3.Effect of serum obtained from rat treated orally with Traditional Chinese Medicine Nao Yi-An on MAPK signal transduction in injured cultured neurons

ObjectiveTo explore the effects of the serum of traditional Chinese medicine Nao Yi An on glutamate induced cell death in cultured hippocampal neurons of rat and the underlying mechanisms. MethodsHippocampal neurons were cultured. The excitatory amino acid induced toxicity on cultured neurons was investigated. The viability of injured neurons was determined with the measurement of Lactate dehydrogenase (LDH) activity. Mitogen activated protein kinase (MAPK) were determined by immunoprecipitation /kinase assays /western blot detection.ResultsThe serum of Nao Yi-An raised cell viability. The serum of Nao Yi-An upregulated the expression of extracellular regulated protein kinases(ERK) and downregulated the expression of c-Jun N terminal kinase/stress activited protein kinase(JNK) in cultured neurons. The serum of Nao Yi-An induced upregulation of ERK and its anti death action were prevented with the specific ERKs inhibitor PD98059. Conclusions Activation of ERK signaling together with inhibition of JNK signaling by Chinese medicine Nao Yi-An appears to be an important mechanism for its survival effects on cultured hippocampal neurons.
4.Effects of amino acid on growth and secondary metabolites contents of adventitious roots of Tripterygium wilfordii.

The adventitious root of Tripterygium wilfordii was used as experiment material to study effects of various concentration of aspartic acid, isoleucine, cysteine and arginine in MS medium on the growth and triptolide, wilforgine, wilforine contents of the adventitious roots. The results showed that compared with the control, supplemented with 0.25 mmol x L(-1) aspartic acid at 3rd week, the growth of the adventitious roots only accounted for 80%, but the content of triptolide of the adventitious roots and the medium was 1.36, 1.30 times, the content of wilforgine was 1.16, 1.37 times, the content of wilforine was 1.22, 1.63 times, respectively. At 3rd week 0.05 mmol x L(-1) isoleucine, the growth of adventitious roots was 97.3%, wilforgine of adventitious roots and medium 1.02, 1.27 times, wilforine 1.36 times and 1.15 times. At 1st week 0.25 mmol x L(-1) cysteine, the growth of the adventitious roots comprised 77.5% of the control, while content of triptolide of adventitious roots reached 1.87 times. At 2nd week 1.00 mmol x L(-1) cysteine, the growth of adventitious roots was 44.6% of the control, the content of wilforine in medium was 2.97 times. At 3rd week 0.50 mmol x L(-1) arginine, the growth of adventitious roots was 124.2%, the content of wilforgine and wilforine was 1.3, 1.4 times, respectively.

6.Neutrophil extracellular traps induce production of MPO-ANCA in chronic bronchitis rats

Objective:To investigate the mechanism of chronic bronchitis with recurrent long duration infections in the production of MPO anti-neutrophil cytoplasmic antibodies(ANCA). Methods:Rats were gave intratracheal injection of LPS and smoked to established chronic bronchitis ( CB ) model. Rat neutrophils were incubated with phorbol-12-myristate-13-acetate ( PMA ) to induce Neutrophil extracellular traps(NETs). NETs were then used to immune chronic bronchitis rats. Serum concentrations of myeloperoxidase anti-neutrophil cytoplasmic antibodies(MPO-ANCA),Citrullinated Histone H3(CitH3),Interferon-α(IFN-α)were evaluated. Results:(1)The serum levels of MPO-ANCA in chronic bronchitis rats immuned with NETs(CB/NETs)were significantly higher than those in chronic bronchitis rats(CB)and healthy control rats,(62. 4975±19. 63764)ng/ml vs (17. 3806±4. 88649)ng/ml,(20. 8884± 4. 39607)ng/ml(both P<0. 01). (2)The serum levels of CitH3 in CB/NETs rat were significantly higher than those in CB rat and healthy control rats,(32. 5974±20. 2915)ng/ml vs (8. 8568±2. 36692)ng/ml,(5. 6225±0. 83738)ng/ml(P<0. 05,P<0. 01). (3)The serum levels of IFN-α were found increased sharply in two CB/NETs rats(3735.7,3428.4 pg/ml). (4)Renal function among three groups showed no significant difference yet. ( 5 ) Serum CitH3 level showed a positive correlation with serum MPO-ANCA level(r=0. 61,P<0. 05). Conclusion:Chronic bronchitis with recurrent infections could induce the production of ANCA probably by the stimulation of NETs,but whether it might develop to AAV remains need a duration.
7.Extraction of three kinds of volatile oil and preparation of ?-CYD inclusion compound of Yuxianling Granules

