming growth factor (TGF)-β (t=9.04,4.86,7.04,6.92,4.51,5.46,respectively,P<0.05 or P<0.01).Conclusions CD11b+GR-1+MDSCs sorted from healthy elderly mice can evidently suppress the proliferation of T cells through cell-cell contact and secretion of suppressive medium.
10.Identification of the subpopulations of myeloid-derived suppressor cells and the function study in elderly tumor-bearing mice

Objective To investigate the level of subpopulations of myeloid-derived suppressor cells (MDSCs) in elderly tumor-bearing mice versus elderly tumor-free mice,and to study the difference in immune suppressive functions between different subpopulations and their mechanisms.Methods A total of 20 healthy C57BL/6 elderly mice(aged 18-20 months) were randomly chosen to establish Lewis lung cancer models.The amount of monocytic-MDSCs (MO-MDSCs) and polymorphonuclear granulocytic-MDSCs(PMN-MDSCs) in tumor-free and tumor-bearing elderly mice was evaluated by using flow cytometry.MO-MDSCs and PMN-MDSCs were separated with Magnetic-Activated Cell Sorting (MACS) MicroBeads and their morphological characteristics were observed after May-Grunwald-Giemsa staining.The effects of MO-MDSCs and PMN-MDSCs on the proliferation of T cells were determined by Brdu-enzyme-linked immunosorbent assay(ELISA).And the immune suppressive mediators secreted by the subpopulations were detected by Real-time polymerase chain reaction(PCR).Results Compared to the tumor-free group,the proportion of MO-MDSCs in the spleen of tumorbearing group were increased [(12.44± 1.20) % vs.(38.42±3.66) %,t=5.67,P<0.001],while PMN-MDSCs were not [(10.34±0.68) % vs.(12.18±1.27) %,t=2.21,P=0.09].The result of Brdu-ELISA showed that MO-MDSCs could suppress the proliferation of T cells [(0.30 ± 0.18) vs.(3.38±0.96),t=8.33,P<0.001],while PMN MDSCs could not [(2.69±0.45)vs.(3.38±0.96),t =1.72,P=0.11].The result of PCR showed that as compared with PMN-MDSCs,Mo-MDSCs had the increased expression levels of arginase-1 (ARG-1),inducible nitric oxide synthase (iNOS),interleukin-10 (IL-10),interferon-γ(IFN-γ) (t =4.31,8.89,1.70,3.13,respectively,P < 0.01 or 0.05),while the expression levels of interleukin-13 (IL-13),transforming growth factor-β(TGF-β) had no differences (t=4.94 and 2.75,P =0.39 and0.47).Conclusions MO-MDSCs are significantly increased in elderly Lewis lung cancer mice models.MO-MDSCs could mediate lung tumor evasion by suppressing the proliferation of T cells through highly expressing ARG-1,iNOS,IL-10 and IFN-γ.