Objective:To specify optimize condition of process for mixture extraction of three kinds of volatile oil and preparation of ? CYD inclusion compound of Yuxianling Granules.Methods: Taking weight of volatile oil to evaluate the extraction process and percentage recovery of inclusion compound, transfer rate of oil, inclusion rate of oil to evaluate the inclusion process. Orthogonal design was applied. Results: The optimum extraction condition was to add five times amount of water into the mixture of crude drugs, to soak for 90 min, to heat up to boiling and reflux 4h. The trituration method is suitable for the productive preparation of inclusion compound, the optimum inclusion condition was ? CYD: Volatile oil in proportion as 8∶1 was mixed, adding two times amount of water into the mixture and trituration 30 min.Conclusions: Above condition of process was the most rational in extraction and inclusion of three kinds of volatile oil.
8.Research hotspots of diabetes management in China

Objective:
To analyze the research status,research hotspots and its changing trends of diabetes management in China.
Methods:
We searched the databases of CNKI,Wanfang and VIP to collect literature about diabetes management until December 7th of 2019;counted the number of published articles and journals;plotted the co-occurrence maps of authors and institutions,and time zone map of keywords by CiteSpace software,so as to analyze the research status,research hotspots and its changing trends of diabetes management.
Results :
A total of 8 435 articles were included,and the number of the articles was increasing year by year. The largest number,8 141 (96.51%) of published articles lay in the period from 2009 to 2019. The articles published in 826 journals,13 of which were in the core area. Diabetes New World published the largest number,419 (4.97%) of articles. Lou Qingqing was the author with the most articles published (26,accounting for 0.31%). Shanghai Jiao Tong University,West China Hospital of Sichuan University and the First Affiliated Hospital of Jinan University ranked the top three in articles published,with 70 articles accounting for 0.83%. The keywords that appeared more frequently were self-management,health education,blood glucose management,gestational diabetes,health management,community management and nursing management. The more centralized keywords were self-management,health education,gestational diabetes and community management. Self-management and health education had always been research hotspots. Family doctors and mobile health were new directions in recent years.
Conclusions
The volume of domestic articles on diabetes management has been increasing. Self-management and health education has always been hotspots,meanwhile,family doctor and mobile health have become new hotspots of diabetes management research.

10.Growth hormone enhances regeneration of nNOS-containing nerve fibers in the aged rat penis.

OBJECTIVETo investigate the effects of growth hormone (GH) on the erectile function and the number of neuronal nitric oxide synthase (nNOS) -containing nerve fibers in the penis of aged rats.
METHODSTwenty-four aged male SD rats (18 months) were randomized into 2 groups: GH intervention group and control group. After four and eight weeks, a half of each group were selected and tested for erectile function after apomorphine (APO) injection and then sacrificed for the detection of nNOS-containing nerve fibers in the penis by streptavidin-peroxidase conjugated method (SP method).
RESULTSAfter four weeks, the erectile function and the number of nNOS-containing nerve fibers showed no significant difference between the GH intervention group and the control group (P > 0.05). After eight weeks, the erection frequency was significantly different (P < 0.05) between the two groups, while the erection rate was not. The number of nNOS-containing nerve fibers in the GH intervention group was significantly larger than that in the control group (P < 0.01).
CONCLUSIONGH enhances the regeneration of nNOS-containing nerve fibers in the penis and improves the erectile function of the aged rat.
